Abstract:Sodium carbonate, cooking, ethanol and lipase were used to degrease for catfish. The results showed that the effect of lipase degreasing is better. The optimal degreasing conditions are ratio of feed liquid 1∶6, pH 9.0, 30 min, enzyme concentration 10 U/mL and the degreasing rate of 85.45%.

Abstract:Purification of eicosapentaenoic acid (EPA)-and docosahexenoic acid (DHA)-ethyl esters (EE) from fish oil ethyl esters by supercritical fluid chromatography(SFC) with carbon dioxide as mobile phase was studied. Reversed stationary phase (C18) and normal stationary phase (silica) were used respectively to find out the operating conditions suitable to enhance the purity and yield. The separation mechanism and efficiency of SFC using C18 and silica as stationary phase respectively were compared and discussed. It was proposed that a combination of SFC with C18 and silica as stationary phase respectively will be an effective way to improve the purity and yield of EPA-EE and DHA-EE.
Extraction of Fish Sauce Flavor Compounds and Optimization of Gas Chromatographic Separation Conditions

Abstract:The extraction solvent and analysis conditions of gas chromatography (GC) for fish sauce flavor compounds were studied. The data showed that ethyl acetate is the most suitable solvent for the fish sauce flavor extraction. A capillary column CP-Sil -8cb-30 m-0.32 mm was selected to quantitatively detect the fish sauce flavor compounds. The optimal temperatures of sample receiver and FID detector are 200 ℃ and 250 ℃, respectively, with nitrogen (purity >99.999%) as the carrier at the speed of 3 ml/min, at the shunting ratio100:1. The starting temperature is 30 ℃ for 2 min and programmed to rise from 30 ℃ to 120 ℃ by 5 ℃/min, continually to 180℃ by 12 ℃/min, then hold for 4 min. Under these optimized conditions, total 44 compounds are separated, and 24 of them can be separated from the baseline.

Abstract:Chitosan was applied to make fish ball from silver carp,and its correlative effects on gelling quality and lipid oxidation were evaluated by colour,texture,water retention,and TBA(2-thiobarbituric acid).The colour,texture and water retention were improved and the lipid oxidation was suppressed when 1.0%chitosan was added in gels.Adding 1.0%chitosan could be considered as a promising approach in the processing of silver carp gels to improve thermal gelling properties and delay lipid oxidation.

Abstract:Iron-carbon reduction method was used to treat high concentrated monosodium L-glutamate wastewater. Results showed that cast-iron was better than stainless steel to raise pH value of monosodium L-glutamate wastewater, and pH value could be increased from 1.97 to 4.88 when cast-iron was used. COD removal rate increased with pH rising in iron-carbon reduction process and the removal reached 52.5% at pH 9.0. The increment of Fe~(2+) concentration was 3395mg/L when pH was raised from 1.96 to 4.88.

Abstract:Catfish ( Clarias magur ) is a popular freshwater fish food worldwide. The processing of this fish generates a significant amount of waste, mainly in the form of viscera, which constitutes around 10–12% of the fish's total weight. This study was focused on extracting polyunsaturated fatty acid (PUFA)-rich oil from catfish viscera, aiming to enhance the extraction process and make the production of oil and handling of fish byproducts more cost-effective. The wet reduction method, a solvent-free approach, was used for extraction, with yield optimization done via the Box–Behnken design. The resulting oil was evaluated for its oxidative quality and chemical characteristics. The optimal conditions for the wet rendering process were as follows: viscera to water ratio, 1:0.5 (w/v); temperature, 90°C; and time, 20 min, yielding 12.40 g/100 g of oil. The oil extracted under optimal wet rendering conditions had quality and oxidative stability comparable to solvent extraction and fewer secondary oxidation compounds. This oil had a higher PUFA content, specifically a 4:1 ratio of omega 6 to omega 3. Such oil, derived from catfish viscera, is suitable for the food industry due to its solvent-free extraction method.

Abstract:A comprehensive processing technology of L.vannamei is developed to produce two kinds of products,seasoning base material and multi-functional feed additives.The operation conditions for the extraction of taste components by flavourzyme and papain are optimized by single factor experiment and Box-Behnken experiments.The results indicate that the optimum conditions are: initial pH value is 6.8,ratio of material and water is 1:1,the addition amount of salt is 3.0%,the dose of enzyme is 1046 U/g,hydrolysis temperature is 62.42 ℃,hydrolysis time is 3.28 h.Under the optimum conditions,the degree of hydrolysis is up to(28.15±0.10)%,and the bitterness value of hydrolysis is 33.05±0.21.The drying methods of hot air drying and intermittent microwave drying for feed additives are investigated,and it is found that intermittent microwave drying is superior to hot air drying.Base on the drying rate and drying power dissipation,the best drying process conditions for feed additives are obtained as 3 W/g(on/off ratio is 0.5) through single factor experiments.

Abstract:Fat deposition and off-flavor in the muscle are the main problems affecting flesh quality in aquaculture fish, especially in catfish, leading to low acceptability and reduced market price. Yellow catfish is an important aquaculture fish in China. In this study, 40 days of depuration and starvation treatment were explored to improve the muscle quality of aquaculture yellow catfish. After depuration and starvation, the body weight, condition factor (CF) and mesenteric fat index (MFI) were all significantly decreased 20 days after treatment. The metabolomic profiles in muscle were characterized to analyze the muscle quality in yellow catfish. The results showed that the content of ADP, AMP, IMP, glutamic acid and taurine were significantly increased between 20 and 40 days post-treatment in the muscle of yellow catfish during the treatment, which was positively associated with the flesh tenderness and quality. In contrast, aldehydes and ketones associated with off-flavors and corticosterone associated with bitter taste were all decreased at 20 days post-treatment. Considering the balance of body weight loss and flesh quality improvement, depuration and starvation for around 20 days is suitable for aquaculture yellow catfish. Our study not only provides an effective method to improve the flesh quality of aquaculture yellow catfish but also reveals the potential mechanism in this process.

Abstract:Effects of acid kind and dosage,water dosage,acid reaction temperature and time on degumming of crude rapeseed oil were investigated.On the basis of single factor experiment,the degumming conditions of crude rapeseed oil were optimized by response surface methodology.The optimal degumming conditions were determined as follows: acid reaction temperature 60 ℃,acid reaction time 24.78 min,citric acid(concentration 50%) dosage 0.22%(based on the mass of crude rapeseed oil) and water dosage 3.34%(based on the mass of crude rapeseed oil).Under these conditions,the phosphorous content of degummed rapeseed oil reduced to 15.67 mg / kg and the degumming rate was 93.76%,which were quite close to the predicted value of 14.70 mg / kg and 94.15%.

Abstract:In order to remove free fatty acids (FFA) in crude oil and improve edible oil quality, deacidification was often used in oil refining. In this work, several liquid enzymes were firstly screened for the selective deacidification of the degummed corn oil, and ethanol was used as a novel fatty acyl acceptor in solvent-free system, which showed a good selective deacidification efficiency and high triglyceride yield. Reaction conditions were optimized by response surface methodology as follows: 3% enzyme dosage and 20:1 substrate ratio (alcohol/FFA) at 50 o C for 50 min. Under these conditions, the acid value, yield of neutral oil, and contents of sterol and vitamin E of corn oil were 0.31 ± 0.06 mgKOH/g, 94.0 ± 0.5%, 1,012.9 ± 11.2 mg/100 g, and 1,320.0 ± 3.8 mg/kg, respectively. Meanwhile, thermodynamic and kinetic parameters were investigated. These results showed that this process was beneficial to selective deacidification, high yields of neutral oil and bioactive components in corn oil.

Abstract:Abstract Supercritical carbon dioxide (Sc-CO 2 ) is an alternative tool to extract lipid for the production of fish oil and enzyme from fish by-products (FBPs). In the application of Sc-CO 2 , this review covers sample preparation, lipid extraction operation, and characterization of fish oil and enzyme as final products. Generally, the fish samples with moisture content less than 20% and particle size less than 5 mm are considered before lipid extraction with Sc-CO 2 . Sc-CO 2 parameters, such as pressure (P), temperature (T), extraction time (t ext ), and flow rate (F), for simultaneous recovery of fish oil, protein, and enzyme were found to be less severe ( P : 10.3–25 MPa; T : 25–45 °C, t ext : 20–150 min; F : 3–50 g/min) than the extraction of fish oil alone ( P : 10–40 Mpa; T : 35–80 °C; t ext : 30–360 min; F : 1–3000 g/min). The enzyme from the Sc-CO 2 defatted sample showed higher activity up to 45 U/mg due to lower denaturation of protein as compared to the organic solvent treated sample albeit both samples having similar pH (6–10) and temperature stability (20–60 °C). Overall, mild extraction of lipid from FBPs using Sc-CO 2 is effective for the production of enzymes suitable in various industrial applications. Also, fish oil as a result of extraction can be produced as a health product with high polyunsaturated fatty acids (PUFAs) and low contamination of heavy metals.

Influence of solvent-free extraction of fish oil from catfish (Clarias magur) heads using a Taguchi orthogonal array design: A qualitative and quantitative approach

Abstract:This study aimed to efficiently utilize catfish heads, enhancing the oil extraction process while improving the cost-effectiveness of fish byproduct management. The study employed the wet rendering method, a solvent-free approach, utilizing a two-factor Taguchi orthogonal array design to identify critical parameters for optimizing oil yield and ensuring high-quality oil attributes. The extraction temperature (80-120°C) and time (5-25 min) were chosen as variables in the wet rendering process. Range analysis identified the extraction time as a more significant (p < 0.05) factor for most parameters, including oil yield, oil recovery, acid value, free fatty acids, peroxide value, and thiobarbituric acid reactive substances. The extraction temperature was more significant (p < 0.05) for oil color. Consequently, the wet rendering method was optimized, resulting in an extraction temperature of 80°C and an extraction time of 25 min, yielding the highest oil yield. This optimized wet rendering process recovered 6.37 g/100 g of oil with an impressive 54.16% oil recovery rate, demonstrating comparable performance to traditional solvent extraction methods. Moreover, Fourier transfer infrared spectra analysis revealed distinct peaks associated with triacylglycerols and polyunsaturated fatty acids (PUFA). The oil recovered under optimized conditions contained higher levels of PUFA, including oleic acid (189.92 μg/g of oil), linoleic acid (169.92 μg/g of oil), eicosapentaenoic acid (17.41 μg/g of oil), and docosahexaenoic acid (20.82 μg/g of oil). Volatile compound analysis revealed lower levels of secondary oxidation compounds under optimized conditions. This optimized wet rendering method offers practical advantages in terms of cost-efficiency, sustainability, reduced environmental impact, and enhanced oil quality, making it an attractive option for the fish processing industries. Future research possibilities may include the purification of the catfish head oil and its application in the food and pharmaceutical industries.
Enzyme-Assisted Extraction of Fish Oil from Whole Fish and by-Products of Baltic Herring (Clupea harengus membras)

Abstract:Baltic herring (Clupea harengus membras) is one of the most abundant commercially caught fish species from the Baltic Sea. Despite the high content of fat and omega-3 fatty acids, the consumption of Baltic herring has decreased dramatically over the last four decades, mostly due to the small sizes and difficulty in processing. At the same time there is an increasing global demand for fish and fish oil rich in omega-3 fatty acids. This study aimed to investigate enzyme-assisted oil extraction as an environmentally friendly process for valorizing the underutilized fish species and by-products to high quality fish oil for human consumption. Three different commercially available proteolytic enzymes (Alcalase®, Neutrase® and Protamex®) and two treatment times (35 and 70 min) were investigated in the extraction of fish oil from whole fish and by-products from filleting of Baltic herring. The oil quality and stability were studied with peroxide- and p-anisidine value analyses, fatty acid analysis with GC-FID, and volatile compounds with HS-SPME-GC-MS. Overall, longer extraction times led to better oil yields but also increased oxidation of the oil. For whole fish, the highest oil yields were from the 70-min extractions with Neutrase and Protamex. Protamex extraction with 35 min resulted in the best fatty acid composition with the highest content of eicosapentaenoic acid (EPA; 20:5n-3) and docosahexaenoic acid (DHA; 22:6n-3) but also increased oxidation compared to treatment with other enzymes. For by-products, the highest oil yield was obtained from the 70-min extraction with Protamex without significant differences in EPA and DHA contents among the oils extracted with different enzymes. Oxidation was lowest in the oil produced with 35-min treatment using Neutrase and Protamex. This study showed the potential of using proteolytic enzymes in the extraction of crude oil from Baltic herring and its by-products. However, further research is needed to optimize enzymatic processing of Baltic herring and its by-products to improve yield and quality of crude oil.

Effects of combined carnosine and ultra-high pressure on the inhibition of fishy off-odor of snakehead fillets and the possible mechanism

Abstract:The effects of combined application of ultra-high pressure (300 Mpa, 900 s) and carnosine on the inhibition of fishy off-odor from snakehead meat were investigated. Results showed that the combination effectively inhibited the formation of fishy volatile organic compounds (VOCs) and trimethylamine (TMA-N). Further studies demonstrated that the reduction of VOCs was mainly due to the inhibition of lipids oxidation based on the antioxidant activity of carnosine and the inactivation of lipoxygenase by UHP. Moreover, the reduction of TMA-N was mainly attributed to the ability of UHP processing to reduce bacterial load, which also extended the shelf-life of snakehead fillets by ∼ 6 days and retarded the production of TVB-N. Additionally, the combined application allowed a better retention of pH, color, and textural quality of the fillets compared with the control. Therefore, the current combination is a promising technique in fishy off-odor removing and better preservation of snakehead meat.

Abstract:Abstract The protease hydrolysis technology is used to extract the polypeptides from the sea cucumber waste liquid. The enzymatic hydrolysis ability of protein in sea cucumber waste liquid by seven proteases shows that the best protease is compound protease by taking the hydrolysis degree as the index. Furthermore, the effects of enzyme dosage, pH, temperature and time of enzymatic hydrolysis on the hydrolysis degree are studied, and Box-Benhnken central composite test and response surface analysis method are used to optimize the enzymatic hydrolysis conditions.. The results show that the optimal enzymatic hydrolysis conditions are: enzyme dosage: 647 U/g; pH: 6.48; temperature: 52.5°C; enzymatic hydrolysis time: 3.44h; protein hydrolysis degree under the best condition: 25.9%. The polypeptides extracted from the sea cucumber waste liquid have high antioxidant activity, and the DPPH· scavenging activity increases with the polypeptide concentration.

Abstract:To address the challenges associated with resource inefficiency, low extraction rates, environmental concerns, and high energy consumption in traditional fish oil production from dotted gizzard shad (Konosirus punctatus), a novel approach is needed. This study aimed to develop and evaluate two innovative methods for fish oil extraction and refinement, focusing on their effects on fish oil quality, fatty acid profile, and volatile compound composition throughout the respective processes. The findings of the study revealed that the ethanol-assisted enzymatic extraction method surpassed the conventional enzymatic approach in extraction efficiency, achieving an optimal extraction rate of 74.94% ± 0.45% under optimized process conditions. Moreover, the ethanol-NaOH one-step degumming and deacidification method proved effective in simultaneously removing phospholipids and free fatty acids. Under optimal conditions, a notable reduction in phospholipid content in dotted gizzard shad oil, from 6.80 ± 0.01 mg/g to 1.18 ± 0.01 mg/g, and a substantial decrease in acid value, from 3.31 mg/g to 0.31 mg/g, were observed. In summary, the study analyzed the physicochemical properties, fatty acid composition, and volatile components of fish oil before and after refinement. The refining process was found to preserve the fatty acid composition while efficiently eliminating hydroperoxides and reducing unpleasant odors in the crude oil.
Combining phospholipase A1 with monoacylglycerol lipase for crude vegetable oil degumming through improved oil-water separation

Abstract:Degumming is essential to eliminate undesired phospholipids from crude vegetable oils before being processed into high-quality products. The degumming efficiency is hampered by the separation of oil and water. To overcome this problem, we introduced an innovative enzymatic degumming strategy for various vegetable oils (rice bran, peanut and flaxseed oil) by combining phospholipase A1 with monoacylglycerol lipase. Compared to using phospholipase A1, the combination of the two enzymes significantly reduced phosphorus content, indicating better hydrolysis of phospholipids. Additionally, the primary hydrolysates by the combined enzymes were glycerylphosphorylcholine and glycerylphosphorylethanolamine that were more affinitive to water to improve oil-water separation and oil yield. Characteristic markers obtained from Partial Least Squares Discriminant Analysis model confirmed the advantages of combined enzymes over single enzyme. This advantage of combined enzymes was further validated in a simulated degumming process. Our findings provide a new enzymatic degumming strategy to expand the toolkit for crude oil refinery.
Optimization of fish oil extraction from Lophius litulon liver and fatty acid composition analysis

Abstract:The Lophius litulon liver was used as raw material for the extraction of fish oil via various extraction methods. The extraction rate by water extraction, potassium hydroxide (KOH) hydrolysis and protease hydrolysis were compared and the results revealed the protease hydrolysis extraction had a higher extraction rate with good protein-lipid separation as observed by optical microscope. Furthermore, subsequent experiments determined neutrase to be the best hydrolytic enzyme in terms of extraction rate and cost. The extraction conditions of neutrase hydrolysis were optimized by single-factor experiment and response surface analysis, and the optimal extraction rate was 58.40 ± 0.25% with the following conditions: enzyme concentration 2,000 IU/g, extraction time 1.0 h, liquid-solid ratio 1.95:1, extraction temperature 40.5°C and pH 6.5. The fatty acids composition in fish oil from optimized extraction condition was composed of 19.75% saturated fatty acids and 80.25% unsaturated fatty acids. The content of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) were 8.06% and 1.19%, respectively, with the ratio (6.77:1) surpassed to the recommendation in current researches (5:1). The results in this study suggest protease treatment is an efficient method for high-quality fish oil extraction from Lophius litulon liver with a satisfactory ratio of DHA and EPA.

Abstract:This study pertains to the selection of the appropriate process parameters of lipid extraction aimed at the valorization of sea-bass gutting and filleting side stream biomass. A comparative kinetic study between an environmentally friendly polar (ethanol) and a conventional non-polar solvent (hexane) was implemented to determine the effect of temperature, solvent to solid ratio and time, providing insights in terms of the yield recovery and quality of omega-3 fatty acids. Maximum recovery yield (43.8 and 34.0 g/100 g of dried gutting and filleting by-products, respectively) was achieved within 10 min. In gutting by-products, ethanol achieved full lipid recovery at 35 °C and a 50:1 ratio, while for hexane, the maximum yield was 83%. In the case of filleting side-streams, hexane achieved exhaustive recovery at a 50:1 ratio and 20 °C compared with ethanol, where a higher temperature (50 °C) was needed for exhaustive recovery to be achieved. Differences were attributed to the diverse fatty acid profiles among the two by-products, with 27.3% and 40.8% polyunsaturated fatty acids in viscera and fish frames, respectively. All extracts at 20 °C and 35 °C remained below the oxidation limit set by the Codex Alimentarius Commission (peroxide value < 5 meqO2/kgoil; p-anisidine value < 20). Therefore, green solvents could assist conventional extraction, combining high efficiency and an improved environmental impact.

Abstract:Catfish (Pangasius hypophthalmus) is one of the main seafood products of Vietnam with a large amount of over 1.3 million tons in 2018. However, after the processing, large amount of the harvested catfish is considered as by-products. In this study, catfish by-products have been used to produce three bioactive components, including fish protein hydrolysate (FPH), hydroxyapatite (HA) and lipid fraction by an enzymatic method. The fish by-products were hydrolyzed by using an Alcalase-substrate ratio of 0.4% (v/w) at 60 °C and natural pH for 9 h with a water-raw material ratio of 1:1 (mL/g). The results show that the protein hydrolysate has degree of hydrolysis (DH) of over 30%, nitrogen recovery (NR) of over 80% and contained a large amount of essential amino acids. HA particles have a size of 50–70 nm with high porosity and quite uniformity, suggesting its potential applications such as bone scaffold or regenerative materials. The lipid fraction has high vitamin A content with a small amount of fatty acids (ca. 90 mg/g) that can be refined for further food applications. This recovery method is simple, environmentally-friendly and can be scale-up easily. In addition, the cheap fish by-products were processed to give value-added products as well as reduce the undesirable environmental impact.Graphic Abstract
