Abstract:Hami melon"Xizhoumi 25" was used in this experiment. Melons were dipped in different concentra-tions of oxalic acid (OA ) with 10, 15 mmol/ L and 20 mmol/ L (add 0.5 mL/L tuwen-20)and melons were dipped in normal temperature distilled water was control at 25 ℃ for 10 min, after the surface were dried com-pletely and stored at 3 ℃-5 ℃ for 42 days, chilling injury index and quality in fruits were measured during the storage period. Compared with that in control, OA treatment inhibited the decrease of firmness and ascorbic acid (ASA)contents and increased the contents of total soluble solids from the initial storage to 21 day (P < 0.05), in this respect, 15 mmol/L OA was the most effective treatment. In addition treatment with 15 mmol/L OA inhib-ited the increase of respiration rate and electrolyte leakage, reducing the chilling injury index of Hami melon. But 20 mmol/L OA promoted the chilling injury index and electrolyte leakage, meanwhile which restrained the decline of chlorophyll content compared to the controls. The results indicated 15 mmol/L OA was the most effec-tive treatment to reduce the CI(chilling injury index)and extend the storage quality.

Abstract:The Hami melon (Cucumis melo var. reticulatus Naud.) is a tropical fruit with a short storage period. To prolong freshness, the fruit is stored at low temperatures (3 degrees C), although this can lead to chilling injury (CI) involving skin lesions, numerous tiny brown spots on the rind of the fruit, susceptibility to decay, especially upon re warming, the formation of small pits, and browning of the surface associated with a loss of sensory quality. Our preliminary research revealed that pretreatment with 15 mmol L-1 oxalic acid (OA) for 10 min at 25 degrees C effectively reduced CI in Hami melons stored at 3 degrees C. The current work was conducted to confirm the above results and determine whether enzymes involved in antioxidative pathways play a role in the decreased CI. The activities of enzymes related to antioxidative pathways and expression levels of the corresponding genes were measured. OA pretreatment prior to storage at 3 degrees C for 42 d increased the activities of glutathione reductase (GR), ascorbate peroxidase (APX), and peroxidase (POD), and led to higher expression levels of the Cm-GR, Cm-APX, and Cm-POD genes relative to the control group. The OA-treated group also maintained higher contents of ascorbic acid (MA) and glutathione(GSH) compared with the control group. OA treatment inhibited H2O2 and O-2(-) accumulation, delayed membrane lipid oxidation, and led to a lower CI index compared with the control fruit. These results indicate that enzymes involved in antioxidative pathways play a role in the alleviation of CI upon OA treatment.

Abstract:The effects of freezing modes and time in frozen storage on aroma, enzymes and micro-organisms in Hami melon, also called cantaloupe melon (Cucumis melo. var. reticulatus Hami melon), were determined. Freezing process and frozen storage could not significantly reduce either micro-organisms counts or the enzymatic activities of polyphenoloxidase (PPO), peroxidase (POD) and lipoxidase (LOX) in melon. Liquid nitrogen ultra-rapid freezing maintained most aromatic components of the melon as compared with slow freezing. The mechanism of aromatic change in frozen melon was mainly ascribed to esters changing continuously in variety and concentration because of enzymolysis or synthetic reactions of esters catalysed by LOX. Meanwhile, increasing concentration of linoleic acid and linolenic acid in frozen melon cells promoted the biochemical reaction of unsaturated fatty acids in LOX system, so that enzymolysis of the double bonds in the ninth and tenth carbon of linoleic acid and linolenic acid was accelerated. Consequently, with the prolongation of time in frozen storage more and more unsaturated alcohols and aldehydes with 6 and 9 carbons were produced, the green notes of frozen storage melon became more and more intense, and the ester fragrance became more and more feeble. The above findings can benefit the development of cantaloupe melon process industry.

Abstract:Rapid postharvest softening largely limits the shelf-life of persimmon (Diospyros kaki L.) fruit. Microwave is a new environmental-friendly inside-out heat-shock approach, whose effect on the fruit softening and quality has not yet been investigated. The current study applied two kinds of microwave treatments (low-power long-time, LPLT, or high-power short-time, HPST) to persimmon fruit with comparison to the hot water (HW) treatments. The results showed both microwave treatments maintained firmness, facilitated the deastringency, and increased soluble solid contents (SSC) and sugar-acid ratio of persimmon fruit. The microwave treatments reduced the cellulose and pectin degradation, and inhibited the cellulase activity, resulting in a significantly higher firmness than HW treatment and control after 2 and 4 days of storage. Moreover, application of HPST treatment down regulated gene expression of DkPG1, DkPE2 and DkEGase1 compared with untreated fruits. These results indicated that microwave treatment is a promising soft-delaying method for the preservation of persimmon fruit.

Abstract:The polyphenol oxidase(PPO) inactivation by microwave irradiation was compared with steam blanching and hot-water blanching.The rudimental conditions of enzyme inactivation by microwave were investigated.The results showed that enzyme inactivation was much faster and the quality of banana slice was better by microwave irradiation than by steam blanching and hot-water blanching.When the power density of microwave was 4w/g,the activity of PPO could be decreased to below 5% in 5 min.The effect of thickness of banana slice on enzyme inactivation by microwave was insignificant,and the banana could be treated by monolayer or double deck under microwave irradiation.

Abstract:Abstract Various applications of microwave have been put into effect in the field of food processing. The microwave assisted thermal sterilization (MATS) was of great significance because of its less time consuming and higher efficient. In this paper, response surface methodology was used to optimize the process. The sterilization of orange sacs was potimiazed using by response surface methodology (RSM). The optimal processing parameters of MATS on orange sacs are as follow: microwave power, 6 kW, microwave sterilization time, 350 s, amount of orange sacs, 4500 g. Sterilized under this condition by MATS, orange sacs only maintained 3.7 CFU lethal magnitude which was consistent with the standards requirement. The key factor in the response surface using microwave sterilization and the interaction between them were also determined and discussed which could make it evident that MATS should be an idea candidate in the sterilization of some fruit or vegetable products as ready to eat. From these results, the best conditions of microwave sterilization are power 6kw, microwave sterilization time 350s and material quantity 4500g.

Abstract:Objective:This paper probed into the effect of microwave treatment on the phenolic acids and flavanone glycosides contents and antioxidant capacity of Huyou peels.Methods:Seven main phenolic acids and four flavanone glycosides were determined by HPLC-PDA.Folin-Ciocalteu method,ABTS free radical-scavenging assay and FRAP assay were adopted to evaluate the antioxidant capacity of methanol extract of Huyou peels.Results:After microwave treatment,the content of free phenolic acid portion increased(P<0.05),while the content of total phenolic acids decreased(P<0.05),and the content of total flavanone glycosides also reduced(P<0.05).Besides,after microwave treatment,the antioxidant capacity of methanol extract of Huyou peels increased(P<0.05),perhaps because of the increase of free phenolic acid portion and flavanone glycosides'decomposition to the corresponding flavanone aglycons with higher antioxidant capacity.Conclusion:Antioxidant capacity of Huyou peels can be obviously elevated by microwave treatment.

Abstract:SummaryThe effects of different thawing methods (room temperature thawing, running water thawing, microwave thawing at 300 W, 500 W, 700 W and high‐pressure thawing at 100 MPa, 150 MPa, 200 MPa) on quality (drip loss, texture, colour, ascorbic acid contents and sensory quality) of frozen hami melon were investigated. The results showed that different thawing treatments affected the quality of hami melon significantly (P < 0.05). Samples thawed by microwave at 500 W best preserved ascorbic acid for nearly half of fresh hami melon. Less drip loss, texture damage and discolouring were observed for the samples thawed by microwave at 700 W than other thawed samples; however, serious ascorbic acid loss was detected. High‐pressure‐thawed samples exhibited shorter thawing time but severe quality losses. The results suggested that microwave thawing at 500 W was an appropriate method to thaw frozen hami melon with the highest sensory score (6.7).

Abstract:ABSTRACT In this study, we investigated the change of the flavor, enzyme activities and microorganism survival in cantaloupe (Cucumis melo L.) juice after UHP treatments, and found the corresponding kinetic models. After the treatment at 500 MPa for 20 min, the pressurized juice can reach the safety standards, i.e., below 100 cfu/100 mL in concentration stipulated for beverage by Chinese national standard; and the activities of POD, PPO and LOX in the juice decreased to about 78, 9 and 5%, respectively. Sensory evaluation indicated that no significant change was observed after UHP treatments. The above findings indicated that UHP treatment is a promising way to process the cantaloupe juice.PRACTICAL APPLICATIONCantaloupe is a favorable fruit with unique aroma produced around the world. However, because there is no effective deep‐processing, a lot of cantaloupes rot away in the farmland every year. The production of freshly squashed juice is a best way to raise the merchandise rate of cantaloupe and to prolong its industrial chain. However, freshly squashed cantaloupe juice has a very short shelf life because it will quickly succumb to microorganism attack if nothing is done. The sterilization of its juice is a difficult issue because the juice is heat sensitive. Therefore, nonthermal processing is desirable. UHP technology used in this study was proven to be a promising way to maintain the flavor and to inactivate enzymes and microorganisms in the cantaloupe juice.

Abstract:The effects of dense phase carbon dioxide (DP-CO2) treatment of 8, 15, 22, 30 and 35 MPa for 5 min, 15 min, 30 min, 45 min, 60 min at 35 degrees C, 45 degrees C, 55 degrees C, 65 degrees C on microorganism, enzyme, and aroma compounds in hami melon juice during storage at 4 degrees C for 4-weeks were investigated. Meanwhile, the color, browning degree, and Vitamin C were also studied. The DP-CO2 treatment had significant effects on inactivation of microorganism and enzyme. It was indicated that higher pressure caused more inactivation of microbial total count and enzyme activity. When it reached 35 Mpa, 55 degrees C, 60 min, the microorganism was totally inactivated. The least residual activity of polyphenol oxidase (PPO), peroxidase (POD), and lipoxygenase (LOX) was 25.26%, 38.46 and 0.02% at 35MP, respectively. The restoration of PPO, POD and LOX residual activity after DP-CO2 treatment was also observed, which was dependent on the pressure level. The aroma compounds were less affected after being treated with DP-CO2, and the flavor of the melon juice was close to the fresh juice after storage at 4 degrees C for 4 weeks and did not produced cook off-odor. The changes of lightness L and browning degree A during storage were well fitted to a first-order kinetic model. The Vitamin C concentration decreased by DP-CO2 processing, but this loss was lower than of the untreated sample.Industrial relevance: Hami melon is highly appreciated for its nutritional quality and special flavor. The flesh of melon is heat sensitive, the sensitive nutrients, color and aromatic profile will be spoiled greatly or off-odour when it was produced with high temperature treatment. Dense phase carbon dioxide processing (DP-CO2) is important to find an innovative food process to inactivate the enzyme and microorganism and protect the nutrient and unique flavor. In this study, the data proved that DP-CO2 processing is a promising non-thermal alternative pasteurization to preserved fresh-squeezed melon juice. (C) 2010 Elsevier Ltd. All rights reserved.
The effects of high-power microwaves on the ultrastructure of Bacillus subtilis

Abstract:Aims: To investigate the microbicidal mechanisms of high-power microwave (2.0 kW) irradiation on Bacillus subtilis and to determine the effect of this procedure on the ultrastructure of the cell wall. Methods and results: We performed viability test, examined cells using transmission electron microscopy (TEM), and measured the release of intracellular proteins and nucleic acids. The inactivation rate of B. subtilis by 2.0-kW microwave irradiation was higher than that of a domestic microwave (0.5 kW). Few proteins were released from either microwaved or boiled cells. However, the leakage of nucleic acids from 2.0-kW-microwaved cells was significantly higher than that of 0.5-kW-microwaved or boiled cells. Therefore, we examined ultrastructural alterations of microwaved or boiled cells to analyse the pattern of release of cytoplasmic contents. Although boiled cells did not show any ultrastructural changes on TEM, 2.0-kW-microwaved cells showed disruption of the cell wall. Conclusion: The microbicidal mechanisms of 2.0-kW microwave irradiation include damage to the microbial cell wall, breakage of the genomic DNA, and thermal coagulation of cytoplasmic proteins. Significance and impact of the study: TEM images showed that the cytoplasmic protein aggregation and cell envelope damage by microwave irradiation were different from the ultrastructural changes observed after boiling.

Abstract:Continuous-flow microwave pasteurization provides important advantages over conventional heat exchangers such as fast volumetric heating, lower tube surface temperature, and possible non-thermal effects that enhance enzymatic and bacterial inactivation. Conventional and microwave-assisted inactivation of pectin methylesterase (PME), polyphenol oxidase (PPO), and peroxidase (POD) in cloudy apple juice were investigated to evaluate non-thermal effects. Experiments were conducted to provide uniform heating with accurate temperature acquisition and similar temperature profiles for conventional and microwave treatments. A two-fraction first-order kinetic model was successfully fitted to the data in a procedure that took into account the whole time-temperature profile instead of assuming isothermal conditions. Predicted inactivation curves for pasteurization at 70 and 80 °C of the cloudy apple juice showed that PME has the highest thermal resistance (residual activity of 30% after 250 s at 80 °C) and that there was no evidence of non-thermal microwave effects on the inactivation of these enzymes.

Abstract:The effects of Conventional Heating (CH) and Microwave Irradiation (MWI) pre-treatment of Egg White Proteins (EWPs) on antioxidative activity of their hydrolysates prepared with pepsin were investigated. MWI pre-treatment of EWPs resulted in a higher degree of hydrolysis (DH) and the highest hydroxyl radical scavenging activity (40.67±1.30%), superoxide anion scavenging activity (75.53±2.79%), inhibitory activity of tea oil autoxidation (67.68±2.50%) and reducing power (0.34±0.01) of Egg White Proteins Hydrolysates (EWPHs) in comparison with CH pre-treatment, although there was no significant difference in DPPH radical scavenging activity between the EWPHs originated from the proteins pre-treated by CH and MWI (p>0.05). According to the experimental results, MWI pre-treatment could improve the antioxidant activities of EWPHs compared with CH treatment.
Effect of High Pressure Carbon Dioxide Processing on Microbial Population and Quality of Fresh-Cut Hami Melon(Cucumis Melo Var.saccharinus)

Abstract:This study aimed to determine the most suitable cleaning process for Hami melon. The changes in microbial populations (total aerobic bacteria (TAB), and total yeast and mold counts), sensory (mass loss, texture and sensory score) and physicochemical properties (total soluble solid (TSS) content, pH, vitamin C and polyphenoloxidase and peroxidase activity) of high pressure carbon dioxide (HPCD)-treated Hami melon during storage at 4 ℃ for 8 days were investigated. Results showed that treatment with strong acid electrolyzed water for 25 min was the best cleaning process. It could reduce TAB of Hami melon by 0.81 (lg (CFU/g)). During the storage, the number of microorganisms of fresh-cut melon HPCD-treated at 2.5, 3.5, 4.5 MPa for 8 min increased significantly and PPO activity also increased. In addition, fruit mass, hardness and ascorbic acid content were decreased respectively by 16.45%-42.70%, 30.38%-48.97% and 96.25%-96.73% after 8 days of storage. This study can provide a theoretical basis for further exploration and application of HPCD on fresh-cut fruits and vegetables.

Abstract:In this study, the thermal and non-thermal effects of microwaves on the enzymatic inactivation of pectin methylesterase in the tomato pulp were validated. An amount of 30 kg of tomato pulp was subject to two different types of treatments: thermal effects and then non-thermal effects using microwaves, implementing a system that allowed simultaneous cooling of microwave heating. An experimental rotational central composite design was used to analyze the effect of the interactions of the variables involved in the process. The activity analysis of the PME enzyme, pH, titratable acidity, total soluble solid, and color was performed on both processed and non-processed tomato pulps, which showed that the heating process with thermal effect did not significantly change physical-chemical properties and were found to be more effective, reaching the enzymatic inactivation of 99% and the temperature of 80oC after 425 s of processing. However, the non-thermal analysis with temperatures at a maximum of 40oC attained 56.8% of enzymatic inactivation.

Abstract:A series of experiments involving microwave irradiation were carried out to evaluate the effect of microwave irradiation on enzymatic hydrolysis of rice straw. Compared with microwave irradiation free hydrolysis, rice straw pretreated by combining microwave irradiation with alkali could increase the initial hydrolysis rate but the hydrolysis yield remained unchanged. When the enzyme solution was treated by microwave irradiation, the initial hydrolysis rate increased slightly, but the yield was decreased remarkably. Its optimal hydrolysis conditions were temperature (45°C), pH (4.8) and enzyme loading (20mgg−1 substrate), which was determined by an orthogonal experiment. When intermittent microwave irradiation was used, initial hydrolysis rate was greatly accelerated but the yield was decreased slightly. Its optimal hydrolysis conditions were temperature (50°C), pH (4.8) and enzyme loading (20mgg−1 substrate), which was determined by another orthogonal experiment.
Effect of glucose oxidase treatment on the aroma qualities and release of cooked off-odor components from heat-treated Hami melon juice

Abstract:Melon juice produces strong cooked off-odors during heat processing, leading to serious deterioration of aroma quality. In this work, the aroma quality of melon juice, the changes in GOD reaction products, and the interactions of reaction products and cooked off-odor components were analyzed by sensory evaluation, gas chromatography-mass spectrometry/olfactory, ultraperformance liquid chromatography-triple quadrupole mass spectrometry, and isothermal titration calorimetry to study the effect mechanism of glucose oxidase (GOD) on the release of cooked off-odor components from heat-treated melon juice. The results showed that GOD treatment improved the aroma quality mainly by controlling off-odor attributes and maintaining characteristic odor attributes. This was because the reaction products (hydrogen peroxide and gluconic acid) of GOD treatment inhibited the release of cooked off-odor components from heat-treated melon juice through oxidation and hydrophobic effects. Furthermore, these products reduced the loss of characteristic odor compounds by restraining Maillard, degradation, and oxidation reactions during heat processing.
Influence of Gamma Irradiation on Enzyme, Microorganism, and Flavor of Cantaloupe (cucumis Melo L.) Juice

Abstract:ABSTRACT: The production of freshly squashed juice is a best way to raise the merchandise rate of cantaloupe and to prolong its industrial chain, and as a low acid and heating sensitive juice how to ensure its safety and quality is a key point. In this study, enzyme activity and microorganism survival in Golden Empress cantaloupe juice after 60Co irradiation were measured and analyzed, and the irradiation off‐odor intensity was evaluated with the Kramer sensory evaluation method. Sensory evaluation indicated that the juice had a slight irradiation off‐odor after treatment at 1 kGy, and had strong off‐odor at 2 kGy and above; therefore the dosage should be less than 1kGy in order to keep the off‐odor in acceptable scope. Enzyme activity determination indicated that lipoxygenase was the easiest one to be inactivated by irradiation, followed by polyphenoloxidase and peroxidase, but the 3 enzymes still remained activities even at 5 kGy. Microorganism survival determination indicated that Escherichia coli was sensitive to irradiation, and can be reduced by 7 log‐cycles at 1 kGy, whereas total colony and target spore bacteria in the juice had stronger endurance to the irradiation, suggested by D10 values of 0.9908 and 1.1923 kGy, respectively, indicating both total colony and target spore bacteria were difficult to inactivate as compared to E. coli. The study indicated that gamma irradiation can not completely inactivate the enzymes, total colony, and target spore bacteria in cantaloupe juice on the premise of acceptable off‐odor.
Microwave Irradiation: Effect of Β-Glucosidase Activity by Modulating Its Structural Properties in Grape Maceration Stage

Abstract:In this paper, microwave as an auxiliary method could activate the 8-glucosidase (8 -GC) activity in grape maceration stage. 8 -GC had higher tolerance to acid and temperature during winemaking process. Microwave parameters were optimized for using response surface methodology (RSM), variables including microwave power, temperature, and time. The results demonstrated that the regression equation obtained by using the quadratic regression orthogonal model could better simulate actual operating conditions, the 8 -GC activity increased by 12.54% under optimum conditions (700 W, 48 degrees C, and 6.9 min) comparing to the untreated 8 -GC. The changes in 8 -GC structural properties confirmed that microwave irradiation did increase 8 -GC activity by affecting 8 -GC's side chain group, hydrolysis degree, microenvironment, stability, composition, and conformation. The study would contribute to understanding the mechanism of aroma enhancement by microwave maceration.

Abstract:This research explored microwave treatment impact on the structuro-functional aspects of oxidized soy protein aggregates (OSPI). Data showed that oxidative treatment promoted the formation of high molecular weight aggregates through hydrophobic interactions, thereby disrupting the structure of natural soy protein isolates (SPI). Microwave treatment for an appropriate time (≤30 s) caused the molecular structure of OSPI to open up and reduction in molecular weight and disulfide bond content, while absolute zeta potential increased. These modifications increased emulsifying capacity of OSPI, as well as the interfacial adsorption of protein. Longer microwave treatment times (>30 s) caused OSPI to exhibit a tendency to aggregate in TEM and CLSM images. It indicated the appropriate microwave electromagnetic field effect and microwave heating effect could coordinatively regulate soy protein functional properties by modifying their aggregation behavior. The results provided new ideas for reducing resource waste, and further expanding soy protein application in the food industry.
