Abstract:Through the summarization of the drug usage characteristics and case analysis of Prof. Wu Wei - ping's experiences in the treatment of cough,it was discovered that based on the clinical feature of cough,Prof. Wu believed that the pathogenesis of cough was the upward perversion of lung qi. The treatment was emphasized on promoting qi activity,especially regulating liver and lung qi. In the paper,it was intro-duced that the modified xiaochaihu tang was used extensively in the treatment of cough. For cough due to in-vasion of exogenous pathogens,the treatment was to harmonize qi circulation and eliminating exogenous patho-gens. For cough due to internal injury,the treatment was to regulate qi activity,harmonize qi and blood and prevent from the invasion of pathogens. Xiaochaihu tang is good at regulating qi activity and smoothing liver and gallbladder. The clinical efficacy was significant.

Abstract:Objective: To observe the diuretic and analgesic action of Sanwei Jili Powder(SJP).Methods:The urinary effects of SJP were studied in loading rats with water,The analgesic effects of SJP were studied with mice writhing response.Results: SJP could significantly increase the urinary output and shorten the latent period of emiction,reduce writhing frequency of mice.Conclusion: SJP exerts the significant diuretic effect in rats and analgesic effect in mice.

Abstract:Objective: To research the main pharmacological actions of Penyan Xiao extract for chronic pelvic inflammation based on clearing heat, promoting qi circulation and relieving pain effect and clarify the mechanism of action about anti-inflammatory action, analgesic action and immunologic function. Methods: The analgesic action of Penyan Xiao extract was studied by mouse hot plate method; the anti-inflammatory action was verified by experiments of mouse agar-induced granuloma; and the immunologic function was studied according to observed the effects of Penyan Xiao Extract on function of mouse mononuclear phagocytes. Results: Penyan Xiao extract could reduced the weight of mouse agar-induced granuloma, increased the pain threshold in mouse, and could also increased the function of mouse mononuclear phagocytes in the 20 minutes, and there is no statistically significant difference between water control group after calculated the K. Conclusion: Penyan Xiao extract has a good anti-inflammatory effect and analgesic action and can improve the body′s immunity.

Abstract:To study the pharmacodynamics of the Liu wei hua shi tong. The effects of Liu wei hua shi tong on diuresis, analgesia, antiinflammation and antithromboxane were observed and studied by using Funnel Test, Pains in Writhing Model, Permeation of Capillary Vessel, Foot Swelling by egg white, and Sheet Glass Test. The Liu wei hua shi tong increased urine volume much significantly and decreased the pain response induced by acetic acid on mice in Writhing experiment, and also reduced the capillary permeability of abdominal cavity induced by acetic acid and inhibited the reaction of Foot Swelling induced by egg white in mice. The drug can shortened the clotting time significantly. [Conclusion] The Liu wei hua shi tong possesses the effects of diuresis,analgesia, antiinflammation and promoting blood coagulatlon.

Abstract:OBJECTIVE:To study the primary effects of Niuhuang Qingwei wan on the gastrointestinal function in aninmal for justifying its efficacies in clinic.METHOD:Mice were twice administered with Niuhuang Qingwei wan (0.83, 1.67, 3.33 g x kg(-1), ig) and rats were twice administered with Niuhuang Qingwei wan (0.59, 1.18, 2.36 g x kg(-1), ig). The effects on the stomach function were evaluated by the gastric emptying test in mice and the gastric analysis in rats. The effect on the intestinal function were evaluated by the propulsive motility of the total gastrointestinal tract test in mice by recording the time and frequency of excreting carbo medicinalis. Its analgesia was explored by using the abdominal constriction test induced by acetic acid.RESULT:Niuhuang Qingwei wan decreased the activity and secretion of pepsin in a dose-dependent manner (P < 0.01, P < 0.05), the gastric juice volume at middle and high doses (P <0.01, P <0.05), and the gastric acid volume at high dose (P <0.05); However, it had no significant effects on the gastric emptying in normal mice and the acidity in gastric juice. It shortened the excreting time of feces and increased the frequency of defecation (P < 0.01, P < 0.05). It also inhibited abdominal constriction responses at high dose, and the inhibition rate was 40.0% (P <0.01).CONCLUSION:Niuhuang Qingwei wan can promote gastrointestinal motility, decrease gastric acid volume and activity of pepsin and show certain analgesia effect. Those findings are consistent with its treating stomach heat in clinic.

Abstract:Qingwei Huanglian Wan (QHW) is a traditional Chinese medicine used for treating inflammation. As the compositions of QHW preparations are complex and varied, it has proven difficult to identify the active ingredients in QHW and control the quality. In this study, high-performance liquid chromatography (HPLC) fingerprints, network pharmacology, molecular docking, and the Griess method were used to screen for and verify the potential active ingredients of QHW. First, sixteen batches of QHW were studied as two groups using multivariate statistical analysis. Thirteen peaks were detected in the HPLC results to establish a fingerprint similarity model, and six chemical constituents (phellodendrine hydrochloride, geniposide, berberine hydrochloride, baicalin, wogonoside, and glycyrrhizic acid) were identified. Among these six constituents, four components (glycyrrhizic acid, berberine hydrochloride, baicalin, and wogonoside) were considered as potential active ingredients. Second, network pharmacology, molecular docking, and the Griess method were used to further elucidate the potential active ingredients. There was no significant difference in the NO content of the active ingredients administration group (dose converted into the medium-dose group) and medium-dose QHW administration group. Then, the contents of berberine hydrochloride, baicalin, wogonoside, and glycyrrhizic acid were determined using the HPLC method. This study provides a comprehensive and reliable strategy for the quality control of QHW preparations and identifies potential active ingredients in QHW. Our methodology may also be useful for studying other traditional Chinese medicines.
Dynamic Analysis on Analgesic Effect of Regulating Qi and Alleviating Mental Depression Prescriptions

Abstract:Objective To investigate the characterstics of the analgesic effect of regulating Qi and alleviating mental depression prescriptions. Methods One hundred and fifty Kunming mice of clean grade, according to randomized blocks by body weight, were divided into 15 groups, including non-treatment model control group(control group), high, middle and low dose groups of Chaiyu decotion(柴郁汤), Shaogan decoction(芍甘汤), Tongxie(痛泻)prescription and Jinlingzi(金铃子)powder, positive control drug Morphine and Ibuprofen groups. A rat pain model was made using tail-immersion in 45 ℃, 50 ℃, 55 ℃ water to induce pain. The pain threshold latency was measured dynamically, measuring the base pain threshold before administration. Analysis of variance of repeated measure definition was applied. Results The analgesic effect of Chaiyu decotion(柴郁汤), Shaogan decoction(芍甘汤)and Tongxie(痛泻)prescription of high doses was more obvious. Pain threshold latency was prolonged by Chaiyu decotion(柴郁汤)in tail-immersion test of 45 ℃, 50 ℃, 55 ℃(P<0.01, P<0.05), the high and middle doses of Chaiyu decotion(柴郁汤)demonstrated good analgesic effect at multiple time points(P<0.05). Conclusion Chaiyu decotion(柴郁汤)has strong analgesic effect, similar to the role of central analgesic drugs; Shaogan decoction(芍甘汤), Tongxie(痛泻)prescription and Jinlingz(i金铃子) powder belong to the peripheral analgesic drugs such as NSAIDs.

Abstract:Introduction: Th study aims to establish the chromatographic fingerprint of Zhachong Shisan pills (ZSP), and to explore its active components and mechanism of analgesic and anti-inflammatory action. Method: First, the Personal Compound Database and Library (PCDL) of ZSP was constructed through literature mining, and then the chemical composition and fingerprint of methanol extractions from 8 batches of ZSP were studied using High-Performance Liquid Chromatography with Quadrupole Time-Of-Flight tandem Mass Spectrometry (HPLC-QTOF-MS) technology. Furthermore, network pharmacology was used to explore the active compounds, potential targets, and signal pathways of analgesic and anti-inflammatory activity of ZSP. A total of 102 compounds were detected in positive and negative ion mode. Results: Fifty-six characteristic peaks in positive ion mode and 78 characteristic peaks in negative ion mode were confirmed as common peaks in the fingerprint of ZSP. Through network pharmacology research, the effective components, key targets, and signal pathways of ZSP for the treatment of cerebral apoplexy by analgesic and anti-inflammatory effects were all analyzed. Conclusion: This study explained the substance basis of the analgesic and anti-inflammatory effects of Zhachong Shisan Pills, and explores its possible mechanisms, providing ideas for rational clinical medication and in-depth pharmacological research.

Abstract:Objective: To explore pharmacodynamic material basis and molecular mechanism of Wutou Tang in pain relief. Methods: Based on the simulation platform of Discovery Studio, the molecular set of Wutou Tang was built and compared with drug set of anti-inflammatory and analgesic actions from DrugBank database in chemical space. After that, network pharmacology based on molecular docking was used to explore the interactions of Wutou Tang and targets related to pain. Results: The molecular sets of Wutou Tang and drugs had better overlaps in chemical space. The main pharmacodynamic material basis of Wutou Tang contains alkaloids, flavonoids, saponins. They could act on six targets including inducible nitric oxide synthase (iNOS), p38, C-Jun N-terminal Kinase 1 (JNK-1), tumor necrosis factor-α (TNF-α), phosphodiesterase 4A (PDE-4A) and Kappa-type opioid receptor. Conclusion: Wutou Tang can mainly act on p38 MAPK and JNK signaling pathways, and has multiple analgesic actions including the inhibition of NO release, the reduction of mechanical hyperalgesia, and the prevention of algesia impulsion conduction and transmission, etc. It has an effect of multi-ingredients, multi-targets and multi-pathways.

Abstract:The analgesic activity of Porcellio laevis Latreille, Rhizoma Corydalis , and Radix Cynanchi Paniculati have been reported in recent years. A new formula named Jia-Yuan-Qing pill (JYQP) is therefore created by combining the three herbs at 9:7:7 ratio according to traditional Chinese theories. The present study aims to evaluate the effect of JYQP as a novel painkiller in various models. Acute toxicity test was applied to evaluate the safety of JYQP. Acetic-acid-induced writhing, hot plate test, formalin test, and naloxone-pretreated writhing test were employed to elaborate the analgesic activity of JYQP and its possible mechanism. A bone cancer pain mouse model was performed to further assess the effect of JYQP in relieving cancer pain. Test on naloxone-precipitated withdrawal symptoms was conduct to examine the physical dependence of mice on JYQP. Data revealed that JYQP reduced writhing and stretching induced by acetic acid; however, this effect could not be blocked by naloxone. JYQP specifically suppressed the phase II reaction time in formalin-treated mice; meanwhile, no analgesic effect of JYQP in hot plate test was observed, indicating that JYQP exerts analgesic activity against inflammatory pain rather than neurogenic pain. Furthermore, JYQP could successfully relieve bone cancer pain in mice. No physical dependence could be observed upon long-term administration in mice. Collectively, our present results provide experimental evidence in supporting clinical use of JYQP as an effective and safe agent for pain treatment.
[Pharmacodynamic substances and therapeutic potential of Wuji Pills:based on UPLC-Q-TOF-MS/MS and network pharmacology]

Abstract:As a classic prescription, Wuji Pills is composed of Coptidis Rhizoma, Euodiae Fructus Preparata, and stir-fried Paeo-niae Radix Alba at the ratio of 6∶1∶6. The practical application of it is limited compared with other famous Chinese medicine prescriptions. Only one company produces Wuji Pills in China. In this study, ultra-performance liquid chromatography quadrupole time of flight mass spectrometry(UPLC-Q-TOF-MS/MS) was used to analyze and identify 26 identical compounds from Wuji Pills and drug-containing plasma of rats. Based on these components, 46 potential targets were screened out with network pharmacology methods, followed by the component-target network construction, Gene Ontology(GO) term enrichment, Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway enrichment, and disease prediction. It was concluded that Wuji Pills acted on core targets such as PTGS2, PTSG1, NCOA2, HSP9 OAD1, and RXRA through magnoflorine, hydroxyevodiamine, daucosterol, and berberine and exerted pharmacodynamic effects through various pathways such as calcium ion signaling pathway, phosphatidylinositol-3-kinase-protein kinase B(PI3 K-Akt) signaling pathway, and vascular endothelial growth factor(VEGF) signaling pathway. Thus, Wuji Pills has therapeutic potential for Alzheimer's disease, diabetes mellitus, myocardial ischemia, and other diseases in addition to the conventional disease(irritable bowel syndrome, IBS). The above research results can provide a reference for the comprehensive interpretation of the pharmacodynamic basis of Wuji Pills and the expansion of clinical application. At the same time, a lot of components in serum and the in vivo transformed and metabolized components of Wuji Pills have similar structure and relative molecular weight. In theory, these components may show additive effects and the competitive/antagonistic effects on the same target. According to the hypothesis of "additive effect of multiple components for a single target" in traditional Chinese medicine, multiple similar components may exert the additive effects on local targets. This study can partly prove the scientificity of this hypothesis and provide laboratory evidence.

Abstract:BACKGROUND: Understanding how pharmaceutical formulas target specific illnesses is crucial for developing effective treatments. Enriching ion channel data is a critical first step in comprehending a formula's mechanism of action. OBJECTIVE: This study aims to explore the effective disease spectrum of the Qi Yu granule formula through network pharmacology analysis and backtracking, and analyze its potential curative effects on liver and spleen system diseases, particularly depression and breast cancer. METHODS: Using pharmacological tools and database analysis, the ion channel data of the formula's components were investigated. The effective disease spectrum was determined, and diseases related to liver and gallbladder, liver depression, and spleen deficiency were identified. Network pharmacology analysis was conducted to backtrack diseases, target gene proteins, and drug compositions. The extraction technology of volatile oil from medicinal herbs was experimentally studied to optimize the preparation process. RESULTS: The effective disease spectrum analysis identified potential curative effects of the Qi Yu granule formula on various diseases, including breast cancer. Backtracking revealed relationships between diseases, target gene proteins, and drug compositions. Experimental studies on volatile oil extraction provided insights into optimizing the preparation process. CONCLUSION: The study underscores the potential therapeutic benefits of the Qi Yu granule formula for liver and spleen system diseases. By integrating network pharmacology analysis and experimental research, this study offers valuable insights into the formulation and efficacy of the Qi Yu granules, paving the way for further exploration and optimization of TCM formulations.

Abstract:Ethnopharmacological relevance Wuwei Shexiang Pills (WWSX), a classic Tibetan medicine, consists of Chebulae Fructus (removed pit), Aucklandiae Radix, Moschus, Aconiti Fiavi Radix, and Acori Calami Rhizoma. It is used clinically in China to treat joint pain, swelling and other symptoms, and has the function of dispelling wind and relieving pain. However, to date, the mechanism of how it works against gout is still unclear. Aims of the study Using network pharmacology, molecular docking and pharmacological verification to explore the potential anti-gout properties of Wuwei Shexiang Pills (WWSX). Materials and methods With the use of UPLC-Q/TOF-MS, the main components of WWSX were obtained and screened for potential anti-inflammatory components by network pharmacology and molecular docking. The anti-inflammatory activity of the components screened from WWSX was also tested by in vitro assays. The anti-gout mechanism of WWSX was predicted by network pharmacology, and the pharmacological validation experiments using gouty arthritis model and mouse air pouch model were used to explore the multifaceted mechanism of WWSX to modify gout. Result Thirty-eight active ingredients were obtained from the UPLC-Q/TOF-MS detection. The network pharmacology and molecular docking analysis showed that 104 co-targets were participated in the treatment of gout, and the main signaling pathways involved were NOD-like receptor pathway, NF-κB pathway and MAPK pathway. Pharmacological evaluation showed that WWSX could significantly improve gout in gouty arthritis models and mouse air pouch models by modulating the above pathways. Conclusion This work has predicted and validated the anti-inflammatory material basis and predicted the anti-gout mechanism of WWSX which was verified by network pharmacology, molecular docking and in vitro cellular studies. The results reveal the mechanism of WWSX in the treatment of gout and provide a theoretical basis for its clinical application.

Abstract:Objective To provide the reference for clinical study of Gentiana odynolysis prescription in Wuwei Medical Bamboo Slips of the Han Dynasty through studying the anti-inflammatory and analgesic effects.Methods Animal models of glacial acetic acid-induced writhing response in mice,xylene-induced ear edema in mice,and carrageenan-induced paw edema in rat were used to observe the effect of anti-inflammation and analgesia of Gentiana odynolysis prescription.Results Gentiana odynolysis prescription had significant effects in anti-inflammation,analgesic and other pharmacological effects compared with control group(p<0.01).Conclusion Gentiana odynolysis prescription has significant effects of anti-inflammatory and analgesia.
Study on Pharmacodynamics of uzhi Dieda Ointmeat

Abstract:Three models were established in this research,including mouse auricular swelling model induced by xylene,mouse pain model induced by intraperitoneal injection of acetic acid and rabbit fracture model. The effects of Wuzhi dieda ointmeat on anti-inflammation,analgesia,promoting fracture healing were observed.The results showed that Wuzhi dieda ointmeat significantly inhibited the auricular swelling in-duced by xylene and intraperitoneal injection of acetic acid-induced pain in mice.Meanwhile,the ointment promoted the healing of fracture in rabbits.

Abstract:Objective It is to observe the antitussive,expectorant and antiashmatic actions of Wuwei Shaji pulvis.Methods The cough models induced by sulfur dioxide in mice and citric acid in guinea pigs were used to observe the antitussive effects.The methods of phenol red excretion in mice and phlegm displacement of rats were used to investigate the expectorant effects.Bronchial asthma model induced by histamine-acetylcholine in guinea pigs was used to observe the antiasthmatic effects.Results The Wuwei Shaji pulvis could obviously decrease the frequency of cough induced by sulfur dioxide in mice and by citric acid in guinea pigs and prolong the latent period of cough.It could obviously increase phenol red output of trachea in mice,promote expectoration in rats,inhibit the asthma induced by histamine-acetylcholine in guinea pigs.Conclusion The Wuwei Shaji pulvis has significant effects of antitussive,expectorant and antiasthmatic.

Abstract:Objective It is to study the influence of Nuangong Qiwei pill on uterus function and the analgesic effect.Methods The influence of drugs on uterine contraction of rat in vivo and in vitro was observed.The analgesic effect of Nuangong Qiwei pill was observed through spastic pain of female rats induced by Estradiol Benzoate and oxytocin and writhing response induced by acetic acid pain.Results Nuangong Qiwei pill can relax uterine smooth muscle of rat in vivo and in vitro and had obvious analgesic effect.Conclusion The influence of Nuangong Qiwei pill on uterine smooth muscle and the analgesic effect are important pharmacological bases for treating irregular menstruation,lumbago due to sluggishness of qi and lower abdomen cold pain.

Abstract:To research the pharmacologic effects of Mongolian Medicine Yunxiang-15weiwan for anti-in- flammatory and anti-fungus.Methods:The experimental animal's model of acute and chronic inflammatory.Re- suits:The tests indicated the Yunxiang-15weiwan could restrain distinctly tumefaction of the mice's ear outline and can restrain dermatophtosis.Conclusion:It is obvious that Yunxiang-15weiwan has the effects of anti-in- flammatory and anti-fungus.
