Effect of MSCs' transplantation on oxidative stress after traumatic brain injury in adult rats

Abstract:Objective To investigated the effect of bone marrow stromal cells (MSCs) administered by the way of internal carotid artery on functional outcome after traumatic brain injury in adult rats.Besides,we observed the influence of anti-oxidation after transplantation.Methods MSCs were harvested from rat bone marrow.A controlled cortical impact was delivered to 38 adult rats to induce traumatic brain injury,and 24 hours after injury,MSCs were injected into the internal carotid artery of the rats (n = 21).Every rat was administered 2×106MSCs.16 rats received PBS in the same way.Neurological function was evaluated according to the modified neurological severity score.In addition,not only the content of SOD、OH-in the experimental group rats brain tissue have been detected,but also the contrast group rats.Results Treatment with 2×106MSCs significantly improved the rats' functional outcomes(P<0.05).After transplantation,higher activity of SOD be detected in experimental group rats brain tissue,on the contrast,content of OH-were lower.Conclusion These data suggest that MSCs may be a potential therapy for traumatic brain injuries in rat.
