Study on TGF-β1 Inducing Transdifferentiation of SUVEC to Smooth Muscle-like Cells

Abstract:To demonstrate the possibility of swine umbilical vein vascular endothelial cells(SUVEC) to transdifferentiate into smooth muscle like cells,porcine vascular endothelial cells were incubated with transforming growth factor-β1 on different concentration.The changes in morphology or expression of specific marker of cells were analyzed by Immunohistochemical staining and image analysis method.It showed that expression of α-SMA increased distinctly when treated with 5～10 ng·mL-1 TGF-β1,whereas expression of F-Ⅷrelated antigen in the cells notably declined.Interestingly,20ng/mL TGF-β1 did not promote up-regulating of α-SMA.SUVECs lost their cobblestone morphology and converted into ragged cells in TGF-β1-treated culture.We demonstrated here that swine vascular endothelial cells can be transdifferentiated into smooth muscle like cells treated with TGF-β1.What's more,the ability of transdifferentiation correlated to the concentration of TGF-β1.
