Abstract:Mitochondria are important organelles in eukaryotes,entire proteins in the mitochondria are called mitochondrial proteome.Recently human mitochondrial proteome was identified that consisted of 1500 proteins which were encoded by nuclear and mitochondrial DNA.Mitochondria are central to cellular energetic metabolism and material metabolism,while mitochondrial dysfunction will directly or indirectly leads to large numbers of diseases.However the mitochondrial proteome systematically analyses of the organelle in physiology and pathogenesis,also discovers the genetic basis of diseases.In the review,we introduce the methods and recent progress in the field of mitochondrial proteome;then we elaborate properties of the mitochondrial proteome and explore future prospects and challenges for using the proteome for systems analysis of the mitochondria.

Abstract:Mitochondrial DNA(mtDNA) is organized in solid particles with a series of proteins,and these nucleoprotein complexes are named as mitochondrial nucleoids because of the prokaryotic origin of mitochondria. Proteins in mitochondrial nucleoids consist of mitochondrial transcription factors,mitochondrial single stranded-DNA binding proteins and multiple functional proteins which catalyze the metabolic reactions in mitochondria.The elucidation of mitochondrial nucleoid structure is meaningful to interpret the mechanisms of the maintenance and inheritance of mitochondrial genome,and in revealing the potential regulation of mitochondrial gene expression.This review focuses on the current opinions in the components,structure and functions of mitochondrial nucleoid, especially how these proteins coordinate the mtDNA and mitochondrial metabolisms like TCA cycle and amino acid metabolism.Meanwhile,abnormality in nucleoid-associated proteins leads to many human diseases.The research in mitochondrial nucleoids is helpful to develop new therapeutics.

Abstract:Iron is a trace element necessary for cell growth, development, and cellular homeostasis, but insufficient or excessive level of iron is toxic. Intracellularly, sufficient amounts of iron are required for mitochondria (the center of iron utilization) to maintain their normal physiologic function. Iron deficiency impairs mitochondrial metabolism and respiratory activity, while mitochondrial iron overload promotes ROS production during mitochondrial electron transport, thus promoting potential disease development. This review provides an overview of iron homeostasis, mitochondrial iron metabolism, and how mitochondrial iron imbalances-induced mitochondrial dysfunction contribute to diseases.

Abstract:Mitochondria are membrane-enclosed organelles found in eukaryotic cells to supply adenosine triphosphate(ATP) as the energy molecules.Mitochondrial DNA(mtDNA) is a circular molecule that codes 22 transfer RNAs,two ribosomal RNAs and 13 proteins.Mitochondrial tRNA is different from cytoplasmic tRNA and does not have a typical clover structure.The high-frequency mutations in mitochondrial tRNAs may cause mitochondrial dysfunction.Several nuclear factors,including processing endonuclease,tRNA modification enzymes and aminoacyl-tRNA synthetase,are involved in the biosynthesis and function of mitochondrial tRNA.Abnormalities of these nuclear factors associated with pathogenic mitochondrial tRNA mutations lead to diseases.

Abstract:Mitochondria are one of the most important organelles for eukaryotes, including humans, to produce energy. In the energy-producing process, mitochondria constantly generate reactive oxygen species as a by-product of electrons leaking out from the electron transport chain react with oxygen. The active oxygen, in turn, plays pivotal roles in mediating several signalings, including those that are implicated in the development of some diseases such as neurodegenerative disease, cardiovascular disease, and carcinogenesis. This signaling, derived from mitochondrial reactive oxygen species, also affects intracellular iron homeostasis by regulating the expression of transporters. Heme iron is incorporated into cells through HCP1, and non-heme iron is transported by DMT1 in absorptive cells. Intracellular iron is exported by ferroportin and bound with transferrin. In most types of cell including erythrocyte, transferrin-bound iron is incorporated through transferrin-transferrin receptor system. We previously reported that the expression of HCP1 and DMT1 was upregulated in cancer cells and that overexpression of manganese superoxide dismutase, which is a mitochondrial-specific superoxide dismutase, downregulated the expression. These findings indicate that mitochondrial reactive oxygen species is associated with iron-related oxidative reactions. Recently, a mitochondria-specific iron transporter, mitoferrin, was identified, and the relationships among mitochondria, iron transportation, and diseases have been increasingly clarified.

Abstract:Iron is essential in various life processes, and its imbalance is closely related to disorders in multiple systems such as the nervous, cardiovascular, and immune systems, as well as many chronic diseases such as infections and cancer. Therefore, accurately assessing the status of iron in the body is of great significance for both basic and clinical research. The gold standard for iron status is still bone marrow stainable iron, but its actual application is very limited due to its invasive sampling and high cost. Other traditional biomarkers such as serum iron, ferritin, and related composite indicators are all affected to varying degrees by factors such as inflammation, age, and circadian fluctuations, but they also have their own suitable application scenarios. However, biomarkers that can fully assess iron status are far from perfect, leading to inaccurate diagnosis and treatment of iron imbalance in many cases. Although new biomarkers such as hepcidin, reticulocyte hemoglobin content, and erythroferrone have emerged, and with the rapid advancement of high-throughput technologies, multi-omics may help diagnose iron status in the future, more exploration and mining are still needed to expand the widespread and rational application of these potential markers. This article reviews the research progress of iron biomarkers, aiming to provide more references and directions for future related research.

Abstract:Iron is a trace metal element necessary to maintain life and is also involved in a variety of biological processes. Aging refers to the natural life process in which the physiological functions of the various systems, organs, and tissues decline, affected by genetic and environmental factors. Therefore, it is imperative to investigate the relationship between iron metabolism and aging-related diseases, including neurodegenerative diseases. During aging, the accumulation of nonheme iron destroys the stability of the intracellular environment. The destruction of iron homeostasis can induce cell damage by producing hydroxyl free radicals, leading to mitochondrial dysfunction, brain aging, and even organismal aging. In this review, we have briefly summarized the role of the metabolic process of iron in the body, then discussed recent developments of iron metabolism in aging and age-related neurodegenerative diseases, and finally, explored some iron chelators as treatment strategies for those disorders. Understanding the roles of iron metabolism in aging and neurodegenerative diseases will fill the knowledge gap in the field. This review could provide new insights into the research on iron metabolism and age-related neurodegenerative diseases.

Abstract:As a cofactor, iron-sulfur (Fe-S) cluster binds to proteins or enzymes that play important roles in various important biological processes, including DNA synthesis and repair, mitochondrial function, gene transcription and translation. In mammals, the core components involved in Fe-S cluster biosynthesis are considered to include the scaffold protein ISCU, cysteine desulfurase NFS1 and its accessory proteins ISD11 and ACP, and frataxin (FXN). Proteins involved in Fe-S cluster transfer have been found to include HSC20/HSPA9, as chaperone system, and Fe-S cluster carriers. The biosynthesis and transfer of Fe-S clusters to Fe-S recipients require fine-tune regulation. Recently, significant progress has been made in the structure and mechanism of mitochondrial Fe-S biosynthesis and transfer. Based on, especially, the development of DNA sequencing technology, bioinformatics, and gene editing technology, diseases caused by mutations of Fe-S cluster-related genes have been revealed in recent years, promoting the rapid development in the field of Fe-S and human health. This review focuses on the function of genes involved in Fe-S cluster biosynthesis and transfer and on the diseases caused by the mutations of the related genes. Finally, some questions we are facing are raised, new hypotheses presented, and the perspectives discussed.

Abstract:Iron, an essential mineral in the body, is involved in numerous physiological processes, making the maintenance of iron homeostasis crucial for overall health. Both iron overload and deficiency can cause various disorders and human diseases. Ferroptosis, a form of cell death dependent on iron, is characterized by the extensive peroxidation of lipids. Unlike other kinds of classical unprogrammed cell death, ferroptosis is primarily linked to disruptions in iron metabolism, lipid peroxidation, and antioxidant system imbalance. Ferroptosis is regulated through transcription, translation, and post-translational modifications, which affect cellular sensitivity to ferroptosis. Over the past decade or so, numerous diseases have been linked to ferroptosis as part of their etiology, including cancers, metabolic disorders, autoimmune diseases, central nervous system diseases, cardiovascular diseases, and musculoskeletal diseases. Ferroptosis-related proteins have become attractive targets for many major human diseases that are currently incurable, and some ferroptosis regulators have shown therapeutic effects in clinical trials although further validation of their clinical potential is needed. Therefore, in-depth analysis of ferroptosis and its potential molecular mechanisms in human diseases may offer additional strategies for clinical prevention and treatment. In this review, we discuss the physiological significance of iron homeostasis in the body, the potential contribution of ferroptosis to the etiology and development of human diseases, along with the evidence supporting targeting ferroptosis as a therapeutic approach. Importantly, we evaluate recent potential therapeutic targets and promising interventions, providing guidance for future targeted treatment therapies against human diseases.

Current Issues in Molecular Biology

Abstract:Iron is essential for many physiological processes, and the dysregulation of its metabolism is implicated in the pathogenesis of various diseases. Recent advances in iron metabolism research have revealed multiple complex pathways critical for maintaining iron homeostasis. Molecular imaging, an interdisciplinary imaging technique, has shown considerable promise in advancing research on iron metabolism. Here, we comprehensively review the multifaceted roles of iron at the cellular and systemic levels (along with the complex regulatory mechanisms of iron metabolism), elucidate appropriate imaging methods, and summarize their utility and fundamental principles in diagnosing and treating diseases related to iron metabolism. Utilizing molecular imaging technology to deeply understand the complexities of iron metabolism and its critical role in physiological and pathological processes offers new possibilities for early disease diagnosis, treatment monitoring, and the development of novel therapies. Despite technological limitations and the need to ensure the biological relevance and clinical applicability of imaging results, molecular imaging technology’s potential to reveal the iron metabolic process is unparalleled, providing new insights into the link between iron metabolism abnormalities and various diseases.

Abstract:Mitochondria are vital to life and provide biological energy for other organelles and cell physiological processes. On the mitochondrial double layer membrane, there are a variety of channels and transporters to transport different metal ions, such as Ca2+, K+, Na+, Mg2+, Zn2+ and Fe2+/Fe3+. Emerging evidence in recent years has shown that the metal ion transport is essential for mitochondrial function and cellular metabolism, including oxidative phosphorylation (OXPHOS), ATP production, mitochondrial integrity, mitochondrial volume, enzyme activity, signal transduction, proliferation and apoptosis. The homeostasis of mitochondrial metal ions plays an important role in maintaining mitochondria and cell functions and regulating multiple diseases. In particular, channels and transporters for transporting mitochondrial metal ions are very critical, which can be used as potential targets to treat neurodegeneration, cardiovascular diseases, cancer, diabetes and other metabolic diseases. This review summarizes the current research on several types of mitochondrial metal ion channels/transporters and their functions in cell metabolism and diseases, providing strong evidence and therapeutic strategies for further insights into related diseases.

Abstract:Mitochondria are small organelles in cells that can produce ATP to provide almost all the energy needed for cell survival. A number of cellular processes, such as signal transduction, energy metabolism, autophagy and apoptosis, are related to mitochondria, which are crucial for the organisms to maintain normal physiological functions. At the same time, the occurrence and development of a variety of illnesses such as cancers and Alzheimer’s disease are closely associated with the dysfunction of mitochondria. Various proteins and inorganic species are required to accomplish the mitochondrial functions; therefore, to timely know the distribution and variation of these species in mitochondria is extremely important. On the other hand, the mitochondrial function could be intervened or modulated by making use of the unique physical and chemical properties of metal complexes, thereby to prevent or treat mitochondrial diseases. With the detection of mitochondrial inorganic species and the modulation of mitochondrial functions as the theme, this review summarizes some of the recent typical achievements made by Chinese scientists on the subject; in addition, it indicates the problems and challenges existing in the development of this field.

Signal Transduction and Targeted Therapy

Abstract:Mitochondria, with their intricate networks of functions and information processing, are pivotal in both health regulation and disease progression. Particularly, mitochondrial dysfunctions are identified in many common pathologies, including cardiovascular diseases, neurodegeneration, metabolic syndrome, and cancer. However, the multifaceted nature and elusive phenotypic threshold of mitochondrial dysfunction complicate our understanding of their contributions to diseases. Nonetheless, these complexities do not prevent mitochondria from being among the most important therapeutic targets. In recent years, strategies targeting mitochondrial dysfunction have continuously emerged and transitioned to clinical trials. Advanced intervention such as using healthy mitochondria to replenish or replace damaged mitochondria, has shown promise in preclinical trials of various diseases. Mitochondrial components, including mtDNA, mitochondria-located microRNA, and associated proteins can be potential therapeutic agents to augment mitochondrial function in immunometabolic diseases and tissue injuries. Here, we review current knowledge of mitochondrial pathophysiology in concrete examples of common diseases. We also summarize current strategies to treat mitochondrial dysfunction from the perspective of dietary supplements and targeted therapies, as well as the clinical translational situation of related pharmacology agents. Finally, this review discusses the innovations and potential applications of mitochondrial transplantation as an advanced and promising treatment.

Abstract:Trace element iron is essential for nearly all living organisms.It is the key component of ironcontaining enzymes and proteins,which participate in many cellular biological processes.It is estimated that nearly one quarter of population worldwide has been suffered from anemia due to iron deficiency.In contrast,iron overload induces a disease termed as Hemochromatosis,which the incidence is approximately 1/200 in Caucasians. Recently,the disease has also been reported in China.It is fatal if the disease progresses to late stage as the sign of heart,pancreas and liver failures.Therefore,maintenance of iron homeostasis is crucial.It is believed that iron is uptake by small intestine,stored in liver,transported in blood,recycled by macrophages,and finally utilized by cells to fulfill the functions.In last "Golden Decade",many novel iron metabolic genes have been cloned and functionally characterized to further understanding of regulation of iron metabolism and maintenance of iron homeostasis. However,more insights need to be learned considering the complexity of the processes.In this review,we summarize the recent findings in this field and discuss remaining questions,and provide our understanding towards future directions.

Abstract:Iron is indispensable in numerous biologic processes, but abnormal iron regulation and accumulation is related to pathological processes in cardiovascular diseases. However, the underlying mechanisms still need to be further explored. Iron plays a key role in metal-catalyzed oxidative reactions that generate reactive oxygen species (ROS), which can cause oxidative stress. As the center for oxygen and iron utilization, mitochondria are vulnerable to damage from iron-induced oxidative stress and participate in processes involved in iron-related damage in cardiovascular disease, although the mechanism remains unclear. In this review, the pathological roles of iron-related oxidative stress in cardiovascular diseases are summarized, and the potential effects and mechanisms of mitochondrial iron homeostasis and dysfunction in these diseases are especially highlighted.

Abstract:The progression of heart failure (HF) is complex and involves multiple regulatory pathways. Iron ions play a crucial supportive role as a cofactor for important proteins such as hemoglobin, myoglobin, oxidative respiratory chain, and DNA synthetase, in the myocardial energy metabolism process. In recent years, numerous studies have shown that HF is associated with iron dysmetabolism, and deficiencies in iron and overload of iron can both lead to the development of various myocarditis diseases, which ultimately progress to HF. Iron toxicity and iron metabolism may be key targets for the diagnosis, treatment, and prevention of HF. Some iron chelators (such as desferrioxamine), antioxidants (such as ascorbate), Fer-1, and molecules that regulate iron levels (such as lactoferrin) have been shown to be effective in treating HF and protecting the myocardium in multiple studies. Additionally, certain natural compounds can play a significant role by mediating the imbalance of iron-related signaling pathways and expression levels. Therefore, this review not only summarizes the basic processes of iron metabolism in the body and the mechanisms by which they play a role in HF, with the aim of providing new clues and considerations for the treatment of HF, but also summarizes recent studies on natural chemical components that involve ferroptosis and its role in HF pathology, as well as the mechanisms by which naturally occurring products regulate ferroptosis in HF, with the aim of providing reference information for the development of new ferroptosis inhibitors and lead compounds for the treatment of HF in the future.

Abstract:Ferroptosis is a distinctive form of iron-dependent cell death characterized by significant ultrastructural changes in mitochondria. Given the crucial involvement of mitochondria in various cellular processes such as reactive oxygen species production, energy metabolism, redox status, and iron metabolism, mounting evidence suggests a vital role of mitochondria in the regulation and execution of ferroptosis. Furthermore, there exists a strong correlation between ferroptosis and various diseases. In this review, we aim to summarize the mechanisms underlying the induction and defense of ferroptosis, emphasizing the influence of mitochondria on this intricate process. Additionally, we provide an overview of the role of ferroptosis in disease, particularly cancer, and elucidate the mechanisms by which drugs targeting mitochondria impact ferroptosis. By presenting a theoretical foundation and reference point, this review aims to contribute to both basic cell biology research and the investigation of clinically relevant diseases.

Abstract:Copper ions serve as co-factors for various enzymes and participate in multiple cellular processes. Mitochondria are essential copper reservoirs within the cell. Previous reviews have extensively summarized the association between mitochondrial copper homeostasis imbalance and hematologic disorders, cardiomyopathies, and skeletal myopathies. However, there is limited information regarding its association with organ fibrosis. This article outlines the role and mechanism of disrupted mitochondrial copper homeostasis in fibrotic diseases, and systematically elaborates copper absorption and transport, as well as the regulation of copper homeostasis within mitochondria. It focuses on the impacts of mitochondrial copper overload and deficiency on fibrotic diseases, and the application of copper chelators as potential anti-fibrotic therapeutic approaches.
