Abstract:Male reproductive development is a complex and highly ordered phenomenon which demands comprehensive understandings of underlying molecular mechanisms to expand its scope for crop improvement. Genetic manipulation of male fertility/sterility is critical for crop hybrid breeding. Although male sterility is not a good trait for the plant itself, its wider application in hybrid rice breeding has made it valuable. The currently widely used male sterile line breeding systems mainly include the following: three-line hybrid rice based on cytoplasmic male sterility and two-line hybrid rice based on environmentally sensitive gene male sterility. The study of male sterility is an excellent thoroughfare to critically understand the regulatory mechanisms essential for the complicated male reproductive developmental process. The unique trait of male sterility also provides valuable resources and convenience for the genetic improvement of rice hybrids. Therefore, deeper and broader understandings about the genetic causes of male sterility are necessary for both basic studies and rice genetic improvement.

Abstract:Rice is one of the most important model crop plants whose heterosis has been well-exploited in commercial hybrid seed production via a variety of types of male-sterile lines. Hybrid rice cultivation area is steadily expanding around the world, especially in Southern Asia. Characterization of genes and proteins related to male sterility aims to understand how and why the male sterility occurs, and which proteins are the key players for microspores abortion. Recently, a series of genes and proteins related to cytoplasmic male sterility (CMS), photoperiod-sensitive male sterility, self-incompatibility, and other types of microspores deterioration have been characterized through genetics or proteomics. Especially the latter, offers us a powerful and high throughput approach to discern the novel proteins involving in male-sterile pathways which may help us to breed artificial male-sterile system. This represents an alternative tool to meet the critical challenge of further development of hybrid rice. In this paper, we reviewed the recent developments in our understanding of male sterility in rice hybrid production across gene, protein, and integrated network levels, and also, present a perspective on the engineering of male-sterile lines for hybrid rice production.

Abstract:Selfish genetic elements are pervasive in eukaryote genomes, but their role remains controversial. We show that qHMS7 , a major quantitative genetic locus for hybrid male sterility between wild rice ( Oryza meridionalis ) and Asian cultivated rice ( O. sativa ), contains two tightly linked genes [ Open Reading Frame 2 ( ORF2 ) and ORF3 ]. ORF2 encodes a toxic genetic element that aborts pollen in a sporophytic manner, whereas ORF3 encodes an antidote that protects pollen in a gametophytic manner. Pollens lacking ORF3 are selectively eliminated, leading to segregation distortion in the progeny. Analysis of the genetic sequence suggests that ORF3 arose first, followed by gradual functionalization of ORF2 . Furthermore, this toxin-antidote system may have promoted the differentiation and/or maintained the genome stability of wild and cultivated rice.

Abstract:Rice ( Oryza sativa L.) occupies a very salient and indispensable status among cereal crops, as its vast production is used to feed nearly half of the world’s population. Male sterile plants are the fundamental breeding materials needed for specific propagation in order to meet the elevated current food demands. The development of the rice varieties with desired traits has become the ultimate need of the time. Genic male sterility is a predominant system that is vastly deployed and exploited for crop improvement. Hence, the identification of new genetic elements and the cognizance of the underlying regulatory networks affecting male sterility in rice are crucial to harness heterosis and ensure global food security. Over the years, a variety of genomics studies have uncovered numerous mechanisms regulating male sterility in rice, which provided a deeper and wider understanding on the complex molecular basis of anther and pollen development. The recent advances in genomics and the emergence of multiple biotechnological methods have revolutionized the field of rice breeding. In this review, we have briefly documented the recent evolution, exploration, and exploitation of genic male sterility to the improvement of rice crop production. Furthermore, this review describes future perspectives with focus on state-of-the-art developments in the engineering of male sterility to overcome issues associated with male sterility-mediated rice breeding to address the current challenges. Finally, we provide our perspectives on diversified studies regarding the identification and characterization of genic male sterility genes, the development of new biotechnology-based male sterility systems, and their integrated applications for hybrid rice breeding.

Abstract:Abstract Development of stable male sterility lines is essential for efficient hybrid seed production. We previously showed that knockout of bHLH142 in rice ( Oryza sativa ) causes pollen sterility by interrupting tapetal programmed cell death (PCD). In this study, we demonstrated that overexpression of bHLH142 (OE142) under the control of ubiquitin promoter also leads to male sterility in rice by triggering the premature onset of PCD. Protein of bHLH142 was found to accumulate specifically in the OE142 anthers. Overexpression of bHLH142 induced early expression of several key regulatory transcription factors in pollen development. In particular, the upregulation of EAT1 at the early stage of pollen development promoted premature PCD in the OE142 anthers, while its downregulation at the late stage impaired pollen development by suppressing genes involved in pollen wall biosynthesis, ROS scavenging and PCD. Collectively, these events led to male sterility in OE142. Analyses of related mutants further revealed the hierarchy of these pollen development regulatory genes. Thus, the findings of this study create a new method to generate genic male sterility in rice. Exploitation of this novel functionality of bHLH142 would confer a big advantage to hybrid seed production. Highlight Overexpression of bHLH142 leads to male sterility in transgenic rice due to early onset of tapetal PCD. This study creates a new method to generate male sterility in rice.

Abstract:Maize ( Zea mays ) represents one of the most successful uses of heterosis among crops. Hybrid maize seeds can be produced by crossing a male-sterile female parent with a male parent as the pollen donor to achieve high seed purity at low cost. Cytoplasmic male sterility (CMS) has been widely used for maize hybrid seed production. Recently, several hybrid seed production technologies based on genic male sterility (GMS) genes have been developed. In addition, the identification of environment-sensitive genic male sterility (EGMS) genes provides opportunities for the two-line system to be applied in maize hybrid seed production. In this review, we systematically summarize the male-sterile genes and molecular mechanisms of male sterility in maize. Future prospects for the study of maize male sterility are highlighted. Future research will enhance our understanding of the molecular regulatory networks of male sterility and promote the process of maize hybrid seed production using male sterile lines.

Abstract:Male sterility is defined as failing to produce functional pollen during stamen development in plants, and it plays a crucial role in plant reproductive research and hybrid seed production in utilization of crop heterosis. High throughput RNA sequencing (RNA-seq) has been used widely in the study of different fields of life science, as it readily detects all the mRNA and non-coding RNA in cells. Recently, RNA-seq has been reported to be applied in different species and kinds of pollen abortion types in plants, which has contributed to the understanding of the molecular mechanism and metabolic networks of male sterility at the transcription level. In this review, we summarize research progress on the mechanisms of male sterility in plants, focusing on RNA-seq analysis encompassing strategies of RNA library construction, differentially expressed genes and functional characteristics of noncoding RNAs involved in stamen abortion. Furthermore, we also discuss application of transcriptome sequencing technology to elucidate pollen abortion mechanisms and map fertility-related genes. We hope to provide references to the study of male sterility in plants.

Abstract:Male sterility is an important inheritance phenomenon in plants and widely used in hybrid seed production. The male sterile plant can't produce normal male gametophyte for reproduction, while the female counterpart is normal. So far, more than 100 of male sterile mutants or genes have been reported in rice. Ms-np is a male-sterile mutant derived from a spontaneous muta- tion. The filaments of the mutant are long and thin, and the withered anthers are white and transparent. Ms-np was confirmed to be a none-pollen type mutant of male sterility, for no pollen grains were stained with I2-KI solution and the anther locules were al- ways hollow. Genetic analysis of five F2 populations and two BC1F1 populations revealed that the mutation was controlled by a single recessive gene. To uncover the molecular basis of ms-np, the F2 population derived from the cross of ms-np/M63 was used for genetic mapping. Screening of 227 F2 mutant individuals with simple sequence repeat (SSR) markers indicated that ms-np(t) was located between the molecular markers RM541 and RM343, on chromosome 6, with the distances of 15.2 and 7.9 cM, re- spectively. The results provide a basis for further gene cloning and understanding of the molecular mechanism underlying rice male fertility.

Abstract:Cytoplasmic male sterility (CMS) is a widespread phenomenon in plants. It is mainly characterized by the absence of functional pollen resulting in abnormal fertilization and fruiting. CMS is caused by cytoplas-mic genes in plants that exhibit male sterility. Recent studies have shown male sterility caused by CMS genes are genetically bound to mitochondria. Accordingly, genes in the nucleus are present to encode a protein which restores pollen sterility, and these genes are known as fertility restorer genes (Rf genes). Furthermore, the pro-duction of hybrid rice depends on the discovery and utilization of CMS and it is also an effective way to in-crease crop production. So far, the mechanism of CMS and fertility restoration were uncovered deeply in rice. This paper mainly introduces the progress of rice CMS genes and the mechanisms of CMS and fertility resto-ration, which may be provide a reference for revealing CMS and fertility restoration mechanism of other crops, and new ideas for the cultivation of hybrid rice.

Abstract:Thermosensitive genic male-sterile (TGMS) lines, which are male-sterile at restrictive (high) temperatures but male-fertile at permissive (low) temperatures, have been widely used in breeding two-line hybrid rice (Oryza sativa L.). Here we find that mutation of thermosensitive genic male sterile 5 (tms5) in rice causes the TGMS trait through a loss of RNase Z(S1) function. We show that RNase Z(S1) processes the mRNAs of three ubiquitin fusion ribosomal protein L40 (UbL40) genes into multiple fragments in vitro and in vivo. In tms5 mutants, high temperature results in increased levels of UbL40 mRNAs. Overaccumulation of UbL40 mRNAs causes defective pollen production and male sterility. Our results uncover a novel mechanism of RNase Z(S1)-mediated UbL40 mRNA regulation and shows that loss of this regulation produces TGMS in rice, a finding with potential applications in hybrid crop breeding.

Abstract:Environment-sensitive genic male sterility is a type of male sterility that is affected by both genetic and environmental factors. Environment-sensitive genic male sterile lines are not only used in two-line hybrid breeding but are also good materials for studying plant-environment interactions. In this study we review the research progress on environment-sensitive genic male sterility in rice from the perspectives of epigenetic, transcriptional, post-transcriptional, post-translational, and metabolic mechanisms as well as signal transduction processes. While significant progress has been made in the genetics, gene cloning, and understanding of the molecular mechanisms of environment-sensitive genic male sterility in recent years, the relevant regulatory network is still poorly understood in rice. We therefore also review studies of environment-sensitive genic male sterility in Arabidopsis and other crops, hoping to promote research in this field in rice. Finally, we analyze the challenges posed by environment-sensitive genic male sterility and provide corresponding suggestions. This review will contribute towards an understanding the molecular genetics of environment-sensitive genic male sterility and its application in two-line hybrid breeding in rice and other species. Abstract:Background Rice ( Oryza sativa ) is one of the main crops in the world, and more than 3.9 billion people will consume rice by 2025. Sterility significantly affects rice production and leads to yield defects. The undeveloped anthers or abnormal pollen represent serious defects in rice male sterility. Therefore, understanding the mechanism of male sterility is an important task. Here, we investigated a rice sterile mutant according to its developmental morphology and transcriptional profiles. Results An untagged T-DNA insertional mutant showed defective pollen and abnormal anthers as compared with its semi-sterile mutant ( sstl ) progeny segregates. Transcriptomic analysis of sterile sstl - s revealed several biosynthesis pathways, such as downregulated cell wall, lipids, secondary metabolism, and starch synthesis. This downregulation is consistent with the morphological characterization of sstl - s anthers with irregular exine, absence of intine, no starch accumulation in pollen grains and no accumulated flavonoids in anthers. Moreover, defective microsporangia development led to abnormal anther locule and aborted microspores. The downregulated lipids, starch, and cell wall synthesis-related genes resulted in loss of fertility. Conclusions We illustrate the importance of microsporangia in the development of anthers and functional microspores. Abnormal development of pollen grains, pollen wall, anther locule, etc. result in severe yield reduction.
