Alterations in arterial responsiveness of abdominal aorta and femoral artery in hyperthyroid rats

Abstract:AIM To investigate the alterations in arterial contractile and dilatory responsiveness of abdominal aorta and femoral artery in hyperthyroid rats.METHODS To induce hyperthyroidism,Levothyroxine(L-T4,Euthyrox) was used to feed rats for 25 days by intragastric administration.Twenty-four male Sprague-Dawley rats weighing 350-400 g were randomly divided into three groups(n=8/group): control group,hyperthyroidism-1 group [(low-dose group,L-T4,0.3 mg/(kg·d)] and hyperthyroidism-2 group [(high-dose group,L-T4,0.6 mg/(kg·d)].Arterial ring preparations from both abdominal aorta and femoral artery were used to determine their contractile and dilatory responses to various agents.RESULTS Compared with those in control group,contractile responses in both abdominal aortic and femoral arterial rings evoked by both KCl(1-9)×10~(-2) mol/L and phenylephrine(PE,10~(-9) mol/L-10~(-4) mol/L) significantly decreased(P<0.05)in hyperthyroid rats.But dilatory responses of 0.5 μmol/L PE preconstricted rings induced by acetylcholine(10~(-9) mol/L-10~(-4) mol/L) and sodium nitroprusside(10~(-9) mol/L-10~(-4) mol/L) significantly increased(P<0.05) in both abdominal aortic and femoral arterial rings of hyperthyroid rats.The sensitivity of both abdominal aorta and femoral artery to KCl,ACh and SNP were significantly enhanced by hyperthyroid state,but EC50 of both abdominal aortic and femoral arterial rings to PE increased in hyperthyroid rats.CONCLUSION Contractile responsiveness is decreased and dilatory responsiveness is enhanced in abdominal aorta and femoral artery of hyperthyroid rats.
