Function of the Lateral Root Development Relating Gene RSI-1 from Tomato in Tobacco Morphogenesis

Abstract:To investigate the function of the lateral root primordium development relating gene RSI-1 and the application possibility in super-root crops breeding, plasmids pBI-SRSI and pBI-ARSI that would lead to constitutive transcription of sense RSI-1 and anti-sense RSI-1 in plant cells and the RSI-1 RNAi interfere vector p1008-RSI-RNAi were constructed and transferred to tobacco (Nicotiana tabaccum) respectively. The transformants with the target chiermic gene constitutively transcribed were screened out by antibiotics medium selecting and Sourthern blotting. Then the morphological variations of the aboveground tissues were analyzed,and the change of the phytohormones involving in rooting was also studied. It was shown that divarication and lateral root formation was enhanced in the transgenic tobacco plants with RSI-1 over-expressed, while the morphology and the contents of gibberellic acid, indole-3-acetic acid, abscisic acid, zeatin and its ribosides were not altered significantly in the tobaccos with the RSI-1 antisense inhibited and RNA interfered. Based on the working mechanism of the function gene over-expressing, antisense inhibiting and RNA interfering, the evidences further conformed that RSI-1 is a gene involved in divaricating and it may be lowly conservative in evaluation. Thus, the root-specific promoters are needed when RSI-1 is used to modify root-deficient crops.
