Abstract:Over the past decade, RNA interference (RNAi) plays an important role in biology, especially for silencing gene expression. RNA interference (RNAi) is a natural process through which expression of a targeted gene can be knocked down with high specificity and selectivity. Methods of mediating the RNAi effect involve small interfering RNA (siRNA) and short hairpin RNA (shRNA). In various applications, RNAi has been used to create model systems, to identify novel molecular targets, to study gene function in a genome-wide fashion, and to create new avenues for clinical therapeutics. This article reviews the current knowledge on the mechanism and applications of shRNA in mammalian and human cells.

Abstract:RNA interference,a phenomenon of gene silencing mediated by double.stranded RNA.is a simple and effective method for blocking specific gene expression in mammalian cells.Oncolytic viruses are a kind of vectors that replicate specifically in tumor cells.With oncolytic viruses.smail interference RNA could be delivered to target tumor cells.and the effects of oncogene silencing are also enhanced with the replication of viruses.Therefore,RNAi model mediated by oncolytic viruses opens up a new way for the gene therapy of tumors.

Abstract:RNA interference (RNAi) is a post-transcriptional gene silencing process by targeting mRNA for degradation in a sequence-specific manner. This powerful platform has enormous potential in functional genomics and medical research. As a tool to knock out expression of specific genes in a variety of organisms, RNAi was used to investigate gene function in a high throughput fashion. Highly conserved in evolution RNAi appears to have evolved as a cellular defense mechanism in plants and animals to suppress viral infection, transposon jumping and endogenous aberrant genes. Exploiting the natural mechanism, the researchers can shut down disease-causing genes and develop novel therapeutics against infection, tumor and other disease.

Abstract:RNA interference(RNAi)is a method of post-transcriptional silencing of genes induced by double-stranded RNA(dsRNA),which can be generated by small interfering RNA(siRNA),repeat-associated small interfererce RNA(rasiRNA)and microRNA(miRNA).Recently,RNAi has become a hot topic as a therapeutic method in neoplastic diseases,infectious diseases and nervous system diseases because of its high efficiency and specificity in silencing target genes.Although some of RNAi drugs have been used in clinical trials,most of them are still in the experimental phase.The application of RNAi in the clinical practice needs further study.

Abstract:The mechanism of a new phenomenon——genetic interference directed by double-stranded RNA was first discovered in 1998 by Andrew Fire and Craig Mello. It degrades mRNA specifically and potently through the mediation of corresponding double-stranded RNA and leads to post-transcriptional gene silencing. Consequently RNA interference was observed is eukaryotic organisms including fungi, Arabidopsis,C. elegans, trypanosomes, hydra, planaria, Drosophila, zabrafish, and mouse. More and more evidence supports that RNAi,co-suppression in plants, and quelling in fungi shares the same basic molecular mechanism. It indicates that this mechanism was acquired during early evolution. RNAi plays important roles in resistance to virus invasion, and inhibition of transportable elements. And it is very likely that RNAi is also important during the normal development and regulation of gene expression.

Abstract:Introduction. In the early 1990s, during experiments aimed at intensifying the colour of Petunia hybryda flowers, a new mechanism of regulation of gene expression was discovered; however, its mechanism, i.e. inhibition of gene expression at a post-transcriptional stage, remained unknown. In 1998 two groups led by A. Fire and C. Mello found a molecular basis for the phenomenon called RNA interference (RNAi). Delivery of a double stranded RNA to a model organism, Caenorhabditis elegans, triggered silencing of complementary messenger RNA sequences. This discovery opened new perspectives for research involving gene functions due to the possibility of inhibiting the expression of a specific gene through its mRNA degradation in the cytosol. Aim. The aim of this paper is to present a potential role of RNAi as a therapeutic method for various diseases. Discussion. RNAi provides a powerful technique for the derivation and analysis of loss-of-function phenotypes in vertebrate cells. This technique may be also applied as a therapeutic strategy, e.g. in genetic and viral diseases, and clinical trials to test this possibility have been already initiated. Conclusions. RNAi-based therapy may become a powerful tool to treat many diseases whose molecular pathogenesis mechanisms have been thoroughly understood.

Abstract:RNAi is a phenomenon of gene silence induced by double-stranded RNA(dsRNA) that is homologous to target gene. Now it is easy to obtain small interference RNA(siRNA), and siRNA is (often)transmitted into organisms by microinjection. RNAi includes encoding region RNAi and promoter region RNAi, which can be used to investigate gene function by degrading a special mRNA target in a cell or organism and thus knocking out or knocking down the level of the encoded protein. They have been used to the analysis of silkworm gene function. With the development of the technique, RNAi can be widely used to genomic analysis, medication target screening, disease treatment and so on.

Abstract:RNA interference (RNAi) is a phenomenon that the endogenous or exogenous double stranded RNA induces the degradation of the homologous mRNA, which leads to the inhibition of specific gene expression. Due to its efficacy and specificity, RNAi has been widely used in gene therapy. There is currently no effective strategy for tumor therapy except for traditional treatment such as surgery and chemotherapy at early stage. However, gene therapy involved in RNAi technology brings new hope to effective treatment of tumor. In this paper, we reviewed the mechanisms involved in RNAi, the advancement of siRNA in tumor gene therapy and challenges faced by people in the application of RNAi.

Abstract:During recent decades there have been remarkable advances in biology, in which one of the most important discoveries is RNA interference (RNAi). RNAi is a specific post-transcriptional regulatory pathway that can result in silencing gene functions. Efforts have been done to translate this new discovery into clinical applications for disease treatment. However, technical difficulties restrict the development of RNAi, including stability, off-target effects, immunostimulation and delivery problems. Researchers have attempted to surmount these barriers and improve the bioavailability and safety of RNAi-based therapeutics by optimizing the chemistry and structure of these molecules. This paper aimed to describe the principles of RNA interference, review the therapeutic potential in various diseases and discuss the new strategies for in vivo delivery of RNAi to overcome the challenges.

Abstract:Ribonucleic acid (RNA) plays a pivotal role in gene regulation and protein biosynthesis. Interfering the physiological function of key RNAs to induce cell apoptosis holds great promise for cancer treatment. Many RNA-targeted anti-cancer strategies have emerged continuously. Among them, RNA interference (RNAi) has been recognized as a promising therapeutic modality for various disease treatments. Nevertheless, the primary obstacle in siRNA delivery-escaping the endosome and crossing the plasma membrane severely impedes its therapeutic potential. Thus far, a variety of nanosystems as well as carrier-free bioconjugation for siRNA delivery have been developed and employed to enhance the drug delivery and anti-tumor efficiency. Besides, the use of small molecules to target specific RNA structures and disrupt their function, along with the covalent modification of RNA, has also drawn tremendous attention recently owing to high therapeutic efficacy. In this review, we will provide an overview of recent progress in RNA-targeted cancer therapy including various siRNA delivery strategies, RNA-targeting small molecules, and newly emerged covalent RNA modification. Finally, challenges and future perspectives faced in this research field will be discussed.

Abstract:RNA interference (RNAi), an effective technique for regulating or silencing specific genes, can be applied to treat various diseases. Multiple clinical trials using RNAi are ongoing, and molecular imaging can serve as a powerful tool in RNAi-based therapies. This brief review will highlight the current progress on in vivo imaging of RNAi delivery and silencing effects. Incorporation of suitable molecular imaging techniques into future RNAi-based clinical trials will provide more pieces of the puzzle, thus facilitating the transformation of RNAi into a powerful therapeutic modality in the clinic.

Abstract:An efficient mechanism for the sequence-specific inhibition of gene expression is RNA interference. In this process, double-stranded RNA molecules induce cleavage of a selected target RNA (see picture). This technique has in recent years developed into a standard method of molecular biology. Successful applications in animal models have already led to the initiation of RNAi-based clinical trials as a new therapeutic option.Only ten years ago Andrew Fire and Craig Mello were able to show that double-stranded RNA molecules could inhibit the expression of homologous genes in eukaryotes. This process, termed RNA interference, has developed into a standard method of molecular biology. This Review provides an overview of the molecular processes involved, with a particular focus on the posttranscriptional inhibition of gene expression in mammalian cells, the possible applications in research, and the results of the first clinical studies.
