In the summer of 2017, leaf blight symptoms were observed on Lanzhou lily in Qilihe District, Lanzhou, resulting in approximately 30 to 40% loss of bulb production in continuous cropping fields. The infection initially manifested as elliptical or irregular oval lesions on the leaves, which gradually expanded and coalesced into blight of grayish brown. Infection often started at the leaf tips and progressed to the base of leaves as symptoms developed. Severely infected leaves were wilted and covered with gray mycelium and conidia, especially under humid conditions. To isolate the pathogen, small pieces of symptomatic leaves (0.5 × 0.5 cm in size) were surface sterilized with 70% ethanol for 30 s, followed by 2.5% sodium hypochlorite for 1 min, and then rinsed three times with sterile distilled water. The disinfested tissues were cultured on potato dextrose agar (PDA) and incubated at 20°C. The pure cultures were obtained by single conidia isolation. The isolated fungus was characterized by forming loosely villiform, grayish aerial mycelium on PDA medium. The hyaline, unicellular, and ovoid conidia (5.8 to 11.2 × 4.2 to 7.1 μm) were borne on the long, branched conidiophores, resembling a typical grape-like cluster. Based on these morphological characteristics, the causal pathogen was preliminarily identified as Botrytis cinerea. To confirm the identity of this fungus, the internal transcribed spacer region (ITS) of the ribosomal DNA was amplified and sequenced using ITS1 and ITS4 primers (White et al. 1990). BLASTn analysis of the resulting ITS sequence (GenBank accession no. MK370589) showed 100% similarity with those of B. cinerea (MH763647, KX443701, and KT266231). Furthermore, the glyceraldehyde 3-phosphate dehydrogenase (G3PDH) gene of this fungus was also sequenced (Staats et al. 2005), and the amplified G3PDH gene (MK372882) exhibited 100% identity with sequences of B. cinerea in GenBank (MH732864, MH974746, and MK051165). Leaves of Lanzhou lily plants were soaked in conidial suspension (106 spores/ml in potato dextrose broth [PDB]) for 1 min to fulfill the pathogenicity test. In the control treatment, the seedlings only received PDB solution. The plants were incubated in a moist climate chamber at 22°C, receiving 16 h of light per day, and irrigated with sterile water. Leaf blight lesions were observed on the plants inoculated with conidial suspension within 10 days, and B. cinerea was reisolated from the infected tissues. No symptoms were observed on control plants inoculated with PDB only. The pathogen was reported to cause leaf blight on ornamental lily in China (Cao et al. 2018); however, to our knowledge, this is the first report of B. cinerea causing leaf blight disease on edible Lanzhou lily in China and globally.The author(s) declare no conflict of interest.References:Cao, X., et al. 2018.
