Abstract:Peanut protein is easily digested and absorbed by the human body, and peanut tofu does not contain flatulence factors and beany flour. However, at present, there is no industrial preparation process of peanut tofu, whereas the quality of tofu prepared by different peanut varieties is quite different. This study established an industrial feasible production process of peanut tofu and optimized the key process that regulates its quality. Compared with the existing method, the production time is reduced by 53.80%, therefore the daily production output is increased by 183.33%. The chemical properties of 26 peanut varieties and the quality characteristics of tofu prepared from these 26 varieties were determined. The peanut varieties were classified based on the quality characteristics of tofu using the hierarchical cluster analysis (HCA) method, out of which 7 varieties were screened out which were suitable for preparing peanut tofu. An evaluation standard was founded based on peanut tofu qualities. Six chemical trait indexes were correlated with peanut tofu qualities (P<0.05). A logistic regressive model was developed to predict suitable peanut varieties and this prediction model was verified. This study may help broaden the peanut protein utilization, and provide guidance for breeding experts to select certain varieties for product specific cultivation of peanut.

Processing technology optimization for tofu curded by fermented yellow whey using response surface methodology

Abstract:<p>The technological applications utilized for tofu processing are diverse and complex, resulting in different yields and quality characteristics of tofu. The current study investigated the gel-forming principle of soybean protein coagulated using fermented yellow whey (FYW) to produce tofu. The effects of several processing parameters (soybean-to-water ratio, boiling temperature, boiling time, and FYW content) on the yield and protein content of tofu produced by the boiling-to-filtering method (BFM) were studied and optimized using response surface methodology. Results indicated significant differences in yield and protein content of tofu using different processing parameters, with FYW content being the most significant (<i>p</i> < .05). Optimum processing parameters of the BFM were found to be: soybean-to-water ratio of 1:5 (kg:kg), boiling time 6.1 min, boiling temperature 105°C, and FYW content of 26%. Under optimum conditions, tofu's yield and protein content were 235.17 g/100 g and 10.60%, respectively, and these were 47.93 g/100 g and 4.16% higher than those before optimization. This study provides practical technical support and a theoretical basis for the standardized industrial production of high-yield and high-protein tofu.</p>

Abstract:Egg make-ups and milk are added into soymilk to increase the sulfur amino acids and the soybean protein's absorbability of tofu, thus different kinds of tofu are made. The nutritional evaluation results show that by adding egg or milk, the protein content of tofu increases 20%～30% and the nutritional value of new tofu products is improved.

Abstract:A combined process with steam flash-explosion,alkaline-extraction and acid-precipitation was used to extract protein from high-temperature peanut meal.Through single factor and orthogonal test,the optimum conditions were figured out.Firstly,the high-temperature peanut meal was soaked in 0.3% sulfuric acid solution at 60℃ for 2h,then drained it and washed with water to remove the acid solution remained in the surface of the meal.The pretreated meal was treated with steam flash-explosion at 1.6MPa for 5min.The protein in the steam flash-exploded meal was extracted by alkaline-extraction and acid-precipitation process.The optimal condition for alkaline-extraction was solid to water ratio:1∶12(g/mL),pH9.5,60℃ and 2h.Under these conditions,the protein extraction rate reached 52.6%,which was 10.8% higher than conventional alkaline extraction and acid-precipitation process.The water holding capacity,emulsifying activity index,foaming capacity and foaming stability of the protein extracted using this combined process were significantly improved,which increased by 67.1%,141.0%,131.3% and 107.4% respectively.R esults showed that the combined process with steam flash-explosion,alkaline-extraction and acid-precipitation was appropriate for extracting protein from high temperature peanut meal,which could increase the protein extraction rate and also improve the functional properties.

Abstract:Summary In this study, sugar beet pectin and soy protein were used to prepare the double‐network ( DN ) modified‐tofu by sequential laccase and microbial transglutaminase treatments. The mechanical properties of modified‐tofu were analysed by uniaxial compression test and texture profile analysis. The DN modified‐tofu exhibited greater mechanical toughness than the corresponding single‐network modified‐tofu, while the ratio of soy glycinin to soy protein isolate reached and exceeded 50%. The enhancement of mechanical toughness may be mainly ascribed to the hierarchical microstructure which is observed in DN modified‐tofu by scanning electron microscopy. DN modified‐tofu provided a distinctive mechanical and sensory tough texture as compared to other tofu samples. In addition, the oral processing measurements indicated that DN modified‐tofu required higher jaw‐closing muscle activities, a greater number of chews and longer chewing duration to be processed. Furthermore, the chewing pattern of DN modified‐tofu was different from other tofu samples.

Abstract:Low bitter peanut polypeptide was prepared by using endoenzyme and exopeptidase treating high temperature peanut meal.The optimum hydrolyzing parameters were established by the single factor and orthogonal test.The optimal processing conditions were as follows:pH 7.0,temperature 50℃,ratio of protein to water 1:25(m/V),As1398 dosage 4 500 U/g,Flavourzyme 500MG dosage 3% and reaction time 1.5 h.Under these conditions,the recovering rate of nitro was 82.33%,and the bitterness of hydrolysate was 2.The peanut polypeptide powder had a higher NSI,and its amino acids kinds were complete.

Abstract:Objective: To study the processing technology and formula of peanut–tea beverage, and further provide reference for the plant protein beverage. Methods: The peanut was baked in the far-infrared oven, soaked with soft water containing 5% NaHCO3, rinsed and then cooked for 30 min, milled at 0.05 mm spacing grinding, filtered and finished peanut slurry. Tea leaves were extracted twice by water of 90°C. The two ingredients were combined with other supplement, refined by colloid mill, homogenized twice, followed by bottling, sealed, sterilization, cooled and other processing. In this study, the formula, stabilizers and additives by orthogonal experiments were investigated. Results: The peanut slurry and sugar were found to be major factors which affected the beverage’s quality. The formula was optimized when the content of peanut slurry, tea and sucrose reached 75 mL, 25 mL and 4 g especially in per 100 mL peanut-tea beverage. As single stabilizer, CMC-Na showed the most significant effect, and storage temperature was found to be better at room temperature than low temperature. Compared with single stabilizer, compound stabilizer was better. The optimal formula was CMC-Na 0.4 g/L, seaweed sodium 0.2 g/L, gelatin 0.8 g/L. Conclusion: The peanut-tea beverage, with unique flavor and rich nutrients, was a novel type of health plant beverage which was suitable for a large variety of people, and had a promising market.

Abstract:Based on the traditional production process of dried tofu,production process of high elastic dried tofu was studied. Using fresh soybean as raw materials,on the basis of single factor experiment,technological parameters of high elastic dried tofu were determined by response surface methodology to optimize production process as follows :micro fluidic homogeneous pressure 120 MPa,steam cooking pressure 0.25 MPa,steam cooking temperature 110℃. Under the optimal process conditions,a kind of high elastic dried tofu with high quality and good sensory organ could be achieved.

Abstract:Peanut polypeptide liquid (＜ 3 000 u) that prepared by enzymatic hydrolysis of peanut protein power from cold-pressed peanut meal (CPPM) was raw metarial and the ferrous chloride was metal chelator.The optimal chelate technological parameters was obtained after the the optimized analysis to the polypeptide-ferrous chelator by using Box-Behnken response surface optimization technique based on single factor experiment,ie.,polypeptide-ferrous mass ratio was 4.31∶1 (g/g),chelating temperature was 25.4 ℃,chelating time was 28.5 min and chelating pH was 7.5.Under the optimization condition,the polypeptide-ferrous chelation rate was (85.68-± 1.27) ％ (n =3),which was well agreement with the model predicted value (89.653 1％),and the deviation between the confirmative result and the predicted result was 4.64％.Through ultraviolet spectroscopy (UV) and infrared spectrometer(FT-IR) analysis,peanut meal albumen polypeptide-ferrous chelate was found,the Fe2+ and NH2 + in the polypeptide and COO-form covalence coordinate bond and form stable conjugated structure,which was a new-type organic metal-chelator.

Abstract:Different proteases were used to hydrolyze soybean meal protein,according to protein dissolution ratio and free amino nitrogen content,alkaline protease was selected as the most appropriate enzyme.The hydrolysis time,enzyme/substrate ratio and substrate concentration were analyzed on the eftects of soybean meal protein hydrolysis.On the basis of single factor experiment,Design-Expert7.0 response surface analysis was then used to optimize the levels of the three factors.The optimum hydrolyzing time,enzyme/substrate ratio and substrate concentration were 4h,6900u/g and 6%,respectively for alkaline protease,and the protein dissolution ratio reached 74.38%.The reaction time and enzyme/substrate ratio of Flavorzyme hydrolysis to prepare low-bitter peptide through the fluorescent probes analysis were 6h and 80u/g respectively.

Abstract:The soybean variety Zhongnong13 was used to study the effect of soaking,heating and coagulating conditions on tofu-gel strength and water-holding capacity.The results suggested that the optimal condition can be obtained as follows: 20℃ soaking 17h,100℃ heating 10min,0.3% GDL,90℃ coagulating 120min.Among these conditions,GDL made the most important effect on tofu-gel strength and water-holding capacity.

Abstract:Three enzymes, GA (Genencor Alkaline Protease), GN (Genencor Neutral Protease) and Protamex were used to prepare protein hydrolysates from defatted peanut meal (DPM). The physicochemical properties of defatted peanut meal hydrolysate (DPMH) and the changes of flavor were determined. DPMHs had lower isoelectric points, zeta potential values and better solubility. The DPMH of GN showed the best emulsifying activity and the emulsion stability. Volatile organic compounds (VOCs) changed significantly with the most drastic changes at 4-6 h. About 50 VOCs were labeled using GC-IMS. The fingerprint of smell change was established, and the enzymolysis reaction could be divided into different stages according to the change of flavor. DPMHs of GA and Protamex showed stronger astringency and bitterness, while stronger umami was shown in DPMH of GN. This research provides basic data support for enzyme selection, sensory and reaction stage control to obtain enzymatic polypeptides with improved properties from DPM.
Preparation of germinated soybean milk with household soymilk machine

Abstract:The objectives of this study were to optimize pulverizing and boiling process for germinated soybean milk.Pulverizing temperature and boiling time were explored through single-factor experiments and the optimal temperature was figured out as 70℃and boiling time as 4min.With optimized process,content ofγ-aminobutyric acid,total freeamino acids,essential freeamino acids in the germinated soybean based milk were 1.36mg/100 g,0.455g/100 g,and 0.219g/100 g,respectively.Compared with dry soybean milk,the above contents were 3.68,1.34,8.87 folds increased respectively.Despite that the trypsin inhibitor activity(TIA)was 1.27 times that of dry soybean based milk,the nutritional value of germinated soybean milk has been remarkably improved.
Effects of Pretreatment on Stability of Peanut Oil Bodies and Functional Characteristics of Proteins Extracted by Aqueous Enzymatic Method

Abstract:Effects of dry and wet grind on peanut oil and protein yield, oil bodies (OBs) stability, fatty acid composition, protein composition and functional characteristics were systematically analyzed. Results showed that peanut oil and protein yields reached highest at dry grind 90 s (92.56% and 83.05%, respectively), while peanut oil and protein yields were 94.58% and 85.36%, respectively, at wet grind 120 s. Peanut oil and protein yields by wet grind was 2.18% and 2.78% higher than that of dry grind, respectively. Surface protein concentration (Г) and absolute value of zeta potential of OBs extracted by wet grind (WOBs) were 11.53 mg/m 2 and 18.51 mV, respectively, which were higher than OBs extracted by dry grind (DOBs), indicating stability of WOBs was higher than DOBs. Relative contents of oleic acid and linoleic acid in peanut oil, essential and hydrophobic amino acids in protein extracted by wet grind were higher than dry grind. There was little difference in protein composition between wet and dry grind, but thermal denaturation degree of protein obtained by wet grind was lower than dry grind. Solubility, oil retention, emulsion stability, foaming and foam stability of protein obtained by wet grind were better than dry grind. Results from this study provided theoretical basis for grind pretreatment selection of aqueous enzymatic method.

Abstract:Tofu has a long history of use and is rich in high-quality plant protein; however, its production process is relatively complicated. The tofu production process includes soybean pretreatment, soaking, grinding, boiling, pulping, pressing, and packing. Every step in this process has an impact on the soy protein and, ultimately, affects the quality of the tofu. Furthermore, soy protein gel is the basis for the formation of soy curd. This review summarizes the series of changes in the composition and structure of soy protein that occur during the processing of tofu (specifically, during the pressing, preservation, and packaging steps) and the effects of soybean varieties, storage conditions, soybean milk pretreatment, and coagulant types on the structure of soybean protein and the quality of tofu. Finally, we highlight the advantages and limitations of current research and provide directions for future research in tofu production. This review is aimed at providing a reference for research into and improvement of the production of tofu.

Abstract:The use of defatted soybean flour (DSF) in food as a source of dietary fiber has been limited due to its rough texture and bitter taste. Our previous work indicates that superfine DSF prepared by jet milling could overcome these problems, as it positively affected physical and sensory properties. Therefore, differently sized DSFs were incorporated in tofu, and their impacts on physical and sensory properties were investigated in this study. Coarse DSF (Dv50 = 341.0 μm), fine DSF (Dv50 = 105.3 μm), and superfine DSF (Dv50 = 5.1 μm) were prepared by conventional sifting and jet milling. Tofu was made with a 5% addition of differently sized DSFs and without DSF (control tofu). The quality of tofu was evaluated by scanning electron microscopy, color measurement, texture profile analysis, and quantitative descriptive analysis. The tofu made with coarse and fine DSF showed negative changes in its physical and organoleptic qualities, such as reduced yields, a less pure color, a harder texture, and a rougher mouthfeel. However, the tofu made with superfine DSF showed only minimal changes in its qualities compared to the control. Therefore, superfine DSF is a promising fiber supplement that does not change the physical and sensory properties in the making of high-quality tofu.

Abstract:ABSTRACTPeanut protein concentrate (PPC) was extracted from commercial defatted peanut flour (DPF) after protease pretreatment. The effect of proteolysis by Alcalase 2.4 L, papain and Protamex at various degrees of hydrolysis (DHs; 1–8%) on the yield and functional properties of PPC were investigated. Proteolysis remarkably increased the nitrogen solubility of DPF in acidic and basic conditions. Enzymatic degradation of denatured and insoluble proteins in DPF was mainly responsible for the increase of nitrogen solubility index. Compared with the control, the yield of PPC was significantly increased by enzymatic pretreatment. In terms of the functional properties, PPCs prepared by papain and Protamex at 4% or 8% DH had high emulsifying activity index. For all three enzymatic treatments, foaming capacity and foaming stability were significantly improved at various DH (P < 0.05). This study demonstrated that protease pretreatment was a highly effective way to extract PPC with good functional properties from DPF.PRACTICAL APPLICATIONSPeanut is an important oilseed crop and a well‐accepted food. After oil production through thermal treatment, the defatted peanut flour is the main by‐product, which possesses large amount of proteins. However, due to the low extraction yield and poor functional properties of these proteins, they are not well utilised in industry till now. In this work, protease was used to pretreat defatted peanut flour. The results indicated that this pretreatment could improve the yield and functional properties of peanut proteins. Functional properties of food proteins are important in food processing and food product formulation. Peanut protein with good emulsifying and foaming properties would have a potential application in some food system, such as salad dressing, sausages, bologna, soups, confectionery, frozen desserts and cakes and so on. Therefore, this work is helpful for extensive utilisation of peanut proteins.
Optimization of Process Parameters of Probiotics-Fermented Soybean Meal and Effects of Coordination Action Between Compound Probiotics and Enzyme on Fermentation Quality

Abstract:This experiment was conducted to optimize process parameters of fermented soybean meal(FSBM) fermented by compound probiotics(Saccharomyces cerevisae∶ Aspergillus oryzae∶ Bacillus subtilis=5∶ 1∶ 2) and investigate the effects of protease supplementation on fermentation quality.The optimal fermentation parameters were obtained through measuring the indexes under imitated factory production scale condition which included temperature,pH and the contents of incubation moisture,crude protein,true protein and total volatile basic nitrogen at 0,24,48 and 72 h during fermentation process at 4 levels(1∶ 0.3,1∶ 0.4,1∶ 0.5 and 1∶ 0.6) of the ratio of material to water.Then the effects of protease supplementation on fermentation quality were compared between control group and experimental group.The results showed as follows: 1) in different ratios of material to water,the temperatures showed a downward trend after the first rise,while pH fell gradually and initial moisture content rose gradually during 72 h fermentation process.Crude protein content(47.29%) was significantly improved in the group with 1∶ 0.6 of the ratio of material to water at 48 h fermentation.True protein content(43.34%) in the group with 1∶ 0.4 of the ratio of material to water at 48 h fermentation was the highest value.Total volatile basic nitrogen content(38.10×10-2 mg/g) reached the peak value in the group with 1∶ 0.6 of the ratio of material to water at 48 h fermentation.2) The contents of true protein and dry matter were decreased by 2.59 and 4.11 percentage points(P0.05),while free amino acids content was increased by 0.36 percentage points through the supplementation of protease(P0.05).Protease supplementation could also significantly eliminate macromolecule protein of soybean meal(P0.05).The results indicate that compound probiotics can ferment FSBM at a low level ratio of material to water and protease supplementation can improve the quality of fermentation.It is concluded that the optimal fermentation parameters were formulated to contain 0.5%(w/w) compound probiotics(Saccharomyces cerevisae∶ Aspergillus oryzae∶ Bacillus subtilis=5∶ 1∶ 2),0.01%(w/w) protease supplementation,1∶ 0.4 of the ratio of material to water and fermented for 48 h.Protease supplementation can improve the quality of available nitrogen in FSBM.
Optimization of defatting technics of cold -pressed peanut meal by quadratic rotatable central composite response surface methodology

Abstract:To take advantage of protein resource from cold -pressed peanut meal,and to reduce the im-pact of residual oil on protein deep processing,anhydrous ethanol was chosen as defatting solvent after comprehensive consideration of solvent safety and the influences of anhydrous ethanol on protein loss rate and defatting rate.The influence rules of defatting temperature(X1 ),defatting time(X2 )and solid -liquid ratio(X3 )were investigated by quadratic rotatable central composite response surface methodology based on single factor experiment,and a second order polynomial nonlinear regression equation and numer-ical model was established.Optimum parameters were determined as defatting temperature 59.5 ℃,defat-ting time 108 min and solid -liquid ratio 1∶19.Under the optimal condition,defatting rate was 92.56%, which was close to the model predicted value 93.07% with the deviation of 0.55%.The results indicated that the combination of single factor experiment and quadratic rotatable central composite response surface methodology can be used in optimization of defatting process of cold -pressed peanut meal.

Abstract:The combined pretreatment of heat-denatured soybean meal using steam flash-explosion (SFE) with sulfuric-acid soaking was investigated to prepare protein from soybean meal. When soybean meal was pre-treated by SFE at 1.8 MPa, 2.2 MPa for 8 min and at 2.0 MPa for 8 min and 10 min, combined with 0.9% sulfuric-acid soaking, the extraction yield of protein increased to 67.72%, 70.54%, 69.47% and 71.21% respectively, compared to untreated soybean meal. Scanning electron micrograph of pretreated samples showed the structural disruption of soybean meal. After pretreatment, the protein yield was improved, while protein content of soy protein isolate (SPI) decreased slightly. The functional properties of SPI from pretreated soybean meal were all improved compared to untreated soybean meal and the relationship between functional properties and the changes of surface hydrophobicity of SPI was discussed. The emulsification properties and fat-binding capacity of pretreated SPI were superior to those of SPI prepared from white flakes. (C) 2013 The Authors. Published by Elsevier Ltd.
