Abstract:The Rh(II)-carbene reaction is dramatically affected by the neighboring substituents. If the neighboring substituent is an OH group, a1,2-H shift is the exclusive pathway. If it is an OAc group, a 1,2-acetoxy migration is observed. If it is p-toluenesulfonyl group, 1,3 and 1,5-C-H insertion are the major pathways, and the 1,2-H shift is completely suppressed. If the adjacent substituent is a trichloroacetyl amino group, 1,5-C-H insertion competes with the 1,2-hydride shift, and no 1,3-C-H insertion can be observed. Both electronic and steric factors are responsible for the switching of the Rh(II)-carbene reaction pathway. The highly stereoselective 1,5-C-H insertions in Rh(II)-catalyzed reaction of alpha-diazocarbonyl compounds, bearing beta-trichloroacetylamino substituent, can be utilized as a novel way to synthesize five-membered cyclic beta-amino acid derivatives.

Abstract:In order to investigate the reaction mechanism of intramolecular C-H insertion by Rh(II)-mediated carbenoids with trans-(2-phenylcyclopropyl)carbinal radical as the mechanistic probe, diazo compounds 2-(2-phenylcyclopropyl)ethyl diazoacetoacetate 8a and methyl 2-diazo-3oxo-6-(2-phenylcyclopropyl)hexanoate 8b have been synthesized. Preliminary investigation of the intramolecular C-H insertion with Rh-2(OAc)(4) as catalyst supports a concerted insertion process.

Abstract:Transition‐metal‐catalyzed C‐H insertion of metal‐carbene represents an excellent and powerful approach for C‐H functionalization. However, despite remarkable advances in metal‐carbene chemistry, transition metal catalysts that capable of enantioselective intermolecular carbene C‐H insertion are mainly constrained to dirhodium(II) and iridium(III)‐based complexes. Here we disclose a new version of asymmetric carbene C‐H insertion reaction with rhodium(I) catalyst. A highly enantioselective rhodium(I) complex‐catalyzed C(sp3)‐H functionalization of 1,4‐cyclohexadienes with α‐aryl‐α‐diazoacetates was successfully developed. By using chiral bicyclo[2.2.2]‐octadiene as ligand, rhodium(I)‐carbene‐induced asymmetric intermolecular C‐H insertion proceeds smoothly at room temperature, allowing access to a diverse variety of α‐aryl‐α‐cyclohexadienyl acetates and gem‐diaryl‐containing acetates in good yields with good to excellent enantioselectivities (up to 99% ee). Furthermore, the synthetic utility of the reaction was highlighted by the facile synthesis of a novel cannabinoid CB1 receptor ligand. This method may offer a new opportunity for the development of therapeutically exploitable cannabinoid receptor type ligands in medicinal chemistry.This article is protected by copyright. Abstract:A Rh(I)-catalyzed three-component reaction of tert-propargyl alcohol, diazoester, and alkyl halide has been developed. This reaction can be considered as a carbene-involving sequential alkyl and alkynyl coupling, in which C(sp)-C(sp(3)) and C(sp(3))-C(sp(3)) bonds are built successively on the carbenic carbon atom. The Rh(I)-carbene migratory insertion of an alkynyl moiety and subsequent alkylation are proposed to account for the two separate C-C bond formations. This reaction provides an efficient and tunable method for the construction of all-carbon quaternary center.

Abstract:A Cp*Rh-III-catalyzed annulative carbooxygenation of challenging 1,1-disubstituted alkenes triggered by C-H activation of N-aryloxyacetamides has been established, which affords 2,3-dihydrobenzofuran derivatives with a quaternary carbon center in good to excellent yields under mild redox-neutral conditions. An amide group on the alkenes is essential for the process, and may inhibit the beta-H elimination from C(sp3)-Rh species by saturating the rhodium center through coordination. Furthermore, mechanistic insights obtained from control experiments suggest a mechanism involving a Rh-III-Rh-V-Rh-III catalytic cycle.

Abstract:The first rhodium( i )-catalyzed asymmetric S–H insertion of α-diazoarylacetates with tert -butyl mercaptan using chiral diene as the ligand has been developed. Asymmetric construction of C–S bonds through transition-metal catalysis is a challenging subject. By using chiral diene as the ligand, we have developed the first rhodium( i )-catalyzed asymmetric carbene insertion approach for C–S bond formation to access sulfur-substituted stereocenters under mild conditions, allowing the facile synthesis of a range of potentially valuable α-thioesters in high yields with moderate to good enantioselectivities.

Abstract:Transition-metal-catalyzed enantioselective transformations of aryl/aryl carbene are inherently challenging due to the difficulty in distinguishing between two arene rings in the reaction process thus remain largely less explored. The few successful examples reported so far, without exception, have all been catalyzed by Rh(II)-complexes. Herein, we describe our successful development of a novel cationic Rh(I)/chiral diene catalytic system capable of efficient enantioselective B-H and O-H insertions with diaryl diazomethanes, allowing the access to a broad range of gem-diarylmethine boranes and gem-diarylmethine ethers in good yields with high enantioselectivities. Notably, previously unattainable asymmetric diarylcarbene insertion into the O−H bond was achieved for the first time. A remarkable feature of this newly designed Rh(I)/diene catalyst bearing two ortho-amidophenyl substitutents is that it can distinguish between two arene rings of the diaryl carbene through a stereochemically selective control of π-π stacking interactions. DFT calculations indicate that the rotation-restricted conformation of Rh(I)/diene complex played an important role in the highly enantioselective carbene transformations. This work provides an interesting and unprecedented stereocontrol mode in diaryl metal carbene transformations.

Abstract:Asymmetric insertion of an arylvinylcarbenoid into the C-H bond for direct enantioselective C(sp2)-H functionalization of aniline derivatives catalyzed by a rhodium(I)-diene complex was developed for the first time. The reaction occurred exclusively at the uncommon vinyl terminus site with excellent E selectivity and enantioselectivities, providing various chiral γ,γ-gem-diarylsubstituted α,β-unsaturated esters with broad functional group compatibility under simple and mild conditions. It provides a rare example of the asymmetric C-H insertion of arenes with selective vinylogous reactivity. Synthesis applications of this protocol were featured by several versatile product transformations. Systematic DFT calculations were also performed to elucidate the reaction mechanism and origin of the uncommon enantio- and regioselectivity of the Rh(I)-catalyzed C(sp2)-H functionalization reaction. The measured and computed inverse deuterium kinetic isotope effect supports the C-C bond-formation step as the rate-determining step. Attractive interactions between the chiral ligand and substrates were also proposed to control the enantioselectivity.

Abstract:A Rh(i)-catalyzed cross-coupling of 2-bromoethyl aryldiazoacetates with tertiary propargyl alcohols has been successfully achieved. This reaction follows a reaction sequence which involves C–C bond cleavage, Rh(i) carbene formation, carbene migratory insertion and nucleophilic substitution to achieve the construction of C(sp2)–C(sp) and C–O bonds. The products feature both enyne and ketene acetal moieties, which have been widely used as building blocks in organic synthesis and polymer science.

Abstract:A mechanistic study was performed on the Rh-catalyzed stereoselective C-C/C-H activation of tert-cyclobutanols. The present study corroborated the previous proposal that the reaction occurs by metalation, β-C elimination, 1,4-Rh transfer, C=O insertion, and a final catalyst-regeneration step. The rate-determining step was found to be the 1,4-Rh transfer step, whereas the stereoselectivity-determining step did not correspond to any of the aforementioned steps. It was found that both the thermodynamic stability of the product of the β-C elimination and the kinetic feasibility of the 1,4-Rh transfer and C=O insertion steps made important contributions. In other words, three steps (i.e., β-C elimination, 1,4-Rh transfer, and C=O insertion) were found to be important in determining the configurations of the two quaternary stereocenters.

Abstract:The first insertion of an alkynyl carbene into N-H bonds under Rh-catalysis is developed. Alkynyl hydrazone carboxylates are used as donor-acceptor carbene precursors and are exquisitely inserted into the N-H bonds of various amines, amides, and 1,2-diamines. Abstract:The synthesis of iridium(III) complexes containing bulky porphyrin ligands is described. The chiral iridium(III) complex of the D4-symmetric Halterman porphyrin ligand [Ir((+)-D4-Por)Me(L)] (L = solvent) is an effective catalyst for enantioselective intramolecular carbene insertion into saturated C-H bonds of α-diazoesters, giving the corresponding cis-β-lactones in good isolated yields (up to 87%), excellent stereoselectivities (cis products exclusively), and good enantioselectivities (up to 78% ee). © 2013 American Chemical Society.
Electronic Effects of Rh(II)-Mediated Carbenoid Intramolecular C−H Insertion: A Linear Free Energy Correlation Study

Abstract:The relative rates of the Rh(II)-mediated carbenoid insertion into the benzylic C-H bonds of a series of para-substituted phenyl substrates have been measured by an indirect intramolecular competition method. Three catalysts, rhodium(II) acetate, rhodium(II) trifluoroacetate, and rhodium(II) acetamide, were investigated. The effect of solvent was also studied on the reactions with rhodium(II) acetate as catalyst. The relative rates were analyzed by the Hammett equation. The relative rates are found to correlate better with sigma than with sigma(+) for all three catalysts when CH2Cl2 is the solvent. For the insertion reactions catalyzed by Rh-2(OAc)(4), Rh-2(O2CCF3)(4), and Rh-2(acam)(4) in CH2Cl2, the Hammett correlations of the relative rates with a give reaction constants of -1.26 (r = 0.98), -0.66 (r = 0.97), and -1.39 (r = 0.98), respectively. The Hammett analysis also indicates no significant solvent effect. The mechanistic significance of these Hammett correlations is discussed.
