In May 2022, symptoms of soft rot on A. fistulosum grown for food production were observed in a greenhouse at the Beijing Academy of Agricultural and Forestry Sciences (39°57′N, 116°18′E). Approximately 10% of the plants in the greenhouse were infected. Diseased plants showed lodging and internal bulb rot accompanied with an acrid odor. Plants died within 2 weeks after appearance of symptoms. Symptomatic bulbs were surface disinfected in 75% ethanol for 30 s, rinsed three times in sterilize water, and ground in sterile water, and a portion of the homogenate was placed on Luria broth (LB) solid medium. Plates were incubated for 2 days at 25°C. Single colonies were reinoculated on LB solid medium to obtain pure cultures. Finally, two types of bacterial colonies were obtained and named ZM1 and ZM2. The colony of ZM1 was light brown and about 18 mm in diameter, while the colony of ZM2 was milky white and about 16 mm in diameter after 4 days of culture on solid LB medium. Gram stain results showed that ZM1 was a gram-positive bacterium and ZM2 was a gram-negative bacterium. Both ZM1 and ZM2 were rod shaped and measured about 0.5 to 0.7 × 3 to 4 μm and 0.4 to 0.6 × 0.8 to 1 μm, respectively. To identify the isolated bacteria, the 16S rDNA sequences of ZM1 and ZM2 were amplified with the primer pairs Eubac27F/1492R (Palkova et al. 2021) and Eubac338F/926R (Mao et al. 2012), respectively. The amplified products were sequenced and deposited in GenBank. BLAST results showed that the amplicon of ZM1 (896 bp; OQ642309) showed 100% homology to Bacillus sp. (MZ021595.1) and ZM2 (520 bp; OQ372972) had 100% homology to Pectobacterium carotovorum (MN393976.1). For pathogenicity testing, suspensions of ZM1 and ZM2 were prepared and adjusted to 107 CFU/ml, which were then injected into bulbs of healthy A. fistulosum plants above the soil line using a medical syringe and needle. The inoculation experiment of each bacterium consisted of three replicates, each with five A. fistulosum individuals, whereas sterile water was used as a control. All plants after inoculation were incubated in a growth chamber under 16-h light and maintained at 28°C and 30% relative humidity. After 1 week, the bulbs injected with ZM2 were rotten inside, which was consistent with the symptoms observed in the original plants, whereas the individuals inoculated with ZM1 and the control group remained disease-free. Subsequently, bacteria were reisolated from the diseased hosts inoculated with ZM2 by using the method described above and identified by amplifying and sequencing the 16S rDNA sequence with the primer pair Eubac338F/926R. To further confirm the identity of ZM2, the full length of gyrB was amplified with the primer pair gyrB-F/gyrB-R (F: 5′-ATGTCGAATTCTTATGACTC-3′, R: 5′-TCAGATATCGATATTCATCG-3′). The BLAST result in NCBI showed that the gyrB sequence of ZM2 (2,384 bp; OQ676993) had 98.84% homology with the P. carotovorum subsp. carotovorum strain DOAC-B772 (MF787381.1). The inoculation experiments were repeated twice, and the same results were obtained, fulfilling Koch’s postulates. P. carotovorum is widely known as a soft rot pathogen and has been reported to occur on many horticultural plants such as potato (Panda et al. 2012), cabbage (Nazerian et al. 2011), and pepper (Gillis et al. 2017); however, A. fistulosum has not been reported previously as a susceptible host to P. carotovorum. To our knowledge, this is the first report of soft rot caused by P. carotovorum on A. fistulosum in China.
