Abstract:The stannylene based approach to regioselective sugar derivatization has been widely employed and reflects the satisfactory regioselectivity.The structure of dimer and polymer complex of tributyhin ether and dibutylstannylene acetal with sugar is well elucidated:the type of oxygen di-coordinated in an apical position of the dipyramid become more negative and hence more nucleophilic as the result of the electron-enriching effect of the tin atom redistribution,which account for the origin of regioselectivity. It is also discussed in detail with their regioselctive derivatization of tin-mediated complex systems of pyran-carbohydrate polyols of different types.

Abstract:A reduction-coupling-etherification reaction between aromatic aldehydes or ketones (1) and alcohols ( 2 ) to yield corresponding diether ( 3 ) of pinacol in the presence of tin and chlorotrimethylsilane. The scope and limitations of this reaction were studied and a reasonable mechanism was proposed.

Abstract:Dibutyltin oxide reacts selectively with hydroxyl groups of carbohydrates to form dibutylstannylene acetal,and polymerization of the acetal in solution enlarges the electronegative and nucleophilic difference of hydroxyl groups situated on the different locations of polymer,which accounts for selective synthesis of sugar derivatives.Hence the chemical characteristic of dibutyltin oxide is well elucidated via its application on sides of protection of adjacent hydroxyl groups of sugars containing active functional groups,regioselective bridging of carbohydrates and glycosylation,which lays foundation for this reagent to extend its application into selective synthesis of sugar derivatives.

Abstract:Regioselectivity of lactose esterification with fatty acid using dibutyltin dimethoxide as stannylating agent were probed. The general factors affecting regioselectivity have been examined. The result shows the C-3' position of galactose and the beta anomer of C-1 position of the glucose residue have some advantage of being acylated at the condition of N-methyl-1-pyrrolidione as solvent and lower temperature (-15 degrees C), the corresponding isomers of lactose fatty acid esters were separated by preparative HPLC method. Explanations for the regioselectivities observed during this tin-mediated reaction were also proposed based on the stannylene acetal in solution.

Abstract:Transition metal-catalyzed hydrostannylation of alkynes is an atom-economic way towards valuable α-vinylstannanes. However, directing-group-free examples for the construction of α-adducts are rare and mostly undergo the M-Sn species insertion process. Here, we report an efficient cobalt-catalyzed highly α-selective hydrostannylation of terminal alkynes via M-H species, using readily available Bu3SnH and easily accessible as well as bench-stable ligand. The robust catalytic system, bearing good functional group tolerance and operational simplicity, provides the α-vinylstannanes in moderate to excellent yields with ⩾97:3 rr averagely. The efficiency of the reaction is investigated with turnover number (TON) up to 10,000 and turnover frequency (TOF) up to 144,000 h−1, respectively. The synthetic utility of the α-vinylstannanes is proved by further derivatizations towards the 1,1-disubstituted alkenes via Kosugi-Migita-Stille cross-coupling, conjugated alkene, α-vinyliodine, α-vinylazide, and allyl alcohol. Several control experiments, deuterium-labeling experiment and density functional theory (DFT) calculations are conducted to reveal the preliminary mechanism.

Abstract:A synthetic method based on Sandmeyer-type reactions to access both tin- and boron-substituted arenes from nitroaniline derivatives is described. This transformation can be applied to the synthesis of a series of functionalized trimethylstannyl arylboronates. In addition, the chemoselective reaction of the Stille and Suzuki-Miyaura cross-coupling reactions is explored, and a series of m- and p-terphenyl derivatives have been synthesized by conducting consecutive one-pot Stille and Suzuki-Miyaura cross-coupling reactions.

Abstract:Rh(acac)(CO)(2) catalyzes addition of (N,N-diisopropylcarbamoyl)trimethylstannane to terminal alkynes to give (Z)-beta-stannyl-alpha,beta-unsaturated amides with terminal attachment of the amide group, while the use of Ni(COd)(2) results in the reversal of the regioselectivity. A mechanism initiated by the oxidative addition of the carbamoylstannane was proposed on the basis of the isolation of (trimethylstannyl)(N,N-diisopropylcarbamoyl)[1,2-bis(dimethylphosphtno)ethane]pallidium.

Abstract:RegioselectiVity of methyl alpha-D-glucopyranoside benzylation using dibutyltin dimethoxide (DBDM) as stannylating agent was probed, general factors affecting regioselectivity have been examined. The results show the O-2 position of glucoside has advantage of being benzylated over the O-6 position. The major isomers of benzyl ether were separated by preparative HPLC method and characterized.

Abstract:Here, we reported a cobalt-hydride-catalyzed Markovnikov-type hydroallylation of terminal alkynes with allylic electrophile to access valuable and branched skipped dienes (1,4-dienes) with good regioselectivity. This operationally simple protocol exhibits excellent functional group tolerance and exceptional substrate scope. The reactions could be carried out in gram-scale with TON (turn over number) up to 1160, and the products could be easily derivatized. The preliminary mechanism of electrophilic allylation of α-selective cobalt alkenyl intermediate was proposed based on deuterium labeling experiment and kinetic studies.

Abstract:The mixture of 3,6-di-O-acetyl-4-O-(2,3,4,6-tetra-O-acetyl-β-D- galactopyranosyl)-2-azido-2-deoxy-D-glucopyranosyl acetates or trichloroacetates and the corresponding mannose type glycosyl donors reacted with spacer arms of di- and triethylene glycol in the solution of dichloromethanc with BF3·Et2O and TMSOTf as promoters at room temperature gave highly selective products. Only the mannose type products were obtained. In addition, some lactose type compounds can be prepared by another way.

Abstract:Carbohydrates, one of the three primary macromolecules of living organisms, play significant roles in various biological processes such as intercellular communication, cell recognition, and immune activity. While the majority of established methods for the installation of carbohydrates through the anomeric carbon rely on nucleophilic displacement, anomeric radicals represent an attractive alternative because of their functional group compatibility and high anomeric selectivities. Herein, we demonstrate that anomeric nucleophiles such as C1 stannanes can be converted into anomeric radicals by merging Cu(I) catalysis with blue light irradiation to achieve highly stereoselective C(sp3)-S cross-coupling reactions. Mechanistic studies and DFT calculations revealed that the C-S bond-forming step occurs via the transfer of the anomeric radical directly to a sulfur electrophile bound to Cu(II) species. This pathway complements a radical chain observed for photochemical metal-free conditions where a disulfide initiator can be activated by a Lewis base additive. Both strategies utilize anomeric nucleophiles as efficient radical donors and achieve a switch from an ionic to a radical pathway. Taken together, the stability of glycosyl nucleophiles, a broad substrate scope, and high anomeric selectivities observed for the thermal and photochemical protocols make this novel C-S cross coupling a practical tool for late-stage glycodiversification of bioactive natural products and drug candidates.

Abstract:Stereochemical control is an important issue in carbohydrate synthesis.Glycosyl donors with participating acyl protective groups on 2-O have been shown to give 1,2-trans glycosides reliably under the pre-activation based reaction condition.In this work,the effects of additives and reaction solvents on stereoselectivity were examined using donors without participating protective groups on 2-O.While several triflate salt additives did not have major effects,the amount of AgOTf was found to significantly impact the reaction outcome.Excess AgOTf led to lower stereochemical control presumably due to its coordination with the glycosyl triflate intermediate and a more SN1 like reaction pathway.In contrast,the stereoselectivity could be directed by reaction solvents,with diethyl ether favoring the formation of glycosides and dichloromethane leading to β isomers.The trend of stereochemical dependence on reaction solvent was applicable to a variety of building blocks including the selective formation of β-mannosides.

Abstract:Stereoselective C-glycosylation reactions are increasingly gaining attention in carbohydrate chemistry because they enable glycosyl precursors, readily accessible as anomeric mixtures, to converge to a single diastereomeric product. However, controlling the stereochemical outcome through transition-metal catalysis remains challenging, and methods that leverage bench-stable heteroaryl glycosyl sulfone donors to facilitate glycosylation are rare. Herein, we show two complementary nonprecious metal catalytic systems, based on iron or nickel, which are capable of promoting efficient C-C coupling between heteroaryl glycosyl sulfones and aromatic nucleophiles or electrophiles through distinct mechanisms and modes of activation. Diverse C-aryl glycosides were secured with excellent selectivity, scope, and functional-group compatibility, and reliable access to both α and β isomers was possible for key sugar residues.

Abstract:The chemical selectivity between Markovnikov addition and acylation in the reaction of thiols and vinyl esters were developed. The addition reaction was efficiently catalyzed by CAL-B at 50°C in isopropyl ether, affording the Markovnikov adducts with good yields. Acylation was carried out under the catalysis of d-amino acylase in DMF at 50°C. A series of thioethers and thioesters were synthesized via the controllable enzymatic methods.

Abstract:Highly regioselective alkylation of sugar hydroxyl groups has always been an important challenge in carbohydrate chemistry, especially for the selective alkylation of trans diols, there is no direct and efficient catalytic method so far. A chiral copper catalyzed universal highly site‐selective alkylation of trans‐diols method is realized. This reaction is performed under mild conditions and has broad substrate scope. The selectivity of the reactions can be controlled by adjusting the copper reagents and the chiral ligands. This method was also applied to the site‐selective modification of drugs and natural products.

Abstract:The development of catalytic enantioselective aldol reactions has been at the forefront of advancements in contemporary asymmetric methodology. Despite significant progress, the catalytic enantioselective synthesis of certain synthetically important motifs such as the ubiquitous aliphatic syn-diol remains a challenge. Herein, we report the application of a benzyloxy-functionalized malonic acid half thioester as an activated ester equivalent in a highly enantioselective glycolate aldol reaction. This robust method operates at ambient temperature, tolerates air and moisture, and generates CO2 as the only by-product. The synthetic applicability of the method is demonstrated by the large-scale enantiodivergent synthesis of -benzyloxy-β-hydroxybutyric acid thioester and its subsequent conversion to diverse polyoxygenated building blocks, deoxy-sugars, and the natural product (–)-angiopterlactone B.

Abstract:Herein, we demonstrate the robustness of a synergistic chiral Pd/organoboron system in tackling a challenging suite of site-, regio-, enantio- and diastereoselectivity issues across a considerable palette of biologically relevant carbohydrate polyols, when prochiral alkoxyallenes were employed as electrophiles. In view of the burgeoning role of noncovalent interactions (NCIs) in stereoselective carbohydrate synthesis, our mechanistic experiments and DFT modeling of the reaction path unexpectedly revealed that NCIs such as hydrogen bonding and CH-π interactions between the resting states of the Pd-π-allyl complex and the borinate saccharide are critically involved in the stereoselectivity control. Our strategy thus illuminates the untapped potential of harnessing NCIs in the context of transition metal catalysis to tackle stereoselectivity challenges in carbohydrate functionalization.

Abstract:A palladium-catalyzed highly C‒S-selective Stille cross-coupling between aryl thianthrenium salts and tri- or tetrasubstituted alkenyl stannanes is described. Herein, critical challenges including site- and chemoselectivity control are well addressed through C‒H thianthrenation and C‒S alkenylation, thereby providing an expedient access to stereodefined tri- and tetrasubstituted alkenes in a stereoretentive fashion. Indeed, the palladium-catalyzed Stille-alkenylation of poly(pseudo)halogenated arenes displays privileged capability to differentiate C‒S over C‒I, C‒Br, C‒Cl bonds, as well as oxygen-based triflates (C‒OTf), tosylates (C‒OTs), carbamates and sulfamates under mild reaction conditions. Sequential and multiple cross-couplings via selective C‒X functionalization should be widely applicable for increasing functional molecular complexity. Modular installation of stereospecific alkene motifs into pharmaceuticals illustrated the synthetic application of the present protocol in drug discovery.
