Abstract:Objective:To establish proteomic profiling using functional magnetic bead based sample fractionation method combinded with matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS) technology and to seek potential biomarkers of ovarian cancer for early diagnosis.Methods:The plasma protein profiling of two groups of patients with ovarian tumors (including 20 ovarian papillary cystadenocarcinoma patients and 9 ovarian papillary cystadenoma patients) and one group of 10 normal controls by the ClinProt magnetic bead enrichment and MALDI-TOF-MS was analyzed.Every group was repeated 3 times.Results:The mean coefficients of variance (CV) for protein peak intensities in three groups were 0.151,0.246 and 0.147,respectively which were small.Two peaks with mass charge ratio (m/z) 1451 and 2673 optimally discriminated ovarian cancer from non-cancer control group.Conclusions:Functional magnetic bead based sample fractionation method combined with MALDI-TOF-MS is accurate and stable.Protein peaks with m/z 1451 and 2673 may be potential biomarkers for ovarian cancer.

Abstract:Objective: To explore the serum cyclic polypeptide biomarkers for ovarian cancer diagnosis. Methods: A total of 54 patients with epithelial ovarian cancer confirmed by pathology in Cancer Hospital, Chinese Academy of Medical Sciences from March 2018 to September 2018 were selected as the study subjects, and 40 healthy women with normal examination results in the cancer screening center were selected as the control. All of the samples were randomly divided into training set and validation set at the ratio of 1∶1 with a random number. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) combined with magnetic bead technology was used for detecting peptide profiling in serum samples to screen significantly differently expressed peptides between ovarian cancer group and control group of the training set (score>5). Receiver operating characteristic (ROC) curve analysis was used to screen differential peptide peaks with area under curve (AUC) ≥0.8, sensitivity and specificity>90% in the training set and validation set. Liquid chromatography-mass spectrometry (LC-MS/MS) was further used to determine the composition of differentially expressed peptides. Results: By comparing the peptide profiles of the two groups, 102 differential peptide peaks were initially detected in the mass-to-charge ratio range of 1 000 to 10 000. ROC curve analysis showed that there were 42 differential peptide peaks with AUC ≥0.8 in both training set and validation set, 19 of which were highly expressed in ovarian cancer group, and 23 were lowly expressed. There were 15 different peptide peaks in highly expressed ovarian cancer group with sensitivity and specificity over 90%. The mass-to-charge ratios were 7 744.27, 5 913.41, 5 329.87, 4 634.21, 4 202.02, 3 879.26, 3 273.35, 3 253.79, 3 234.34, 2 950.33, 2 664.51, 2 018.38, 1 893.37, 1 498.69 and 1 287.55. There were 15 different peptide peaks in lowly expressed ovarian cancer group with sensitivity and specificity over 90%, the mass-to-charge ratios were 9 288.46, 7 759.77, 5 925.24, 4 652.77, 4 210.42, 3 887.02, 3 279.90, 3 240.82, 2 962.15, 2 932.70, 2 022.42, 1 897.16, 1 501.69, 1 337.38 and 1 290.13. No protein composition was identified in 15 different peptide peaks in lowly expressed ovarian cancer group. The two protein compositions identified in 15 different peptide peaks in highly expressed ovarian cancer group were recombinant serglycin (SRGN) and fibinogen alpha chain (FGA), the mass-to-charge ratios of which were 1 498.696 and 5 913.417, respectively. The sensitivity and specificity of the two proteins for ovarian cancer diagnosis were 100%, 100% and 90.9%, 100%, respectively. Conclusion: SRGN and FGA are highly expressed in the serum of ovarian cancer patients, which may be potential diagnostic markers for ovarian cancer.
An Integrated Approach Utilizing Proteomics and Bioinformatics to Detect Ovarian Cancer

Abstract:Objective: To find new potential biomarkers and establish the patterns for the detection of ovarian cancer. Methods: Sixty one serum samples including 32 ovarian cancer patients and 29 healthy people were detected by surface-enhanced laser desorption/ionization mass spectrometry (SELDI-MS). The protein fingerprint data were analyzed by bioinformatics tools. Ten folds cross-validation support vector machine (SVM) was used to establish the diagnostic pattern. Results: Five potential biomarkers were found (2085 Da, 5881 Da, 7564 Da, 9422 Da, 6044 Da), combined with which the diagnostic pattern separated the ovarian cancer from the healthy samples with a sensitivity of 96.7%, a specificity of 96.7% and a positive predictive value of 96.7%. Conclusions: The combination of SELDI with bioinformatics tools could find new biomarkers and establish patterns with high sensitivity and specificity for the detection of ovarian cancer.

Abstract:Early detection and monitoring the treatment remain the most important factors in improving long-term survival of ovarian cancer patients. New biomarkers that individually or in combination improve the diagnostic performance of existing tumor markers are critically needed. This study uses proteomic approaches to identify new biomarkers for detection and monitoring of ovarian cancer.We analyzed protein profiles of three sets of sera using surface enhanced laser desorption and ionization time-of-light mass spectroscopy (SELDI-TOF-MS) on IMAC ProteinChip arrays and ProPeak software for bioinformatics data analysis. The first set of patients included 21 ovarian cancers, 18 benign diseases, and 20 normal patients. The second set included 32 ovarian cancers, 30 benign ovarian diseases, and 30 age-matched healthy controls. The third set included samples collected before and after chemotherapy from 18 ovarian cancer patients. All samples were collected at the Gynecology and Obstetrics Hospital of Fudan University in Shanghai, China. The datasets from low-intensity and high-intensity spectra were analyzed separately.Seven peaks were selected for their contribution to the separation of ovarian cancers from controls using the first and second set of samples. The same dysregulation trends were confirmed for six of the seven peaks in independent validation using the third set of samples.Using SELDI-TOF analysis of 195 unique specimens, we discovered with preliminary validation six distinct peaks that may potentially be useful in the detection and monitoring of ovarian cancer. Additional studies are required to determine the protein identities of these peaks and to further validate their performance as biomarkers.
Proteomic markers for early detection of ovarian cancer

Abstract:Objective To screening the early detection marker for ovarian cancer from multiple kinds of body fluid of patients with ovarian cancer to further understand the mechanism of the carcinogenesis of ovarian cancer.Methods Weak cation beads(WCX) combining with MALDI-TOF-MS were used to screen the ascites,urine,cyst fluid and serum of the same patient(total 30 cases of patients).Different peaks were checking in the serum of training set and testing set.Results The candadate peaks screened from the four kinds of body fluid were 6081 and 4466Da.The sensitivity and specificity of the candidate peaks for diagnosis of ovarian cancer were 100% and 100%,by leave-one-off compared with 100% and 87.5% by blinding method.Conclusions Biomarkers screened from four kinds of body fluids have highly specific and valuable for early detection of ovarian cancer to a certain extent.

Abstract:OBJECTIVE:The purpose of this study was to screen potential serum tumor biomarkers for the diagnosis of ovarian cancer.METHODS:The study includes 3 sets. The first set of patients included 37 ovarian cancers and 31 healthy women (healthy controls). The second set included 42 ovarian cancers, 33 patients with benign ovarian tumor, and 29 healthy women (noncancer controls). The third set included 39 ovarian cancers and 35 patients with benign ovarian tumor (benign controls). Serum samples from ovarian cancers, healthy controls, noncancer controls, and benign controls were analyzed by surface-enhanced laser desorption/ionization time-of-flight mass spectrometry.RESULTS:A 3-peak model (peaks of mass-to-charge ratio values at 5766.379 d, 5912.586 d, and 11695.56 d) was established in the training set that discriminated cancer from noncancer with high sensitivity (10/11, 90.90%) and specificity (19/20, 95.00%). The peaks corresponding to 3 potential biomarkers increased significantly with the degree of malignancy.CONCLUSIONS:The proteins represented by these 3 peaks are biomarker candidates for ovarian cancer diagnosis and/or monitoring treatment response.
Proteomic Studies of Early-Stage and Advanced Ovarian Cancer Patients.

Abstract:Objectives. The objectives of this Study were to evaluate the diagnostic value for ovarian cancer using proteomic pattern established by surface-enhanced laser desorption/ionization (SELDI-TOF-MS) profiling of plasma proteins coupled with support vector machine (SVM) data analysis, and to investigate whether the proteomic pattern established by advanced ovarian cancer could be used for diagnosis of early-stage ovarian cancer patients.Methods. The Study included 44 ovarian cancer patients (11 early-stage and 33 advanced ovarian cancer patients) and 31 age-matched non-cancer controls. SELDI-TOF-MS coupled with SVM analysis was performed to establish a proteomic pattern to discriminate 33 advanced ovarian cancer patients from 31 non-cancer controls. A blind test, including 11 early-stage ovarian cancer cases, was performed to investigate whether proteomic pattern established by advanced ovarian cancer could be used for diagnosis of early-stage ovarian cancer patients.Results. A seven-peak proteomic pattern was established which discriminated 33 advanced ovarian cancer patients from 31 non-cancer controls effectively. A sensitivity of 93.94% (31/33) and a specificity of 93.55% (29/31) were yielded from the proteomic pattern. Among the 7 protein peaks, 5 with mass charge ratio (m/z) 4099 Da, 5488 Da, 4144 Da, 4479 Da and 3940 Da were up-regulated, while 2 peaks, with m/z 13 783 Da and 8588 Da were down-regulated in the advanced ovarian cancer group compared with non-cancer control group. After blind test, 9 of 11 early-stage ovarian cancer patients were successfully diagnosed with the accuracy of 81.82% (9/11).Conclusions. This study demonstrated that SELDI-TOF-MS coupled with SVM is effective in distinguishing protein expression between ovarian cancer and non-cancer plasma and it may be feasible to diagnose early-stage ovarian cancer using proteomic pattern established by advanced ovarian cancer. The gained and lost protein peaks in plasma may exist in both early-stage and advanced ovarian cancer plasma. Further studies should be performed using larger sample numbers. (C) 2008 Elsevier Inc. Abstract:The need for specific and sensitive markers of ovarian cancer is critical. Finding a sensitive and specific test for its detection has an important public health impact. Currently, there are no effective screening options available for patients with ovarian cancer. CA-125, the most widely used biomarker for ovarian cancer, does not have a high positive predictive value and it is only effective when used in combination with other diagnostic tests. However, pathologic changes taking place within the ovary may be reflected in biomarker patterns in the serum. Combination of mass spectra generated by new proteomic technologies, such as surface-enhanced laser desorption ionization time-of-flight (SELDI-TOF) and artificial-intelligence-based informatic algorithms, have been used to discover a small set of key protein values and discriminate normal from ovarian cancer patients. Serum proteomic pattern analysis might be applied ultimately in medical screening clinics, as a supplement to the diagnostic work-up and evaluation.

Abstract:Abstract Objective This study explored serum protein biomarkers for predicting the efficacy of first-line chemotherapy in advanced epithelial ovarian cancer using MALDI-TOF-MS combined with magnetic bead technology. Methods A total of 182 epithelial ovarian cancer patients were admitted to the Department of Gynaecologic Oncology, Cancer Hospital of the Chinese Academy of Medical Sciences between March 2018 and September 2018. Sixty-four of these patients had stage III/IV disease and received neoadjuvant chemotherapy followed by interval debulking surgery (NACT-IDS) and adjuvant chemotherapy (mucinous and low-grade serous carcinoma were excluded). Serum samples were collected before treatment. After a median follow-up time of 16 months, 25 chemosensitive patients with progression-free survival (PFS) >12 months and 10 chemoresistant patients <6 months after adjuvant chemotherapy were selected as the research subjects. Matrix-assisted laser desorption/ionisation time-of-flight mass spectrometry (MALDI-TOF MS) was used for peptide profiling of the serum samples to compare significantly differentially expressed peptides between the chemosensitive group and chemoresistant group (score >5), and peptide peaks with AUC ≥ 0.8 were identified. Liquid chromatography–mass spectrometry (LC‒MS/MS) was further used to determine the composition of the differentially expressed peptides. Results In the range of 1000-10000 m/z, 71 differential peptide peaks between the chemoresistant and chemosensitive groups were initially detected. ROC curve analysis showed that among the 71 differential protein peaks, there were 22 differential polypeptide peaks with an AUC ≥0.8, of which 10 were highly expressed in the chemoresistant group and 12 were highly expressed in the chemosensitive group. LC‒MS/MS analysis identified the protein components of 4 polypeptide peaks among the 10 differentially expressed polypeptide peaks in the chemoresistant group: 4634.22 m/z as fragments of serum amyloid A-1, 1287.56 m/zas fragments of prothrombin, and 1099.56 m/z and 2018.39 m/z as fragments of complement C3; the sensitivity and specificity of these three proteins for predicting first-line chemotherapeutic efficacy were 100%, 100%, and 96% and 80%, 100%, and 100%, respectively. No protein component was identified among the 12 differential protein peaks in the chemosensitive group. Conclusion Serum amyloid A-1, prothrombin and complement C3 were highly expressed in chemoresistant patients; thus, they might be related to the poor efficacy of paclitaxel/carboplatin first-line chemotherapy in advanced ovarian cancer. It is necessary to further explore their predictive value.
Alterations in the serum proteome profile during the development of ovarian cancer

Abstract:Ovarian cancer causes more deaths than any other malignant tumor of the female reproductive system. This is because the condition usually goes undetected until the late stages. The purpose of the present study is to identify alterations in the serum proteome profile during the development of ovarian cancer and to provide an experimental basis for discovering new and valuable serum biomarkers for the early detection of ovarian carcinoma. Surface-enhanced laser desorption/ionization-time of flight (SELDI-TOF-MS) was used to profile changes in the serum proteome of Fischer 344 rats with ovarian cancer during the progress of tumor development. Sera were collected from the rats on day A (1 week before injection of tumor cells), day B (4 weeks after injection), and day C (6 weeks after injection). Each sample was subjected to SELDI-TOF-MS testing. Peak detection and alignment and selection of peaks with the highest discriminatory power were performed using proteinchip biomarker software. Decision tree analyses were performed using biomarker pattern software. Finally, 3 peaks were found to be the most valuable ones (3759, 4659 and 9318 Da). The expression frequency of m/z 3759-Da peaks was downregulated and another two frequencies (4659 and 9318 Da) were upregulated, and the levels of expression of these three proteins showed the same tendency as the expression frequency during the development of ovarian cancer. The total accuracy rate of diagnosis at 4 and 6 weeks post-injection was 94.7 and 97.3%, respectively. Profiling the serum proteome changes during the process of the cancer development using SELDI-TOF-MS may provide useful information regarding carcinogenesis and facilitate discovery of novel serum biomarkers for early detection.
A Multiplex Methylation-Specific Pcr Assay For Detection Of Early-Stage Ovarian Cancer Using Cell-Free Serum Dna

Abstract:5535 Background: Epithelial ovarian cancer (EOC) remains the most lethal disease among gynecological malignancies. Prompt diagnosis is challenging because of the non-specific symptoms exhibited during the early stage of the disease. So there is an urgent need for better detection methods. Here we performed this work to build up a platform of multiplex methylation-specific PCR (MSP) assay to improve the early detection of ovarian cancer, via identifying the methylation status of cell-free serum DNA. Methods: After screening, we chose seven genes (APC, RASSF1A, CDH1, RUNX3, TFPI2, SFRP5 and OPCML) with a high frequency of methylation as candidate genes to construct the multiplex-MSP assay. When methylation of at least one of the seven genes was observed, the multiplex-MSP assay was considered positive. We performed the retrospective and screening study to verify its specificity and sensitivity in the detection of EOC. Results: The methylation status of cell-free serum DNA was examined in the preoperative serum of 202 patients, including 87 EOC cases (stage I, n=41, stage II-IV, n=46), 53 benign ovarian tumors and 62 healthy controls. As expected, multiplex MSP assay achieved a sensitivity of 85.3% and a specificity of 90.5% in stage I EOC, strikingly higher than that of single CA125, producing a sensitivity of 56.1% at 64.15% specificity [p=0.0036](Table). Conclusions: Multiplex MSP assay analyzing the methylation status of cell-free serum DNA is a suitable and reliable approach to improve the early detection of ovarian cancer, potentially benefiting a broad range of applications in clinical oncology. Identifying Potential Markers for Monitoring Progression to Ovarian Cancer Using Plasma Label-free Proteomics

Abstract:Background: Cancer antigen 125 (CA125) is considered to have high sensitivity but poor specificity for ovarian cancer. New biomarkers utilized to early detect and monitor the progression of ovarian cancer patients are critically needed.Methods: A total of 80 patients including 16 early stage, and matched with 17 late stage, 23 benign ovarian tumor (BOT) and 24 uterine fibroid (UF) patients were utilized to perform plasma proteomics analysis using isobaric tag for relative and absolute quantitation (iTRAQ) method to identify differential diagnostic proteins of ovarian cancer patients. A validation set of 9 early stage, 11 late stage, 17 BOT and 16 UF collected by an independent cohort of samples with the same matching principles was examined to confirm the expressed levels of differential expression proteins by ELISA analysis.Results: CRP and ARHGEF 11 were identified as potential diagnostic biomarkers of ovarian cancer. Results of area under the curve (AUC) analysis suggested that combination of diagnostic proteins and CA125 achieved a much higher diagnostic accuracy compared with CA125 alone (AUC values: 0.98 versus 0.80), especially improved the specificity (0.97 versus 0.77). In addition, elevated plasma CRP levels were associated with increased risk of ovarian cancer.Conclusions: Current study found that plasma protein CRP was an indicator for monitoring the progression of ovarian cancer. Combination of plasma protein biomarkers with CA125 could be utilized to early diagnose of ovarian cancer patients.

Abstract:Purpose Noninvasive and convenient biomarkers for early diagnosis of breast cancer remain an urgent need. The aim of this study was to discover and identify potential protein biomarkers specific for breast cancer. Methods Two hundred and eighty-two (282) serum samples with 124 breast cancer and 158 controls were randomly divided into a training set and a blind-testing set. Serum proteomic profiles were analyzed using SELDI-TOF-MS. Candidate biomarkers were purified by HPLC, identified by LC-MS/MS and validated using ProteinChip immunoassays and western blot technique. Results A total of 3 peaks ( m / z with 6,630, 8,139 and 8,942 Da) were screened out by support vector machine to construct the classification model with high discriminatory power in the training set. The sensitivity and specificity of the model were 96.45 and 94.87%, respectively, in the blind-testing set. The candidate biomarker with m/z of 6,630 Da was found to be down-regulated in breast cancer patients, and was identified as apolipoprotein C-I. Another two candidate biomarkers (8,139, 8,942 Da) were found up-regulated in breast cancer and identified as C-terminal-truncated form of C3a and complement component C3a, respectively. In addition, the level of apolipoprotein C-I progressively decreased with the clinical stages I, II, III and IV, and the expression of C-terminal-truncated form of C3a and complement component C3a gradually increased in higher stages. Conclusions We have identified a set of biomarkers that could discriminate breast cancer from non-cancer controls. An efficient strategy, including SELDI-TOF-MS analysis, HPLC purification, MALDI-TOF-MS trace and LC-MS/MS identification, has been proved very successful.

Abstract:OBJECTIVE:To develop the rationales for ovarian cancer-specific protein profiles in serum and to analyze the protein profiles of multidrug resistance P-glucoprotein (MDR1) and multidrug resistance-associated protein (MRP) positive and negative expression sets for a rapid and sensitive serum protein pattern.METHOD:Serum protein profiles from 36 epithelial ovarian cancer cases were compared with 30 healthy controls using surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (DELDI-TOF-MS). Blinded test was conducted subsequently for identification of the protein pattern. Expression of MDR and MRP were detected in set of training cases by immunohistochemistry. The spectra were analyzed statistically between positive and negative groups.RESULTS:Twenty-nine peaks were significantly different between ovarian cancer and healthy controls (P < 0.01, 15 peaks up-regulated and 14 down-regulated). A set of three peaks, at 5 486, 6 463 and 8 575 m/z respectively, were selected from 29 sense peaks as an ovarian cancer biomarker. The sensitivity was 100% and specificity 93.33%. Identification by blinded validation indicated a positive predictive value of 90%. MDR1 expression in ovarian cancer was 69.4 %. Twenty peaks were significantly different between positive and negative sets (P < 0.01). MRP expression in ovarian cancer was 63.8%. But one peak was detected (P < 0.01).CONCLUSION:It was an ideal pattern for employing the SELDI mass spectrum approach to diagnose ovarian cancer and select the multidrug resistance cases. Serum biomarkers for detecting drug resistance in ovarian cancer can be established on the basis of MDR1 immunohistochemistry.
Evaluation of proteomics-identified CCL18 and CXCL1 as circulating tumor markers for differential diagnosis between ovarian carcinomas and benign pelvic masses

Abstract:A lack of sensitive and specific tumor markers for early diagnosis and treatment is a major cause for the high mortality rate of ovarian cancer. The purpose of this study was to identify potential proteomics-based biomarkers useful for the differential diagnosis between ovarian cancer and benign pelvic masses. Serum samples from 41 patients with ovarian cancer, 32 patients with benign pelvic masses, and 41 healthy female blood donors were examined, and proteomic profiling of the samples was assessed by surface-enhanced laser desorption/ionization time-of-flight (SELDI-TOF) mass spectroscopy (MS). A confirmatory study was also conducted with serum specimens from 58 patients with ovarian carcinoma, 37 patients with benign pelvic masses, and 48 healthy women. A classification tree was established using Biomarker Pattern Software. Six differentially expressed proteins (APP, CA 125, CCL18, CXCL1, IL-8, and ITIH4) were separated by high-performance liquid chromatography and identified by matrix-assisted laser desorption/ionization (MALDI)-MS/MS and database searches. Two of the proteins overexpressed in ovarian cancer patients, chemokine CC2 motif ligand 18 (CCL18) and chemokine CXC motif ligand 1 (CXCL1), were automatically selected in a multivariate predictive model. These two protein biomarkers were then validated and evaluated by enzyme-linked immunosorbent assay (ELISA) in 535 serum specimens (130 ovarian cancer, 64 benign ovarian masses, 36 lung cancer, 60 gastric cancer, 55 nasopharyngeal carcinoma, 48 hepatocellular carcinoma, and 142 healthy women). The combined use of CCL18 and CXCL1 as biomarkers for ovarian cancer had a sensitivity of 92% and a specificity of 97%. The multivariate ELISA analysis of the two putative markers in combination with CA 125 resulted in a sensitivity of 99% for healthy women and 94% for benign pelvic masses, and a specificity of 92% for both groups; these values were significantly higher than those obtained with CA 125 alone (p and lt;0.05). We conclude that serum CCL18 and CXCL1 are potentially useful as novel circulating tumor markers for the differential diagnosis between ovarian cancer and benign ovarian masses.

Abstract:Despite many years of studies, ovarian cancer remains one of the top ten cancers worldwide. Its high mortality rate is mainly due to lack of sufficient diagnostic methods. For this reason, our research focused on the identification of blood markers whose appearance would precede the clinical manifestation of the disease. ITRAQ-tagging (isobaric Tags for Relative and Absolute Quantification) coupled with mass spectrometry technology was applied. Three groups of samples derived from patients with: ovarian cancer, benign ovarian tumor, and healthy controls, were examined. Mass spectrometry analysis allowed for highlighting the dysregulation of several proteins associated with ovarian cancer. Further validation of the obtained results indicated that five proteins (Serotransferrin, Amyloid A1, Hemopexin, C-reactive protein, Albumin) were differentially expressed in ovarian cancer group. Interestingly, the addition of Albumin, Serotransferrin, and Amyloid A1 to CA125 (cancer antigen 125) and HE4 (human epididymis protein4) improved the diagnostic performance of the model discriminating between benign and malignant tumors. Identified proteins shed light on the molecular signaling pathways that are associated with ovarian cancer development and should be further investigated in future studies. Our findings indicate five proteins with a strong potential to use in a multimarker test for screening and detection of ovarian cancer.

Abstract:The serum MALDI-TOF MS spectrum includes signals for serum proteins and peptides between 1000 and 12,000 Da in size, presenting a fingerprint-like pattern. However, whole serum MALDI-TOF MS signals are complex and prejudiced for data analysis. Pre-treatment with specific nanomaterials can simplify the mass spectrum while retaining the characteristics of the fingerprint pattern. In the present study, we used hydrophilic interaction chromatography nanoparticles (HICNPs) to enrich proteins and peptides in serum from a large number prostate cancer samples and controls. After pre-treatment with HICNPs, the serum MALDI-TOF MS signals for samples were simpler, with more analysable fingerprint-like patterns. Principal component analysis and partial least squares discriminant analysis of the samples demonstrated a significant difference in the MALDI-TOF signals between prostate cancer and controls, with an analytical accuracy of 77%, approaching that of methods based on prostate-specific antigen. Due to the low cost and high flux, MALDI-TOF MS fingerprinting can be used in largescale evaluation of various cancers, including prostate cancer.
Identification and validation of differential plasma diagnostic biomarkers of ovarian cancer based on lipidomics

Abstract:OBJECTIVE Ovarian cancer is one of the most common gynecologic maligancies.About 90％ of ovarian cancer patients were diagnosed at an advanced stage (Ⅲ/Ⅳ).Despite improvements in mulitimodality treatment,the 5-year overall survival for advanced ovarian cancer is less than 20％,while patients recognized at early stage (Ⅰ / Ⅱ),the 5-year survival rates are more than 90％.This study aimed to identify the differential plasma lipids biomarkers to improve the accuracy of ovarian carcinoma diagnosis.METHODS We combined the results from multivariate sparse-group lasso method and univariate Wilcoxon rank sum test to identify the potentially differential lipids biomarkers of the plasma from 139 ovarian carcinoma and 76 benign tumor patients which collected from Department of Gynecology of Harbin Medical University Tumor Hospital between March 1,2011 and July 31,2013.We identified the biomarkers that are not related with CA125 and assessed the diagnostic accuracy of differential lipids and the combined diagnostic accuracy of those complementary biomarkers with CA125 using the 7-fold cross-validation method.In addition,cytoscape software was used to construct the network of these differential lipids to study the relationship of them.Finally,we identified the differential lipids biomarkers to distinguish early-stage from late-stage patients.RESULTS We identified 20 differential lipids between ovarian cancer and controls,of which Stearamide,Stearic acid,Arachidic acid,PI(42 ∶ 9) were not related with CA125 (P＜0.05),the AUC value of the combination of these lipids with CA125 was 0.94,which was larger than that with CA125 alone.Of the 20 lipids,8 lipids were selected as potential biomarkers to distinguish those early-stage from late-stage patients,they are PC(35∶4),PC(38 ∶ 6),PC(46 ∶ 4),PC(P-35 ∶ 2),PE(P-36 ∶ 6),PG(34 ∶ 2),Cer (d18 ∶ 1/16 ∶ 0),3-Deoxyvitamin D3,respectively,and all P＜0.05.CONCLUSIONS Studying on the plasma lipidomics of ovarian cancer has improved the diagnostic accuracy of CA125 alone.Eight biomarkers could be used for early diagnosis of ovarian carcinoma.These results should be further validated in a large prospective cohort study.
Discrimination Analysis of Mass Spectrometry Proteomics for Ovarian Cancer Detection

Abstract:Aim: A discrimination analysis has been explored for the probabilistic classification of healthy versus ovarian cancer serum samples using proteomics data from mass spectrometry (MS). Methods: The method employs data normalization, clustering, and a linear discriminant analysis on surface-enhanced laser desorption ionization (SELDI) time-of-flight MS data. The probabilistic classification method computes the optimal linear discriminant using the complex human blood serum SELDI spectra. Cross-validation and training/testing data-split experiments are conducted to verify the optimal discriminant and demonstrate the accuracy and robustness of the method. Results: The cluster discrimination method achieves excellent performance. The sensitivity, specificity, and positive predictive values are above 97% on ovarian cancer. The protein fraction peaks, which significantly contribute to the classification, can be available from the analysis process. Conclusion: The discrimination analysis helps the molecular identities of differentially expressed proteins and peptides between the healthy and ovarian patients.

Abstract:Background. Currently available tests are insufficient to distinguish patients with epithelial ovarian cancer (EOC) from normal individuals. Metabolomics, a study of metabolic processes in biologic systems, has emerged as a key technology in the measurements of small molecular metabolites in tissues or biofluids. Material and methods. To investigate the application of metabolomics on selecting EOC-associated biomarkers, 173 plasma specimens (80 newly diagnosed EOC patients and 93 normal individuals) were analyzed using ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC/QTOF/MS). A two-step strategy was performed to select EOC-associated biomarkers. The first step was to select potential biomarkers in distinguishing 42 cancer patients from 58 normal controls through partial least-squares discriminant analysis (PLS-DA) and database searching, and the second step was to validate the discrimination performance of these biomarkers in a dataset contained 38 EOCs and 35 controls. Results. Eight candidate biomarkers were selected. The combination of these biomarkers resulted in the area of receiver operating characteristic curve (AUC) of 0.941, a sensitivity of 0.921, and a specificity of 0.886 at the best cut-off point for detecting EOC. Discussion. Our findings suggested that sharp differences in metabolic profiles exist between EOC patients and normal controls. The identified eight metabolites associated with EOC may be served as novel biomarkers for diagnosis.
