Abstract:Objective: Evidence for the importance of genetic factors in male fertility is accumulating. It is most likely due to the obstacle of spermatogenesis. Cloning of new testis-specific gene related to spermatogenesis is of momentous physiological and pathological significance to understand the molecular mechanism of spermatogenesis. Design: The digital differential display (DDD) program of the National Center for Biotechnology Information (NCBI) was performed between seven testis libraries (“tester”) and seventy-six libraries derived from other tissues (“driver”). In this process,tags in testis libraries and multi-tissue pooled libraries were quantitatively compared by statistics analysis. Select a new expression sequence tags (ESTs) result from DDD which was unique to human testis libraries. To validate the use of DDD approaches in gene discovery, the ESTs which was predicted to be testis-specific, was chosen for further study. Multi-tissue reverse transcriptase polymerase chain reaction (RT-PCR) analysis of mRNA was performed. Use bio-information to obtained the full-length cDNA of the ESTs, PCR technology and sequencing confirm the predicted new coding frame. Perform semi-quantitative RT-PCR analysis of mRNA from diffrental normal tissues and sperm to clarify express pattern of the new gene. Materials and Methods: Using the DDD program, we identified a new contig of the ESTs which were unique to testis libraries and represented a novel gene. Multi-tissue RT-PCR analysis indicated that the contig was specifically expressed in human testis. The full-length cDNA of the new gene was obtained through BLAST program. Gene-specific primers were designed for RT-PCR in human testis. Sequencing was performed and the sequcece result was analysed. Semi-quantitative RT-PCR analysis of mRNA from diffrental normal tissues and sperm was performed to clarify the expression pattern of the gene. Results: A novel full-length cDNA sequence-SRG5 in human were identified (GeneBank accession number AY221117), SRG5 was 2430 bp in length, located in chromosome 3p21.1. The sequence of the opening reading frame was 676∼1248 bp, as was confirmed by RT-PCR and sequencing in human testis. The cDNA encodes a novel protein of 190 amino acids with a theoretical molecular weight of 20417.8 and isoelectric point of 5.23. The sequence share no significant homology with any known protein in databases. Semi-quantitative RT-PCR analysis of multiple tissues showed that SRG5 is expressed significantly in human male testis and sperm. Sequencing result confirm the predict nucleotide sequence. Conclusion: Identification of SRG5 using DDD approaches validates gene discovery using computational approaches.We hypothensize that SRG5 functions as a testis-specific and spermatogenesis relate gene that plays the roles in spermatogenesis.
