Abstract:A direct catalytic asymmetric aldol-type reaction of 3-substituted-2-oxindoles with glyoxal derivatives and ethyl trifluoropyruvate, catalyzed by a chiral N,N'-dioxide-Sc(OTf)(3) (Tf = trifluoromethanesulfonyl) complex, has been developed that tolerates a wide range of substrates. The reaction proceeds in good yields and excellent enantioselectivities (up to 93% yield, 99:1 diastereomeric ratio (dr), and >99% enantiomeric excess (ee)) under mild conditions, to deliver 3-(alpha-hydroxy-beta-carbonyl) oxindoles with vicinal quaternary-tertiary or quaternary-quaternary stereocenters. Even with 1 mol % catalyst loading or on scaleup (10 mmol of starting material), maintenance of ee was observed, which showed the potential value of the catalyst system. In studies probing the reaction mechanism, a positive nonlinear effect was observed and Sc(III)-based enolate intermediates were detected by using ESIMS. On the basis of the experimental results and previous reports, a possible catalytic cycle was assumed.

Abstract:We describe herein an intriguing method for the synthesis of biaryl aldehydes bearing both axial and central chirality through a desymmetric [3 + 2] cycloaddition reaction of activated isocyanides with prochiral biaryl dialdehydes under silver catalysis. This protocol features excellent enantioselectivity, 100% atom economy, good functional group compatibility, and operational simplicity.

Abstract:Two novel chrial bisaminoalcohols, 1, 3-bis-[(2' S)-(diphenylhydroxymethyl)pyrrolidin-1-yl]-2-propanone (3) and 1,3-bis-[(2'S)-(diphenylhydroxymethyl)pyrrolidin-1-yl]-2-propanol (4), were prepared using the 1, 3-dibromo-2-propanone and ( 21 S) - ( diphenylhydroxymethyl) pyrrolidine as the starting materials. The yields were 39% and 35%, respectively. The asymmetric direct aldol reactions of unmodified ketones and aldehydes were achieved with the ee up to 9% by using the bisaminoalcohol-diethylzinc as the catalyst.

Abstract:A series of poly(2-oxazoline) (POX) derivatives bearing prolinamide pendants were designed as organocatalysts and evaluated in the direct asymmetric aldol reaction between aromatic aldehydes and cyclic ketones. The structural variation of the alkyl spacer connecting the polymer backbone with the catalytic unit was applied so as to deduce structure–performance relationships combined with comparable experiments from model catalysts. Results showed that the POX-bound prolinamides can promote the aldol reaction more effectively as compared to their small-molecular and non-POX-bound analogs. The catalyst P3 containing the pyrrolidine moiety closer to the tertiary amide backbone exhibited the overall best catalytic efficiency, affording anti-products in 84% yield with 89% ee in the representative aldol addition of cyclohexanone to 4-nitrobenzaldehyde at a 10 mol.% catalyst loading. Furthermore, the influence of trifluoroacetic acid as an additive on the asymmetric transformation was investigated. Theoretical calculations revealed that the protonation of the aldehyde carbonyl group switched the activation mode of the aldol acceptor through hydrogen bond interactions, thereby changing the relative energy barrier of the enamine/aldehyde reaction transition states, which accounted well for the significant improvement in the enantioselectivity of the acidic additives observed experimentally.

Abstract:The direct asymmetric aldol reactions of equivalent molar amounts of aldehydes and ketones were carried out at -20 degrees C over alkaline Al(2)O(3) with 20 mol % of Pro-Trp as catalyst and 20 mol % of N-methylmorpholine or 1,4-diazabicyclo[2.2.2]octane as additive. After simple and environmentally friendly work-up, moderate to high isolated yields (up to 95%), good diastereoselectivities (>99:1), and enantioselectivities (up to 98% ee) have been achieved for the reactions of different kinds of ketones with various aldehydes. The catalytic system could be reused without decrease of activity by addition of 10 mol % catalyst and base in the catalytic system.

Abstract:Asymmetric allylboration of aldehydes with allylboronic acid pinacol ester catalyzed by chiral hydroxyl carboxylic acid is described. This reaction provides synthetically useful homoallyl alcohols in high yield with good to high enantioselectivity. The present catalytic protocol was also examined in asymmetric allenylboration of aldehydes at elevated temperature to afford chiral homopropargyl alcohols with reasonable asymmetric induction.

Abstract:Heterogeneous asymmetric addition of diethylzinc to aldehydes catalyzed by chiral polymer catalysts has been achieved. The conjugated polymers based on a BINOL skeleton were synthesized in five steps and applied to this reaction with yield and e.e. up to 95 and 84.5%, respectively. These chiral polymers can be easily quantitatively recovered and reused without decreasing of e.e. or catalytic activity.

Abstract:The development of catalytic enantioselective aldol reactions has been at the forefront of advancements in contemporary asymmetric methodology. Despite significant progress, the catalytic enantioselective synthesis of certain synthetically important motifs such as the ubiquitous aliphatic syn-diol remains a challenge. Herein, we report the application of a benzyloxy-functionalized malonic acid half thioester as an activated ester equivalent in a highly enantioselective glycolate aldol reaction. This robust method operates at ambient temperature, tolerates air and moisture, and generates CO2 as the only by-product. The synthetic applicability of the method is demonstrated by the large-scale enantiodivergent synthesis of -benzyloxy-β-hydroxybutyric acid thioester and its subsequent conversion to diverse polyoxygenated building blocks, deoxy-sugars, and the natural product (–)-angiopterlactone B.

Abstract:We report a copper‐catalyzed reductive aldol addition to aldehydes and ketones, with pinacolborane as stoichiometric reductant, that results in the generation of stereodefined syn‐aldol products. Cyclic, acyclic, fused and spirocyclic aldols bearing contiguous stereocenters are obtained with excellent yields and diastereoselectivities. Moreover, enantioselective reactions could be carried out with cycloalkenones to deliver aldols bearing three contiguous stereocenters and with up to 98% ee. Computations reveal that the enolborate intermediate undergoes the syn‐aldol reaction via a twist‐boat transition state that is stabilized by non‐classical hydrogen bonding interactions.

Abstract:We present herein simple primary-tertiary diamine-Brønsted acid conjugates that catalyze both syn- and anti-aldol reactions of dihydroxyacetones (DHAs) with high diastereoselectivities and enantioselectivities. This type of organocatalysts functionally mimics all four DHA aldolases, namely L-fuculose-1-phosphate aldolase, D-tagatose-1,6-diphosphate aldolase, D-fructose-1,6-diphosphate aldolase, and L-rhamnulose-1-phosphate aldolase.

Abstract:Compared with the well-explored enamine catalysis with secondary amines, the development of efficient enamine-based primary amine catalysts has remained as an elusive goal until recently. We present herein that a simple chiral primary-tertiary diamine 1d in combination with TfOH acts as an efficient enamine-based primary amine catalyst. The 1d-TfOH catalytic system could effectively catalyze the asymmetric direct aldol reaction with broad substrate scope as well as high regio- and diastereoselectivity and enantioselectivity under ambient temperature. Significantly, the reactions accommodate the synthetically important but challenging substrates, such as linear aliphatic ketones, with high regioselectivity and unprecedented syn diastereoselectivity. These results are in sharp contrast with the secondary amine mediated similar reactions wherein anti diastereoselectivity was normally observed.

Abstract:Catalytic enantioselective alkenylation of aldehydes with easily accessible alkenyl halides promoted by a chiral cobalt complex derived from a newly developed tridentate bisoxazolinephosphine is presented. Such processes represent an unprecedented reaction pathway for cobalt catalysis and a general approach that enable rapid construction of highly diversified enantioenriched allylic alcohols containing a 1,1-, 1,2-disubstituted and trisubstituted alkene as well as axial stereogenicity in up to 99% yield and 99:1 er without the need of preformation of alkenyl–metal reagents. DFT calculations revealed the origin of enantioselectivity.

Abstract:Using a single chiral phosphoramide-Zn(II) complex as the catalyst, the asymmetric β-H transfer reduction of aromatic α-trifluoromethyl ketones and enantioselective addition of aromatic aldehydes with Et2Zn in one pot were successfully realized, affording the corresponding additive products of secondary alcohols in high yields (up to 99%) with excellent enantioselectivities (up to 98% ee) and the reduction products of α-trifluoromethyl alcohols in good to excellent yields with up to 77% ee.
