Abstract:The decapeptide gonadotropin-releasing hormone (GnRH) is a kind of regulative neuropeptide synthesized by GnRH-producing cell bodies in hypothalamus and is an important information molecule among nervous, immune and neuroendocrine systems. GnRH plays a pivotal role in the regulation of reproduction. In recent years, GnRH-analogues have been one of the most widely applied hormonal medicine. The research progresses of GnRH were reviewed, including structure, localization and distribution of GnRH and its receptor. A series of evidences on GnRH regulating reproduction activity at different levels such as pituitary and gonad, factors affecting secretion of GnRH, as welt as prospects of studies on GnRH are also introduced.

Abstract:The physiology of reproduction is very complex and is regulated by multiple factors, including a number of hypothalamic neuropeptides. In last few decades, various neuropeptides have been discovered to be involved in stimulation or inhibition of reproduction. In 2000, Tsutsui and colleagues uncovered gonadotropin-inhibitory hormone (GnIH), a neuropeptide generating inhibitory drive to the reproductive axis, in the brain of Coturnix quail. Afterward, GnIH orthologs were discovered in other vertebrates from fish to mammals including human. In these vertebrates, all the discovered GnIH and its ortholgs have LPXRFamide (X=L or Q) sequence at C-terminus. GnIH orthologs of mammals and primates are also termed as RFamide-related peptide (RFRP)-1 and -3 that too have an LPXRFamide (X=L or Q) motif at their C-terminus. GnIH and its orthologs form a member of the RFamide peptide family. GnIH signals via its canonical G protein coupled receptor 147 (GPR147). Both GnIH and GPR147 are expressed in hypothalamus and other brain regions. Besides actions through the hypothalamic GnRH and kisspeptinergic neurons, GnIH-GPR147 signaling exerts inhibitory effect on the reproductive axis via pituitary gonadotropes and directly at gonadal level. Various factors including availability and quality of food, photoperiod, temperature, social interaction, various stresses and some diseases modulate GnIH-GPR147 signaling. In this review, we have discussed expression and actions of GnIH and its orthologs in vertebrates. Special emphasis is given on the role of GnIH-GPR147 signaling pathway in the regulation of reproduction. We have also reviewed and discussed currently available literature on the participation of GnIH-GPR147 signaling pathway in the stress modulation of reproduction.

Abstract:Gonadotrophin-releasing hormone(GnRH),a neuropeptide with 10 amino-acid,plays a critical role in the regulation of reproduction.There are 2 genome types of GnRH in human,GnRH-I and GnRH-II,and 2 types of receptor,GnRHR-I and GnRHR-II.They regulate the reproduction and maturity through the hypothalamicpituitary-gonadal axis,and also have the effect on some extrapituitary compartments by autocrine and/or paracrine,including the breast,placenta,ovary and endometrium.GnRH may be a valuable effective method of treatment and prevention of deterioration for breast cancer.GnRH analogues have been used to treat many hormones related diseases,especially uterine diseases,such as endometrial hyperplasia,endometrial cancer and endometriosis(EMs),as well as reduce the risk of cervical cancer.Placenta can generate GnRH or GnRH-like peptides.Studies of placental tissue in vitro indicate that GnRH can significantly increase the release of human chorionic gonadotropin(hCG) from placenta tissue.Local GnRHs not only plays an important role in maintaining ovarian function,but also is involved in normal and abnormal reproductive endocrine regulation in ovary.
Gonadotropin-releasing hormone: molecular and cell biology, physiology, and clinical applications

Abstract:The observations in recent years that gonadotropin-releasing hormone ( pyroGlu1 -His2-Trp3- Ser4 -Tyr5- Gly6 -Leu7-Arg8-Pro9- G ly10NH2 [GnRH]) has considerable clinical efficacy as a contraceptive, an ovulation-inducing agent, and a drug that is useful in the therapy of a number of diseases are owed in no small part to advances that have been made in the basic molecular biology and physiology of this peptide. It is reasonable then that this symposium should be broken down into three separate yet integrated areas: 1) the molecular and cellular biology of GnRH, 2) the physiology of GnRH, and 3) the clinical efficacy of this compound. A number of recent advances have helped the progress of work in all three of these areas. The observation that substitution of a D-amino acid in the sixth amino acid position leads to considerable metabolic stability and reduced degradation of releasing hormone analogs has been especially useful. In addition, the observation that the further derivatization of such substituted compounds by deletion of the 10th amino acid and addition of an ethylamide group to Pro9 has resulted in compounds with a markedly increased ability to bind to the receptor. These compounds have been useful clinically and have allowed development of radioligand assays to be used in animal studies and in vitro. The availability of a large number of agonists and antagonists has allowed formulation of molecular models for GnRH and resulted in development of clinically useful compounds. In a short paper of this nature we cannot hope to provide an encyclopedic review; rather we hope to provide an overview and offer references to the literature for those who wish a more in-depth treatment.

Abstract:The hypothalamic decapeptide gonadotropin releasing hormone (GnRH) plays an important role in the control of human reproduction. The physiological function of GnRH is mediated by GnRH receptor(GnRHR). GnRH binds to its receptor in high specificity. The distribution of GnRH and GnRHR is not only in the hypothalamic pituitary gona- dal axis but also in several peripheral organs such as placenta, gonads and mammary glands. Recent studies show that GnRH and GnRHR are also expressed in a number of human malignant tumors, including cancers of the breast, ovary and endometrium. Antiproliferative effect of GnRH and its analogs in cell lines derived from these can- cers have been observed by various investigators. The classical GnRH receptor signal transduction mechanisms, operated in the pituitary, are not involved in the mediation of antiproliferation of GnRH analogs in cancer cells. The GnRHR interacted on the mitogenic signal transduction of growth factor receptors and related oncogene products is associated with tyrosine kinase activity, via activation of a phosphotyrosine phosphatase, resulting in downregulation of cancer cell proliferation.

Abstract:RF-amide related peptide (RFRP) is the orthologue of gonadotropin-inhibitory hormone (GnIH) in mammals. The bodies of RFRP cell are located in the dorsomedial nucleus of the hypothalamus (DMH) and the fibers project to preoptic area (POA) and median eminence of the hypothalamus. Its receptor mainly distributes in hypothalamus. RFRP fibers project to GnRH cells to regulate mammalian reproduction axis. This paper reviews the progress of current researches on RFRP in regulation of animal behaviors, including reproduction, food intake, anxiety and stress response.

Abstract:Gonadotropin-releasing hormone (GnRH), a conservative neuro-decapeptide family, plays a crucial role in regulating the gonad development and controlling the final sexual maturation in vertebrates. Since the first GnRH, namely mGnRH, was identified, 16 kinds of GnRH variants have been characterized from protochordate and vertebrate brain tissues to this day. cGnRH-Ⅱ peptide exist in all the brain tissue of detected organisms, and it is highly conservative during the vertebrate evolution. However, the main function peptide is the GnRH, which is predominately expressed in hypothalamus and telencephalon. Corresponding to the multiplicity of GnRH peptide,different subtypes of GnRH-R is also presented in gonadtropnn cell membrane of some vertebrates. So, it is proposed that the GnRH-R molecular have three kinds of subtypes which take form by homologous evolution. Thedeletion or mutant of GnRH gene or GnRH-R gene will lead to the abnormity or pathological changes of organism individual in propagate physiology. According to the above theory, transgenic fish are produced using antiense transgenic technique, and sterile transgenic fish are finally gained by inhibiting the expression of the sGnRH gene in brain level. Transgenic fish generated by antisense transgenic technology is safe in genetic and ecological aspects, which will supply a new technique scheme for the industrialization of transgenic fish.

Abstract:C-type natriuretic peptide(CNP),a member of the natriuretic peptide family of structurally related peptides,is a autocrine / paracrine growth factor with important roles in cardiovascular homeostasis,regulation of cell proliferation and skeletal development;moreover,CNP also plays a key role in modulating animal reproductive processes.Notably,recent studies have revealed that CNP acts as a regulator to inhibit meiotic resumption in mouse oocytes in vivo;additionally,CNP can enhances superovulation response by the stimulation of follicular development prior to gonadotropins treatment.In light of the findings above,CNP may be of special importance in the promotion of oocyte development in vitro as one meiotic inhibitor,as well as the improvement of superovulation efficiency in domestic animals.Hence,this paper reviewed the structure of CNP,its role as one of functional molecules that modulate reproductive processes in animals and its potencial value.

Abstract:Mammalian reproduction is mainly driven and regulated by the hypothalamic-pituitary-gonadal (HPG) axis. Follicle-stimulating hormone (FSH), which is synthesized and secreted by the anterior pituitary gland, is a key regulator that ultimately affects animal fertility. As a dimeric glycoprotein hormone, the biological specificity of FSH is mainly determined by the β subunit. As research techniques are being continuously innovated, studies are exploring the underlying molecular mechanism regulating the secretion of mammalian FSH. This article will review the current knowledge on the molecular mechanisms and signaling pathways systematically regulating FSH synthesis and will present the latest hypothesis about the nuclear cross-talk among the various endocrine-induced pathways for transcriptional regulation of the FSH β subunit. This article will provide novel ideas and potential targets for the improved use of FSH in livestock breeding and therapeutic development.

Abstract:Growth hormone-releasing hormone (GHRH) has classically been considered a regulatory neuropeptide of the hypothalamic-pituitary system, which mediates its anabolic effects through hepatic GH/IGF-I axis. However, during the last decades it has been demonstrated that this key regulatory hormone may be produced in numerous peripheral tissues outside the central nervous system, participating in fundamental physiological functions through a complex balance between its purely endocrine action, and the recently local (autocrine/paracrine) discovered role. Among peripheral sites, its presence in the male and female reproductive systems stands out. In this review, we will first explore the role of the GHRH/GHRH-R hormone axis as a central player in the gonadal function; then, we will discuss available information regarding the presence of GHRH/GHRH-R and the potential physiological roles in reproductive systems of various species; and finally, we will address how reproductive system-related disorders—such as infertility problems, endometriosis, or tumor pathologies (including prostate, or ovarian cancer)—could benefit from hormonal interventions related to the manipulation of the GHRH axis.

Abstract:Luteinizing hormone (LH) and human chorionic gonadotropin (CG), like follicle-stimulating hormone, are the most important regulators of the reproductive system. They exert their effect on the cell through the LH/CG receptor (LHCGR), which belongs to the family of G protein-coupled receptors. Binding to gonadotropin induces the interaction of LHCGR with various types of heterotrimeric G proteins (Gs, Gq/11, Gi) and β-arrestins, which leads to stimulation (Gs) or inhibition (Gi) of cyclic adenosine monophosphate-dependent cascades, activation of the phospholipase pathway (Gq/11), and also to the formation of signalosomes that mediate the stimulation of mitogen-activated protein kinases (β-arrestins). The efficiency and selectivity of activation of intracellular cascades by different gonadotropins varies, which is due to differences in their interaction with the ligand-binding site of LHCGR. Gonadotropin signaling largely depends on the status of N- and O-glycosylation of LH and CG, on the formation of homo- and heterodimeric receptor complexes, on the cell-specific microenvironment of LHCGR and the presence of autoantibodies to it, and allosteric mechanisms are important in the implementation of these influences, which is due to the multiplicity of allosteric sites in different loci of the LHCGR. The development of low-molecular-weight allosteric regulators of LHCGR with different profiles of pharmacological activity, which can be used in medicine for the correction of reproductive disorders and in assisted reproductive technologies, is promising. These and other issues regarding the hormonal and allosteric regulation of LHCGR are summarized and discussed in this review.

Abstract:Follicle stimulating hormone (FSH) is an essential glycoprotein hormone for human reproduction, which functions are mediated by a G protein-coupled receptor, FSHR. Aberrant FSH-FSHR signaling causes infertility and ovarian hyperstimulation syndrome. Here we report cryo-EM structures of FSHR in both inactive and active states, with the active structure bound to FSH and an allosteric agonist compound 21f. The structures of FSHR are similar to other glycoprotein hormone receptors, highlighting a conserved activation mechanism of hormone-induced receptor activation. Compound 21f formed extensive interactions with the TMD to directly activate FSHR. Importantly, the unique residue H6157.42 in FSHR plays an essential role in determining FSHR selectivity for various allosteric agonists. Together, our structures provide a molecular basis of FSH and small allosteric agonist-mediated FSHR activation, which could inspire the design of FSHR-targeted drugs for the treatment of infertility and controlled ovarian stimulation for in vitro fertilization. ### Competing Interest Statement The authors have declared no competing interest.
Discovery of new receptors regulating luteinizing hormone and follicle-stimulating hormone secretion by bovine gonadotrophs to explore a new paradigm for mechanisms regulating reproduction

Abstract:Previous studies in the 1960s and 1970s have reported that both gonadotropin-releasing hormone (GnRH) and estradiol-activated nuclear estrogen receptors regulate gonadotropin secretion in women. However, I had previously reported that gonadotroph function is regulated by complex crosstalk between several membrane receptors. RNA-seq had previously revealed 259 different receptor genes expressed in the anterior pituitary of heifers. However, the biological roles of most of these receptors remain unknown. I identified four new receptors of interest: G protein-coupled receptor 30 (GPR30), anti-Mullerian hormone (AMH) receptor type 2 (AMHR2), and G protein-coupled receptors 61 and 153 (GPR61 and GPR153). GPR30 rapidly (within a few minutes) mediates picomolar, but not nanomolar, levels of estradiol to suppress GnRH-induced luteinizing hormone (LH) secretion from bovine gonadotrophs, without decreasing mRNA expressions of the LHα, LHβ, or follicle-stimulating hormone (FSH) β subunits. GPR30 is activated by other endogenous estrogens, estrone and estriol. Moreover, GPR30 activation by zearalenone, a nonsteroidal mycoestrogen, suppresses LH secretion. AMHR2, activated by AMH, stimulates LH and FSH secretion, thus regulating gonadotrophs, where other TGF-β family members, including inhibin and activin, potentially affect FSH secretion. I also show that GPR61, activated by its ligand (recently discovered) significantly alters LH and FSH secretion. GPR61, GPR153, and AMHR2 co-localize with the GnRH receptor in unevenly dispersed areas of the bovine gonadotroph cell surface, probably lipid rafts. The findings summarized in this review reveal a new paradigm regarding the mechanisms regulating reproduction via novel receptors expressed on bovine gonadotrophs.

Abstract:Follicle stimulating hormone( FSH),a glycoprotein hormone,plays an important role in human assisted reproduction in clinic as one of the main drugs in the treatment of man and woman infertility. However,its relatively short half-life requires frequent administration,which makes poor patient compliance. In recent years,the development of long-acting follicle stimulating hormones,which have a longer half-life and higher in vivo activity than wild-type FSH,has become a hot research. Currently the methods of designing a long-acting FSH include increasing glycosylation sites and making Fc fusion proteins. In this paper,we reviewed the long-acting follicle stimulating hormone drugs and their main biological properties.
