Tolerability Trial of Peramivir Trihydrate and Sodium Chloride Injection in Healthy Volunteers

Abstract:Objective: To assess the tolerability and safety of peramivir trihydrate and sodium chloride injection in healthy volunteers.Methods: Twenty-six healthy Chinese subjects were recruited to the single dose study.They were divided into 150,300,450,600 and 750 mg dose step,and 150 mg was the first dose step.Safety and tolerability findings at the previous dose level were the basis for the decision to proceed to the next higher dose level.Twenty healthy Chinese subjects were enrolled to the multiple dose study,the escalated dose level were 150 and 300 mg once a day for 5 consecutive days.The safety was assessed based on vital sign measurements,electrocardiograms(ECGs),physical examination,clinical laboratory testing and record of adverse events.Results: The main adverse events judged by the investigators to be related to peramivir trihydrate and sodium chloride injection were abnormalities in partly clinical laboratory values and ECGs.All adverse events were mild in severity and resolved without any treatment.No severe adverse events were observed in the trial process.Conclusion: Peramivir trihydrate and sodium chloride injection were safe and well tolerated in Chinese healthy subjects after a single dose of 150～750 mg and multiple doses of 150～300 mg,qd for 5 days.
