Abstract:N-Substituted 1,2,3-triazole is an important structural unit in organic chemistry, and has widely utilized in organic synthesis, medicinal chemistry and material science. The chemistry of N-1-substituted 1,2,3-triazoles has attracted much attention from organic chemists, while the synthesis and application of their N-2-isomers have been far less explored. The recent progress on the research field of N-2-alkyl, allyl, propargyl, vinyl, aryl substituted 1,2,3-triazoles since year 2000 is summarized, including some research of our group. The content is classified by different synthetic methods, such as selective functionalization of 1,2,3-triazoles and oxidative cyclization of bisarylhydrazones or azobenzenes.

Abstract:s-Triazine has special cycle aromatic structure and can constitute various trisubstituented s-triazine derivatives, which possess star-shaped molecular structure, by linking directly or indirectly with hydrocarbon group. These new trisubstituented s-triazine derivatives were widely applied in function materials, medicine and other fields for their good physical and chemical properties. In this paper, the progresses in the synthesis of trisubstituented s-triazine derivatives were summarized by cyclotrimerization of nitrile compounds and side-chain modification of trisubstituented s-triazine.

Abstract:We have developed a novel and efficient method for the preparation of 2,4-disubstituted-1,2,3-triazoles. These compounds 3a-3p were synthesized in good yield by refluxing alpha-aminoacetophenones and hydrazines in glacial acetic acid. The reaction proceeds under mild conditions and is compatible with various functional groups.

Abstract:During the past few decades, there has been considerable growth in the development of denitrogenative reactions of triazole skeletons to construct valuable cyclic compounds, particularly heterocycles. Despite the inherent difficulty of the ring-opening of triazole derivatives, many novel and efficient approaches have arisen in this area mainly with the use of transition metal (such as rhodium, palladium, silver, copper) catalysis, photolysis, or free radical mediated reactions. Generally, these procedures begin with the ring-opening of 1,2,3-triazoles or benzotriazoles followed by N2 extrusion and subsequent diverse transformations, which enable the rapid synthesis of various heterocycles in a single step. To avoid overlap with other related reviews, this minireview covers the recent advances in the denitrogenative cyclization of 1,2,3-triazoles since 2016 and the denitrogenative cyclization of benzotriazoles since 2012.

Abstract:NH-1,2,3-Triazoles are a kind of N-containing heterocylic compounds with important biological activities, which have found broad use in medicinal intermediates. Besides, NH-1,2,3-triazoles serve as valuable synthetic intermediates for the preparation of 2-substituded-1,2,3-triazoles through N-alkylation reaction. Because of their potent applications in both synthetic organic chemistry and medicinal chemistry, the progess in the synthesis of NH-1,2,3-triazoles, especially those reported in the past decade, is surveyed according to the research by our group and other groups. Some classical reaction mechanics are also reviewed in this paper.

Abstract:The reactions of α-bromoacetophenones with methylhydrazine in refluxing acetic acid generated 2-methyl-4-aryl-2H-[1,2,3]triazoles in good yields. The method was developed by the reactions of α-bromoacetophenones with phenylhydrazines in the presence of cupric ion, leading to 2,4-diary-2H-[1,2,3]triazoles. The structures were established on the basis of corresponding IR, 1H NMR, and elemental analysis data.

Abstract:This feature article aims to review the evolution and recent advances in different protocols for synthesizing fused hybrids of N‐heterocycles, specifically [1,2,3]triazolo[1,5‐a] quinoline. Approaches to synthesize this unit will be discussed, including dehydrogenative cyclization, N−N oxidative coupling, Dieckmann condensation, intramolecular Heck reactions, (3+2) cycloaddition, Ullmann‐type coupling, and direct intramolecular arylation reactions. This report outlines the evolution and recent progress about the different protocols to synthesize the N‐heterocycles fused hybrids, specifically [1,2,3]triazolo[1,5‐a]quinoline. This review encompasses a broad range of approaches, describing several reactions for obtaining this since, such as dehydrogenative cyclization, oxidative N−N coupling, Dieckmann condensation, intramolecular Heck, (3+2)‐cycloaddition, Ullman‐type coupling and direct intramolecular arylation reactions. We divided this review in three section based in the starting materials to synthesize the target [1,2,3]triazolo[1,5‐a]quinolines. Starting materials containing quinoline or triazole units previously formed, as well as starting materials which both quinoline and triazole units are formed in situ. Different methods of obtaining are described, such as metal‐free or catalyzed conditions, azide‐free, using conventional heating or alternative energy sources, such as electrochemical and photochemical methods. Mechanistic insights underlying the reported reactions were also described in this comprehensive review.

Abstract:A series of novel 1,2,4-triazole derivatives were synthesized, and their structures were characterized by IR, UV-Vis, FL, NMR, ESI-MS, and elemental analysis. In the meanwhile, the single crystal structures of 3,4-diethyl-5-(4-pyridyl)-1,2,4-triazole and 3,4-dimethyl-5-( o -hydroxyphenyl)-1,2,4-triazole were determined by X-ray diffraction.

Abstract:Dibenzotriazonine represent a new class of nine-membered cyclic azobenzenes with a nitrogen atom embedded in the bridging chain. To enable future applications of this photoactive backbone, we propose in this study the synthesis of mono- and dihalogenated triazonines, that allow the late-stage introduction of different functionalized aryl groups and heteroatoms (N, O, and P) via palladium-catalyzed reactions. Indeed, different diphenylphosphoryl-triazonines were synthesized with functional groups such as aniline or phenol. Bis(diphenylphosphoryl)phenyl mono- and bis-carbamate-triazonines were also isolated in good yields.

Abstract:Seven novel 2-aryl-5-[(4-arylthiazol-2-yl)methyl]-1,3,4-oxadiazoles(7a~7g) were synthesized by cyclization of N,N′-diacylhydrazines(6) in the presence of POCl3.6 were obtained from thioamides and α-bromoketones by a four-step reaction.The structures of 6 and 7 were characterized by 1H NMR,IR,MS and elemental analysis.

Abstract:The solvent‐free synthesis of 1,2,4‐triazols from N‐tosylhydrazonyl chlorides and nitriles at room temperature was developed. The transformation was suitable for a wide spectrum of substrates, and the acid‐promoted cyclization mechanism was proposed. 1,2,4‐Triazols represent important heterocyclic compounds which have found extensive application in the pharmaceutical agrochemical and material industries. Herein, we reported an acid‐promoted solvent‐free synthesis of 1,2,4‐triazols from N‐tosylhydrazonyl chlorides and nitriles at room temperature. Various N‐tosylhydrazonyl chlorides and nitriles were examined, and the transformation was suitable for a wide spectrum of substrates. Two simple acid‐promoted cyclization pathways were proposed for the transformation.

Abstract:Carbazole and its derivatives are a class of important nitrogen-containing heterocyclic compounds possessing various unique properties and biological activity. Herein the current progress of their synthetic strategies reported in the past 5 years is reviewed. The representative examples, which have been divided into four categories according to the usage of different key intermediates, are selected and discussed in detail. Special emphasis is put on the newly developed cyclization reactions which may be useful for the construction of the tricyclic backbone of carbazoles.

Abstract:Fourteen new 2-acylhydrazino-5-(2,4-dichlorophenyl)-1,3,4-oxadiazoles were prepared by nucleophilic displacement of acylhydrazine at the 2-position of 2-methylsulfonyl-5-(2,4-dichlorophenyl)-1 ,3,4-oxadiazole. Fourteen new 3-substituted-6-(2,4-dichloro phenyl)-S-triazolo [3,4-b]-1,3,4-oxadiazoles were obtained by treating 2-acylhydrazino-5-(2,4-dichloro-phenyl)-1,3,4-oxadiazoles with K2CO3 solution. The structures of new compounds were determined by elementary and spectral analyses, All the new compounds showed a significant antibacterial activity.

Abstract:N-heterocyclic compounds have a wide range of applications in medicinal chemistry, bioconjugation and materials chemistry due to their high biological activity, low toxicity and high systemic nature. Dibromo-triazoles and their amino derivatives are important functional materials. 3,5-Dibromo-4H-1,2,4-triazole (1, yield 74%) can be easily synthesised from 1H-1,2,4-triazole (2) using bromine, N-bromosuccinimide or other brominating agents in a high yield. The 3,5-dibromo-4-amino-1,2,4-triazole and 3,5-dibromo-1-amino-1,2,4-triazole (3, yield 21%) can be prepared by reacting compound 1 with hydroxylamine-O-sulphonic acid in weakly alkaline aqueous solutions at reflux temperature. Pure compound 3, a colourless solid, was finally obtained after a column chromatography. 4,5-dibromo-1-amino-1,2,3-triazole and 4,5-dibromo-2-amino-1,2,3-triazole (4, yield 30%) can also be prepared with 1H-1,2,3-triazole (5) as the starting reactant and 4,5-dibromo-1H-1,2,3-triazole (6, yield 78%) as the intermediate. The structures of the compounds above were confirmed using H-1-NMR, C-13-NMR, infrared (IR), LC-MS, XRD and elementary analysis. According to the characterisation results, these compounds have two isomers.
