Effect of CDK Inhibitor Olomoucine on the S-phase Arrest Induced by Benzo(a) Pyrene in 16HBE Cells

Abstract:Objective To observe the effect of olomoucine on cell cycle arrest induced by benzo(a)pyrene in 16HBE cells,to explore molecular mechanism of effect of BaP on cell cycle.Methods After exposure to 10 μmol /L BaP for 4 h in FBS-free medium,the 16HBE cells were added 50 μmol /L olomoucine,and then were harvested for cell cycle analysis and western blot analysis at 8 h,12 h and 24 h.Results The cell cycle analysis showed that olomoucine induced a time dependent increase in the number of cells in G0 /G1 phase,and decreased the effect of increase in the number of cells in S phase but prolonged the period of increase arose by BaP.Compared with BaP group,in group of olomoucine and BaP co-exposure the expression levels of CDK2 decreased at 8 h and 12 h,but increased at 24 h;CyclinE increased significantly,but CyclinA remained unchanged;phosphorylated Chk1 remained unchanged at 8 h,but decreased little at 12 h and 24 h;p53 increased significantly at 8 h,but inconspicuously at 12 h and 24 h;Cdc25A decreased at 8 h,but remained unchanged at 12 h and 24 h.Conclusion Olomoucine attenuated and prolonged the effect of BaP on the S-phase arrest by inhibiting the activity of CDK2,which the checkpoint proteins Chk1,p53,Cdc25A and CyclinE /CDK2 participated in.
