Effects of Valerian on number of p-CREB positive neurons in cerebral hippocampus of rats with depression induced by chronic mild stress

Abstract:Objective To explore the effects of the optimal dosage Valerian on behavior,and phosphorylated cAMP responsive element-binding protein(p-CREB) positive neurons in cerebral hippocampus of rats with depression induced by chronic mild stress.Methods Thirty five rats were randomly divided into 7 groups:normal control group,untreated group,negative control group,positive control group,and low-,medium-and high-dose Valerian-treated groups.During the intragastric administration,the body weight,tap water intake and 1% sucrose intake were examined to all groups of rats once a week.After the administration,the neuronal number of cerebral hippocampus was counted,the optimal dosage Valerian was obtained eventually.According to the above methods,all rats were perfused and fixed after the intragastric administration was over.The pCREB and brain-derived neurotrophic factor(BDNF) positive neurons of hippocampal sections were detected by immunohistochemistry and neutral red staining.Results 1% sucrose intake of rats with depression recovered to the normal level after the optimal dosage Valerian was administered,and the number of neurons and p-CREB positive neurons in cerebral hippocampus of rats with depression were also increased to the normal level.BDNF positive neurons of hippocampus in the Normal control group were lightly stained.There was no obvious difference in BDNF positive neuron staining of cerebral hippocampus in the rest groups of rats compared with the Normal control group.Conclusion High-dose(100 mg·kg-1·d-1) Valerian may be a optimal dosage to treat the rats with depression induced by chronic mild stress in the present study.Valerian may improve the behavior activity and recover the number of neurons and p-CREB positive neurons of cerebral hippocampus in the depressive rats to the normal level.
