The changes in biological behavior and phenotype of human epidermal keratinocytes in culture

Abstract:Objective:To provide experimental basis for the study of dedifferentiation through obtaining a high proportion of differentiating keratinocytes in vitro.Methods:Using routine culture method of epidermal cells,the human epidermal keratinocytes (HEKs) were cultivated and subcultured.The cells of each passage were deter- mined for the markers of both epidermal stem cell and epidermal cell by immunocytochemical staining and Western blot analysis.These markers includedβ1 integrin,keratin 19(K19),and keratin 10(K10).Results:Cells of the first passage were clonogenic and were positive forβ1 integrin and K19,whereas negative for K10.The morpholo- gy and phenotype of HEKs began to change after several passages of subculture.The number of clones decreased gradually and the distribution of cells appeared to be dispersed.The number of cells positive forβ1 integrin and K19 showed a tendency of decrease,whereas those positive for K10 increased.The first turning point for the change was the 5th to 6th passage,during which the proliferation ability of HEKs declined and the proportion of cells positive for K10 significantly increased.The second turning point for cells occurred in 10th to 11th passage. HEKs in this period showed no proliferation ability and the number of cells significantly decreased.The cells be- came large and with a low karyoplasmic ratio.The cells positive forβ1 integrin and K19 disappeared,and all the cells were positive for K10.Conclusion:HEKs of certain passages can be used in the study of dedifferertiation, and it provided an experimental basis for a novel utilization of HEK.
