Abstract:Introduction: Herbal preparations are extensively utilised for the treatment of diseases in Asian countries. However, the variations in origin, climate, and production processes can lead to inconsistencies in the quality of herbal preparations. Existing quality control methods only target a few components in the finished product but ignore the control in the pharmaceutical process. Therefore, this study intends to develop a comprehensive component analysis method for intermediates in the pharmaceutical process to reveal the change patterns of substances and deepen the process understanding. Objective: This study aims to develop a rapid and comprehensive process characterisation and critical process identification method for herbal preparations. Methods: Six batches of Trichosanthis Pericarpium injection (TPI) intermediates were collected from the production process. Proton nuclear magnetic resonance (H-1-NMR) spectra were acquired for qualitative and quantitative analysis of the se intermediates. Subsequently, chemometrics were used to identify critical processes and potential chemical markers. Results: A total of 39 components in intermediates were identified, and the transfer of 25 components during the production process was investigated. Column chromatography was determined as the critical process. Nine components were identified as chemical markers. Conclusion: The application of H-1-NMR facilitated a comprehensive reflection of the chemical composition information of process intermediates, enabling investigations into the transfer of multi-component substances and accurate identification of critical processes and chemical markers.
[The Effect of Preparation Method on the Quality of Shen-Mai Injection].

Abstract:OBJECTIVE:To investigate the effect of different preparation method on the quality of Shen-mai injection.METHOD:The Shen-mai injection samples were prepared using three different methods. Fingerprints of Shen-mai extracts red ginseng, and its intermediates were obtained using an HPLC analytical procedure. The contents of ginsenoside Rg1, Rc and Rb1, and the gross saponins of Shen-mai extract were quantitatively mensured with HPLC procedures.RESULT:There was significant difference in fingerprints and chemical contents of the injections prepared by the three different methods.CONCLUSION:The quality of Shen-mai injection was greatly influenced by the preparation method. HPLC fingerprinting method can be applied for the determination of the Shen-mai preparations.

Abstract:Xiaochaihu (XCH) is a classic Chinese medicine formula. XCH tablet, XCH granule, XCH capsule, and XCH effervescent tablet are included in the Chinese Pharmacopoeia. In this review, the formula and quality standards of XCH preparations at home and abroad were compared. The differences in manufacturing process of XCH preparations are discussed. The progress of research on the qualitative identification, quantitative detection and fingerprint chromatogram/specific chromatogram of XCH preparations was reviewed. The characteristic components of Pinelliae Rhizoma Praeparatum Cum Zingibere Et Alumine and Jujubae Fructus was rarely analyzed for XCH preparations. It is suggested that the specificity of drug quality detection methods should be improved. Considering drug safety and drug efficacy, it is suggested to set the upper and lower limits of the content of saikosaponins. The standards for heavy metals and other limited items for XCH preparations are also suggested to be set.

Abstract:A simple and reliable method of high-performance liquid chromatography with diode array detection method was developed for fingerprint analysis and simultaneous determination of six compounds including puerarin, salvianolic acid B, berberine hydrochloride, palmatine chloride, dehydrocorydaline, and icariin in the Chinese medicine preparation Yangxinshi Pian. The separation was performed on an Agilent Eclipse XDB-C18 reserved-phase column (250 mm × 4.6mm I.D., 5 μm) using gradient elution with 50 mmol/L monopotassium phosphate aqueous solution and methanol as mobile phase at a flow rate of 1.0 mL/min. The column operating temperature was set at 30°C, and the detection wavelength was 280 nm. The method was validated by linearity, precision, accuracy, stability, and recovery. For fingerprint analysis, 25 peaks were selected as the common peaks, and four kinds of similarities including cosine similarity (S), ratio of similarity (S'), projection content similarity (C), and content similarity (P) were applied to evaluate the quality consistency of different batches of Yangxinshi Pian. The results showed that the developed method was an efficient tool for quality evaluation of Yangxinshi Pian.

Abstract:Shenmai Injection is a Chinese medicinal injection prepared from Ginseng Radix et Rhizoma Rubra and Ophiopogonis Radix, which is widely used in clinical practice for the treatment and adjuvant therapy of cardiovascular diseases with significant pharmacological effects. Proton nuclear magnetic resonance spectroscopy(~1H-NMR) has the advantages of simple and nondestructive sample pretreatment, fast analysis, abundant chemical information, quantification and no need to follow the standard curve. It is widely used in the analysis and research of complex mixtures of traditional Chinese medicine, clinical blood and urine samples. In this study, the ~1H-NMR fingerprint of Shenmai Injection was established. Thirty-two chemical components were identified, including seven amino acids, eight small molecular organic acids, one alkaloid, four sugars, two nucleosides, seven saponins, and three other components. Pearson's correlation coefficient and multivariate analysis of variance(principal component analysis combined with hierarchical cluster analysis) were applied based on the ~1H-NMR fingerprint to evaluate the quality consistency. The results showed high-quality consistency of 82 batches of Shenmai Injection. This study confirms that the ~1H-NMR fingerprint has great potential in the application of quality control of Chinese medicinal injection.

Abstract:Very few efforts have been directed toward interpreting the differences between chromatographic and spectroscopic fingerprints for the purpose of quality control, although similarity analysis based on fingerprints is commonly applied to quality consistency evaluation of herbal medicines (HM). In the current study, comparative evaluation of high-performance liquid chromatography (HPLC), ultraviolet (UV), and near-infrared (NIR) fingerprint techniques combined with chemometric methods has been conducted to control the quality of Chinese herbal injections. A set of Danshen injections (DSI) are chosen as an example of the relevance to contemporary HM. Similarity analysis by principal component analysis, identification using linear discriminant analysis and k nearest neighbor classifiers, and control charts based on Hotelling's T-2 and DModX statistics are employed to evaluate the quality consistency of the DSI samples from three different manufacturers. The results in this study demonstrate that the UV spectroscopy technique combined with chemometric methods is useful for the identity and consistency evaluation of DSI. Such an approach is believed to be equally applicable to other HM.
Quantitative Profiling of Comprehensive Composition in Compound Herbal Injections: an NMR Approach Applied on Shenmai Injection

Abstract:INTRODUCTION:Compound herbal injections (CHIs) can be regarded as a significant innovation in the modernisation of herbal medicine. Therefore, improving the quality control level of CHIs has always been an active research topic in traditional herbal medicine.OBJECTIVES:In this study, Shenmai injection was used as a representative sample for investigating the ability of proton nuclear magnetic resonance (1 H NMR) in the quality evaluation of CHIs.METHODS:A quantitative 1 H NMR method was developed to simultaneously determine the contents of total ginsenosides, polysorbate 80, and 20 primary metabolites in Shenmai injection. Multivariate statistical analysis was combined to compare differences between samples from different manufacturers.RESULTS:It was found that the combined measurement uncertainty of each component is less than 1.61%, which demonstrates the reliability of the method. Furthermore, the components determined by this method account for up to 92.64% of the total solids, which is an unprecedented success in the analysis of Shenmai injection. In the end, the method was applied to the quality comparison of Shenmai injection from six manufacturers. The results showed that the differences among the samples from the six manufacturers were reflected in multiple types of components.CONCLUSION:This study fully demonstrates the superiority of the quantitative 1 H NMR method in comprehensive composition profiling of CHIs, which is conducive to improving the quality control level of Shenmai injection. Further, the present study can be used as a reference study for the research on the quality and safety of CHIs.
Fingerprints of Qingkailing Injection by HPLC/UV/MS/MS

Abstract:AIM: To develop a method of the HPLC/UV/MS/MS fingerprints of Qingkailing Injection(Radix Isatidis,Fructus Gardeniae,Flos Lonicerae Japonicae,etc.) and provide a novel conception for its quality control. METHODS: Separation was performed on Phenomenex Luna C_(18) analytical column(250 mm×4.6 mm,5 μm).The mobile phases consisted of water containing 0.1% formic acid and acetonitrile was used as gradient elute.The flow rate was 1 mL/min.An Electrospray Ion Trap mass spectrometer was utilized for qualitative analysis and both positive and negative secondary ion scan mode were applied. RESULTS: Perfect fingerprints with excellent separation and reproducibility were obtained and 13 compounds were identified.Especially three isomers,including Neochlorogenic acid,Chlorogenic acid and Isochlorogenic acid,were differentiated successfully. CONCLUSION: The method is suitable for establishing the HPLC/MS/MS fingerprints of Qingkailing Injection and hence could be used as an effective and credible pattern for analyzing traditional Chinese medicine as well as its quality control.Moreover,based on this approch,further traditional Chinese medicine research can be performed.

Abstract:Chromatographic fingerprinting has been recommended as a potential and reliable strategy for the quality control of herbal medicines. Although varieties of chromatographic techniques, particularly HPLC, have been widely employed, hyphenated chromatographic approach has not been sufficiently exploited in chromatographic fingerprinting. In this work, LC/MS fingerprinting of Shenmai injection was developed. Thirty ginsenosides as well as seven ophioponins were selected to construct the LC/MS fingerprint using selective ion monitoring (SIM) mode, while previous HPLC fingerprint [H.J. Zhang, Y.J. Wu, Y.Y. Cheng, J. Pharm. Biomed. Anal. 31 (2003) 175–183] only represents the ginsenosides. Subsequently, the proposed LC/MS fingerprints were applied to identifying the product manufacturers. All the samples were accurately classified based on their LC/MS fingerprints in conjunction with principal components analysis (PCA). This study would be potentially helpful to improve the quality control ability of fingerprinting-based strategy for complex herbal medicines.

Abstract:Full-quantified fingerprint technique combines the fingerprint technique and multiple target determination technique. The development of full-quantified fingerprint technique includes the preparation of full-quantified fraction, the quality control in preparation process with fingerprint technique and determination of products. In this paper, the full-quantified fractions of Salvia miltiorrhiza Bge. were prepared by water extraction, deposition with the ethanol, membrane filter, separation on macroporous resin and separation with preparative high performance liquid chromatography (HPLC). Fingerprint technique was used to test the reproducibility of preparation. Three ingredients, protocatechualdehyde, rosmarinic acid, salvianolic acid B were determined and the sum of their contents was more than 50%. The pharmaceutical active compounds were taken as the preparation target. The impurities were removed effectively by multiple preparations, and the specification of full-quantified fraction was improved greatly.

Abstract:Background: Xiaochaihu capsule is composed of seven traditional Chinese medicines. The pharmacopoeia only focuses on the quantitative detection of baicalin, which cannot fully reflect the quality of the preparation. Some medium polar components were used to establish the fingerprint of Xiaochaihu capsule, but there was no report on the strong polar components. Methods: A high performance liquid chromatography-corona charged aerosol detection technology was used to establish a fingerprint analysis method for Xiaochaihu capsules following an analytical quality by design approach. Definitive screening designed experiments were used to optimize the method parameters. A stepwise regression method was used to build quantitative models. The method operable design region was calculated using the experimental error simulation method. Plackett–Burman designed experiments were carried out to test robustness. Results: The contents of four components were simultaneously determined. There were seven common peaks in the fingerprint. The common peak area accounted for 91.72%. Both fingerprint and quantitative analysis methods were validated as applicable in the methodology study. The quantitative fingerprint analysis method for sugar components can fill the gap in the detection of strong polar components in the existing methods. It provides a new technology for the comprehensive overall evaluation of Xiaochaihu capsule.

Abstract:Qualitative and quantitative analysis of Shuanghuanglian fenzhenji was studied in the present paper by FTIR. The fingerprint characteristics of main components in the injections were assigned and can be used as the criterion in the qualitative quality control. Calibration models by PLS algorithm were built to get the quantitative information of baicalin and chlorogenic acid in the injection samples. The R2 of calibration models are both above 0.99 and the average relative deviations between predicted and actual value measured by HPLC are less than 4% for the both properties. The qualitative and quantitative analysis by FTIR is simple and rapid, so it may provide a new on-line approach to the quality control of main constituent in Shuanghuanglian fenzhenji and some other traditional Chinese medicines.

Abstract:OBJECTIVES:This study aimed to establish a rapid and comprehensive method for quantitative determination of complex ingredients in Traditional Chinese Medicine injections.METHODS:A 1H quantitative nuclear magnetic resonance method was developed to simultaneously quantify comprehensive chemical components in Danshen Injection. Multivariate statistical analysis technique was applied to quality evaluation of multiple batches of Danshen injection.KEY FINDINGS:A complete signal attribution to the 1H nuclear magnetic resonance spectrum of Danshen injection was developed and performed for the first time. A total of 32 chemical components were identified from Danshen Injection. Among them, 20 were quantified simultaneously, accounting for up to 80% (w/w) of the total solids and 95% (w/w) of total organic matter, representing success compared to the previous studies. The developed method was further applied to analyze 13 batches of Danshen Injection from three manufacturers to make a realistic analysis.CONCLUSION:It was found that the comprehensive chemical information provides an adequate characterization for quality profiles among different commercial batches of Danshen Injection. The developed method further offered a guarantee for improving the consistency and safety of Traditional Chinese Medicine injections.
[The Study on Fingerprint of Compound Ginseng Injection by Powder X-ray Diffraction].

Abstract:OBJECTIVE:To establish the fingerprint of "Compound Ginseng Injection", including Ginseng and Pericarpium Citri Reticulatae (Chenpi) raw material, their intermediate and injection by powder X-ray diffraction.METHOD:Powder X-ray diffraction was used to analyze Ginseng and Chenpi raw material, their intermediate and injection, contrasted and analyzed the graphs to establish the fingerprint.RESULT:The fingerprint of Ginseng and Chenpi raw material, their intermediate and injection was established. Furthermore, the fingerprint of them showed an excellent recurrence and stability.CONCLUSION:Powder X-ray diffraction method can be used to control the quality of "Compound Ginseng Injection".

Abstract:To establish a method for quality control of the preparation process of Shenkang granules by means of fingerprint technique combined with quantitative determination of index components.According to the preparation process,the multi-wavelength fusion and similarity evaluation were carried out on the fingerprints of herbs,the intermediates (decoction,alcohol solution,water sink) and Shenkang granules,then quantitative analysis of the index group.The fusion fingerprints of rhubarb and salvia miltiorrhiza,the intermediates and granules had a total of 21 common peaks,the fusion fingerprints of safflower and radix astragali,the intermediates and granules had 27 common peaks.The index components can be detected in all samples.The similarity of the fusion fingerprints of the intermediates and granules was more than 0.89,indicating that the material remained well during the preparation process.The results of the content determination of the index group can characterize the quantitative transfer of material during the process.The comprehensive evaluation method based on the combination of fingerprint technique and multi-index components can effectively control the production process from medicine to preparation,and provide the guarantee for the quality control of Shenkang granules production.

Abstract:A simple, reliable method for simultaneous determination of the ten components in Danhong injection (DHI) in combination of chromatographic fingerprint analysis was developed by high performance liquid chromatography coupled with diode-array-detector (HPLC-DAD). The separation was performed on an Agilent Zorbax SB-C18 column with a linear gradient elution of acetonitrile and 0.5% formic acid. The method was validated by linearity, precision, stability and recovery. The developed method was subsequently applied to evaluate 15 batches of DHI and testified to be suitable for its quality control.
A Novel and Practical Chromatographic “Fingerprint-ROC-SVM” Strategy Applied to Quality Analysis of Traditional Chinese Medicine Injections: Using KuDieZi Injection as a Case Study

Abstract:Fingerprinting is widely and commonly used in the quality control of traditional Chinese medicine (TCM) injections. However, current studies informed that the fingerprint similarity evaluation was less sensitive and easily generated false positive results. For this reason, a novel and practical chromatographic "Fingerprint-ROC-SVM" strategy was established by using KuDieZi (KDZ) injection as a case study in the present article. Firstly, the chromatographic fingerprints of KDZ injection were obtained by UPLC and the common characteristic peaks were identified with UPLC/Q-TOF-MS under the same chromatographic conditions. Then, the receiver operating characteristic (ROC) curve was used to optimize common characteristic peaks by the AUCs value greater than 0.7. Finally, a support vector machine (SVM) model, with the accuracy of 97.06%, was established by the optimized characteristic peaks and applied to monitor the quality of KDZ injection. As a result, the established model could sensitively and accurately distinguish the qualified products (QPs) with the unqualified products (UPs), high-temperature processed samples (HTPs) and high-illumination processed samples (HIPs) of KDZ injection, and the prediction accuracy was 100.00%, 93.75% and 100.00%, respectively. Furthermore, through the comparison with other chemometrics methods, the superiority of the novel analytical strategy was more prominent. It indicated that the novel and practical chromatographic "Fingerprint-ROC-SVM" strategy could be further applied to facilitate the development of the quality analysis of TCM injections.

Abstract:Guanxinning injection (GXNI) is a traditional Chinese medicine preparation derived from Salviae Miltiorrhizae Radix et Rhizoma and Chuanxiong Rhizoma. It is produced through a series of processes involving water extraction, ethanol precipitation, and other techniques. GXNI is primarily used for treating angina pectoris in coronary heart disease, and it has shown remarkable efficacy in clinical practice. One of the key components responsible for its pharmacological effects is salvianolic acids. However, these components are known for their poor stability and susceptibility to pH and temperature variations. Therefore, it is crucial to focus on ensuring the quality and stability of salvianolic acids in GXNI. In this study, we employed Proton nuclear magnetic resonance (1H NMR) to analyze the intermediate products formed during the manufacturing of GXNI. We thoroughly examined the spectra of these intermediates and developed a precise 1H-qNMR method for the accurate quantification of various classes of chemical components present in GXNI. Additionally, we applied this method to investigate how the composition of GXNI evolves and transfers throughout the entire production process. To further enhance our understanding, we employed Principal Component Analysis (PCA) and Orthogonal Partial Least Squares Discriminant Analysis (OPLS-DA) to identify critical steps in the production process and to identify potential quality markers (Q-markers) associated with these processes. Our study sheds light on the dynamic changes in composition that occur during the production of GXNI. Quantitative Chromatographic Fingerprint Analysis of Sanye Tangzhiqing Decoction Based on Quality by Design Concept

Abstract:In this work,a high performance liquid chromatography-ultraviolet( HPLC-UV) detection technology was used to establish fingerprint analysis method for Sanye Tangzhiqing Decoction following an analytical quality by design( AQb D) approach. Firstly,column temperature,flow rate,and gradient elution conditions were determined as the method parameters needing to be optimized. Then according to the results of definitive screening design,three critical method attributes( CMAs) were identified,including peak number,the percentage of common peak area to total peak area,and retention time of the last peak. A stepwise regression method was used then to build quantitative models between CMAs and method parameters. Probability-based design space was calculated and successfully verified using the experimental error simulation method. After the analysis conditions were optimized,the contents of six components,namely chlorogenic acid,paeoniflorin,rutin,hyperoside,quercetin-3-O-β-D-glucuronide,and salvianolic acid B were simultaneously determined. There were 19 common peaks in the fingerprint and their common peak area accounted for 96% of the total peak area. Both fingerprint and quantitative analysis methods were validated applicable in methodology study,and they can be applied to determine new samples.
