Effects of Temperature on the Key Enzymes and Metabolic Flux Distribution of Glucose Metabolism in Zymomonas Mobilis

Abstract:In this work,we present a new method for combining the advantages of in vitro enzyme assays and metabolic flux analysis.Zymomonas mobilis strain ATCC31821 was used to study the effect of the key enzymes and metabolic flux distribution of the glucose metabolism on different temperature conditions. All experiments were carried out in a bioreactor (IOL) with 5.0L working volume.The fermentation medium was continually flushed with purging nitrogen gas and was agitated at 100r/min for anaerobic conditions, the pH was automatically controlled at 5.5 with the addition of 0.5mol/L NaOH, and the temperature was kept constant at 25,30,35 and 40℃, respectively. The concentrations of metabolites in the growth medium for 24h were determined by HPLC, and the activities of intracellular enzymes in ED, HMP and TCA under different temperature conditions were performed with freshly prepared cell-free extracts.The results showed that compared temperature 25℃ or 40℃,the activities of alcohol dehydrogenase (ADH), glucose- 6- phosphate dehydrogenase ( G- 6- PDH ), Pyruvate decarboxylase ( PDC ), glucosekinase ( GK), pyruvatekinase (PK) and glyceraldehyde-3- phosphate dehydrogenase ( GD-3- PDH ) were higher,the activities of malate dehydrogenase(MDH) and isocitrate dehydrogenase(ICDH) were lower at 30- 35℃.In addition,the amount of carbon flux into the TCA cycle was at least,and the carbon flux into the ED pathway for glucose catabolism was higher at 30-35℃.Therefore,in order to reduce energy distribution and the generation of by-product,fermentative temperature control on the metabolic pathway is more meaningful.
