Abstract:The significance of bionanomotors in nanotechnology is analogous to mechanical motors in daily life. Here the principle and approach for designing and constructing biomimetic nanomotors with continuous single-directional motion are reported. This bionanomotor is composed of a dodecameric protein channel, a six-pRNA ring, and an ATPase hexamer. Based on recent elucidations of the one-way revolving mechanisms of the phi29 double-stranded DNA (dsDNA) motor, various RNA and protein elements are designed and tested by single-molecule imaging and biochemical assays, with which the motor with active components has been constructed. The motor motion direction is controlled by three operation elements: (1) Asymmetrical ATPase with ATP-interacting domains for alternative DNA binding/pushing regulated by an arginine finger in a sequential action manner. The arginine finger bridges two adjacent ATPase subunits into a non-covalent dimer, resulting in an asymmetrical hexameric complex containing one dimer and four monomers. (2) The dsDNA translocation channel as a one-way valve. (3) The hexameric pRNA ring geared with left-/right-handed loops. Assessments of these constructs reveal that one inactive subunit of pRNA/ATPase is sufficient to completely block motor function (defined as K = 1), implying that these components work sequentially based on the principle of binomial distribution and Yang Hui's triangle.

Abstract:The realization that many essential functions of living cells are performed by nanoscale motors consisting of protein complexes has given rise to an intense effort to understand their mechanisms. Considerable progress has been made in the past two years by a combination of biophysical techniques and theoretical analysis. Single-molecule studies have played a spectacular role for a variety of motors including kinesin, myosin, and polymerases. The understanding of F1-ATPase, the smallest biomolecular rotary motor, has made particular progress by the interplay of experimental and theoretical studies; the latter have provided information not available from experiment.

Abstract:Biological molecular motors transform chemical energy into mechanical work by coupling cyclic catalytic reactions to large-scale structural transitions. Mechanical deformation can be surprisingly efficient in realizing such coupling, as demonstrated by the F1FO ATP synthase. Here, we describe a synthetic molecular mechanism that transforms a rotary motion of an asymmetric camshaft into reciprocating large-scale transitions in a surrounding stator orchestrated by mechanical deformation. We design the mechanism using DNA origami, characterize its structure via cryo-electron microscopy, and examine its dynamic behavior using single-particle fluorescence microscopy and molecular dynamics simulations. While the camshaft can rotate inside the stator by diffusion, the stator's mechanics makes the camshaft pause at preferred orientations. By changing the stator's mechanical stiffness, we accelerate or suppress the Brownian rotation, demonstrating an allosteric coupling between the camshaft and the stator. Our mechanism provides a framework for manufacturing artificial nanomachines that function because of coordinated movements of their components.
The Lawnmower: an autonomous, protein-based artificial molecular motor

Abstract:Inspired by biology, great progress has been made in creating artificial molecular motors. However, the dream of harnessing proteins - the building blocks selected by Nature - to design autonomous motors has so far remained elusive. Here we report the synthesis and characterization of the Lawnmower, an autonomous, protein-based artificial molecular motor comprised of a spherical hub decorated with proteases. Its "burnt-bridge" motion is directed by cleavage of a peptide lawn, promoting motion towards unvisited substrate. We find that Lawnmowers exhibit directional motion with average speeds of up to 80 nm/s, comparable to biological motors. By selectively patterning the peptide lawn on microfabricated tracks, we furthermore show that the Lawnmower is capable of track-guided motion. Our work opens an avenue towards nanotechnology applications of artificial protein motors.

Abstract:We report the hierarchical assembly of a chloroplast-derived rotary FoF1-ATPase motor-propelled flasklike pentosan colloidal motor (FPCM) with the ability of the synthesis, storage, and triggered release of biological energy currency ATP. These streamlined and submicrometer-sized hollow flasklike pentosan colloidal motors are prepared by combining a soft-template-based hydrothermal polymerization with a vacuum infusion of chloroplast-derived proteoliposomes containing rotary FoF1-ATPase motors. The generation of proton motive force across the proteoliposomes by injecting an acidic buffer solution promotes the rotation of FoF1-ATPase motors to drive the self-propelled motion of FPCMs, accompanying the inner ATP synthesis and storage. These rotary FoF1-ATPase motor-powered FPCMs exhibit a chemotactic behavior by migrating from their neck opening to their round bottom along a proton gradient of the external environment (negative chemotaxis). Such rotary biomolecular motor-driven flasklike pentosan colloidal motors with ATP synthesis and on-demand release make them promising candidates for engineering novel intelligent nanocarriers to actively regulate cellular metabolism.

Abstract:Nano/micromotors (NMMs) are tiny objects capable of converting energy into mechanical motion. Recently, a wealth of active matter including synthetic colloids, cytoskeletons, bacteria, and cells have been used to construct NMMs. The self-sustained motion of active matter drives NMMs out of equilibrium, giving rise to rich dynamics and patterns. Alongside the spontaneous dynamics, external stimuli such as geometric confinements, light, magnetic field, and chemical potential are also harnessed to control the movements of NMMs, yielding new application paradigms of active matter. Here, we review the recent advances, both experimental and theoretical, in exploring biological NMMs. The unique dynamical features of collective NMMs are focused on, along with some possible applications of these intriguing systems.

Abstract:Significance Single-molecule techniques have elucidated how isolated molecular motors generate piconewton forces with unprecedented detail. However, in diverse biological and synthetic settings, force-generating proteins collectively power nonequilibrium dynamics, including continuous large-scale rearrangements and persistent fluid flows. Characterizing motor-generated forces in these dense and dynamical environments remains a challenge. We assembled a reversible DNA-based force-sensing probe that, by an optical readout, reveals the molecular arrangements and the force loads experienced by kinesin motors. These probes provide insight into motor-generated forces that collectively power the unique dynamics of microtubule-based active nematics, a noteworthy example of an internally driven active matter system. DNA-based force probes can be extended to study forces and stresses in various synthetic systems as well as diverse cellular environments.

Abstract:Cells display a range of mechanical activities enabled by the cytoskeleton, a viscoelastic hydrogel manipulated by motor proteins powered through catalysis. This raises the question of how the acceleration of a chemical reaction can enable the energy released from that reaction to be transduced, and thereby work to be done, by a molecular catalyst. Here we demonstrate the molecular-level transduction of chemical energy to mechanical force in the form of the powered contraction and powered re-expansion of a crosslinked polymer gel driven by the directional rotation of embedded artificial catalysis-driven molecular motors. Continuous 360° rotation of the rotor about the stator of motor-molecules incorporated within the polymeric framework of the gel, twists the polymer chains of the crosslinked network around one another (either clockwise or anti-clockwise, depending on the chirality of the fuelling system). This progressively increases writhe and tightens entanglements, causing macroscopic contraction of the gel to ~70% of its original volume. The limit of contraction corresponds to the stall force of the motor; the point at which, despite catalysis continuing, the untwisting force exerted by the entwined strands balances conformation selection in the motor’s catalytic cycle. Subsequent addition of the opposite enantiomeric fuelling system powers rotation of the motor-molecules of the contracted gel in the reverse direction, unwinding the entanglements and causing the gel to re-expand. Continued powered twisting of the strands in the new direction causes the gel to contract once again. The experimental demonstration of work against a load by a synthetic catalyst, and the mechanism of the transduction of energy by a catalyst through kinetic asymmetry in its acceleration of a fuel-to-waste reaction, informs both the debate surrounding the mechanism of force generation by biological motors and the design principles for artificial molecular nanotechnology.

Abstract:Self-propelled micromotors are emerging as important tools that help us understand the fundamentals of motion at the microscale and the nanoscale. Development of the motors for various biomedical and environmental applications is being pursued. Multiple fabrication methods can be used to construct the geometries of different sizes of motors. Here, we present an overview of appropriate methods of fabrication according to both size and shape requirements and the concept of guiding the catalytic motors within the confines of wall. Micromotors have also been incorporated with biological systems for a new type of fabrication method for bioinspired hybrid motors using three-dimensional (3D) printing technology. The 3D printed hybrid and bioinspired motors can be propelled by using ultrasound or live cells, offering a more biocompatible approach when compared to traditional catalytic motors.

Abstract:Micro-/nanomotors (MNMs) are able to propel themselves in fluids through converting different energies from environment into kinetic energy. Recently, layer-by-layer (LbL) assembly, a versatile assembly approach, has been employed to access MNMs with advantages such as regulated motion, stimuli-response properties, and multifunctionality. In this chapter, we review the recent progress on controlled fabrication, motion control, and biomedical applications of MNMs based on controlled molecular assembly. Through integrating diverse functional building blocks such as nanoparticles, enzymes, and metal shells, MNMs with various structures (e.g., hollow capsules and nanotubes) have been prepared, and the control over the on/off state of the MNM motion has been realized. In addition, we also discuss a special type of MNMs which is derived from the combination of as-assembled biological aggregates and artificial nanostructures. These MNMs can be driven by bubble recoil, irradiation by near-infrared light, and ultrasonic fields. We have also demonstrated the potential applications of these assembled MNMs in biomedical fields such as targeted drug delivery, photothermal therapy, and detoxification.
A synthetic autonomous rotary nanomotor made from and fuelled by DNA

Abstract:DNA nanostructures are made using synthetic DNA strands, the sequences of which are designed such that they will self-assemble into the desired form by hybridization of complementary domains. Various structures and devices have been presented, including DNA tweezers, nanorobots and a range of linear motors such as bipedal walkers. Inspiration for the latter is drawn from naturally occurring molecular motors like kinesin. This paper describes a concept for an autonomous rotary nanomotor made from DNA, which utilizes the well-known and widely-studied phenomenon of toehold-mediated DNA strand displacement. The motor is to be driven by a series of strand displacement reactions, the order of which is controlled by steric constraints arising from the secondary structure of the DNA strands comprising the motor mechanism. The capabilities of DNA motors would be extended significantly if autonomous rotary motion could be achieved. The device has a range of potential applications, including molecular computation and single-molecule manipulation.

Abstract:In recent years, there has been a growing interest in engineering dynamic and autonomous systems with robotic functionalities using biomolecules. Specifically, the ability of molecular motors to convert chemical energy to mechanical forces and the programmability of DNA are regarded as promising components for these systems. However, current systems rely on the manual addition of external stimuli, limiting the potential for autonomous molecular systems. Here, we show that DNA-based cascade reactions can act as a molecular controller that drives the autonomous assembly and disassembly of DNA-functionalized microtubules propelled by kinesins. The DNA controller is designed to produce two different DNA strands that program the interaction between the microtubules. The gliding microtubules integrated with the controller autonomously assemble to bundle-like structures and disassemble into discrete filaments without external stimuli, which is observable by fluorescence microscopy. We believe this approach to be a starting point toward more autonomous behavior of motor protein–based multicomponent systems with robotic functionalities.

Abstract:Biomolecular motor proteins that generate forces by consuming chemical energy obtained from ATP hydrolysis are pivotal for organizing broad cytoskeletal structures in living cells. The control of such cytoskeletal structures benefits programmable protein patterning; however, our current knowledge is limited owing to the underdevelopment of an engineering approach for controlling pattern formation. Here, we demonstrate the tailoring of assembled patterns of microtubules (MTs) driven by kinesin motors by designing the boundary shape in fabricated microwells. We found an MT bundle structure along the microwell wall and a bridging structure perpendicular to the wall. Corroborated by the theory of self-propelled rods, we further showed that the alignment of MTs defined by the boundary shape determined the transition of the assembled patterns, providing a blueprint to reconstruct bridge structures in microchannels. Our findings provide a geometric rule to tailor the self-organization of cytoskeletons and motor proteins for nanotechnological applications.

Abstract:Self-propelled chemically powered synthetic micron and nano-scale motors are being intensively studied because of the wide range of potential applications that exploit their directed motion. This paper considers even smaller Ångström-size synthetic motors. Such very small motors in bulk solution display effects arising from their self-propulsion. Recent experiments have shown that small-molecule catalysts and single enzyme molecules exhibit properties that have been attributed to their chemical activity. Molecular dynamics is used to investigate the properties of very small Ångström-size synthetic chemically powered sphere-dimer motors in a simple atomic-like solvent confined between walls separated by distances of tens of nanometers. Evidence for strong structural ordering of the motors between the walls, which reflects the finite size of solvent molecules and depends on solvent depletion forces, is provided. Dynamical properties, such as average motor velocity, orientational relaxation, and mean square displacement, are anisotropic and depend on the distance from the walls. This research provides information needed for potential applications that use molecular-scale motors in the complex confined geometries encountered in biology and the laboratory.

Abstract:An efficient enzyme-powered micromotor device was fabricated by assembling multiple layers of catalase on the inner surface of a poly(3,4-ethylenedioxythiophene and sodium 4-styrenesulfonate)/Au microtube (PEDOT-PSS/Au). The catalase assembly was achieved by programmed DNA hybridization, which was performed by immobilizing a designed sandwich DNA structure as the sensing unit on the PEDOT-PSS/Au, and then alternately hybridizing with two assisting DNA to bind the enzyme for efficient motor motion. The micromotor device showed unique features of good reproducibility, stability and motion performance. Under optimal conditions, it showed a speed of 420 mu m s(-1) in 2% H2O2 and even 51 mu m s(-1) in 0.25% H2O2. In the presence of target DNA, the sensing unit hybridized with target DNA to release the multi-layer DNA as well as the multi-catalase, resulting in a decrease of the motion speed. By using the speed as a signal, the micromotor device could detect DNA from 10 nM to 1 mu M. The proposed micromotor device along with the cyclic alternate DNA hybridization assembly technique provided a new path to fabricate efficient and versatile micromotors, which would be an exceptional tool for rapid and simple detection of biomolecules.

Abstract:Self-propelled micro/nanomotors (MNMs) have shown great application potential in biomedicine, sensing, environmental remediation, etc. In the past decade, various strategies or technologies have been used to prepare and functionalize MNMs. However, the current preparation strategies of the MNMs were mainly following the pre-designed methods based on specific tasks to introduce expected functional parts on the various micro/nanocarriers, which lacks a universal platform and common features, making it difficult to apply to different application scenarios. Here, we have developed a modular assembly strategy based on host-guest chemistry, which enables the on-demand construction of imaging-trackable nanomotors mounted with suitable driving and imaging modules using a universal nanocarrier platform, according to different application scenarios. These assembled nanomotors exhibit ed enhanced diffusion behavior driven by enzymatic reactions. The loaded imaging functions were used to dynamically trace the swarm motion behavior of assembled nanomotors with corresponding fuel conditions both in vitro and in vivo. The modular assembly strategy endowed with host-guest interaction provides a universal approach to producing multifunctional MNMs in a facile and controllable manner, which paves the way for the future development of MNMs systems with programmable functions.

Abstract:Inspired by the machines that have changed the world through their autonomous functions, scientists have focused their attention on performing similar functions at the micro/nanoscale. The motility of these micro/nanomachines (micro/nanomotors) offers enormous opportunities for cargo delivery, biodetection, and environmental and biomedical applications. Among the various geometries of micro/nanomotors, a tubular shape provides asymmetric inner and outer surfaces, where the inner wall hosts chemical reactions that supply the moving power and the outer wall can be modified for specific chemical and biological functions. This review describes the concept of tubular micro/nanomotors, including their basic principles, fabrication methods and control over their motion. With the assistance of catalytic reactions, tubular micro/nanomotors can generate a powerful thrust force to navigate in complex natural and in vitro environments. Modification of the tubular micro/nanomotors allows the motors to capture, transport, and release selected cargos on demand. In addition, their application in sensing and decontamination benefits from their collection behavior and self-mixing effect. Furthermore, noncatalytic reaction-driven tubular micro/nanomotors, such as redox-based and biohybrid tubular micro/nanomotors, provide new possibilities for practical in vivo applications. The state-of-the-art tubular micro/nanomotors could offer a promising platform for various applications, e.g., lab-on-chip devices and in vivo theranostics.
Fundamentals and applications of enzyme powered micro/nano-motors

Abstract:Micro/nanomotors (MNMs) are miniaturized machines that can convert many kinds of energy into mechanical motion. Over the past decades, a variety of driving mechanisms have been developed, which have greatly extended the application scenarios of MNMs. Enzymes exist in natural organisms which can convert chemical energy into mechanical force. It is an innovative attempt to utilize enzymes as biocatalyst providing driving force for MNMs. The fuels for enzymatic reactions are biofriendly as compared to traditional counterparts, which makes enzyme-powered micro/nanomotors (EMNMs) of great value in biomedical field for their nature of biocompatibility. Until now, EMNMs with various shapes can be propelled by catalase, urease and many others. Also, they can be endowed with multiple functionalities to accomplish on-demand tasks. Herein, combined with the development process of EMNMs, we are committed to present a comprehensive understanding of EMNMs, including their types, propelling principles, and potential applications. In this review, we will introduce single enzyme that can be used as motor, enzyme powered molecule motors and other micro/nano-architectures. The fundamental mechanism of energy conversion process of EMNMs and crucial factors that affect their movement behavior will be discussed. The current progress of proof-of-concept applications of EMNMs will also be elaborated in detail. At last, we will summarize and prospect the opportunities and challenges that EMNMs will face in their future development.
Functional Chemical Motor Coatings for Modular Powering of Self-Propelled Particles

Abstract:Inspired by the locomotion of semiaquatic insects, a variety of surface swimming microrobots propelled by surface tension Marangoni forces have been developed over the years. However, most Marangoni micromotor systems present limitations in their applications due to poor performance, short lifetime, low efficiency, and toxicity. We have developed a functional chemical motor coating consisting of protein microfilms with entrapped fuel to functionalize inactive substrates or particles. This motor material system generates large Marangoni propulsive forces with extremely small amounts of fuel due to a self-regulated fuel release mechanism based on dynamic nanostructural changes in the protein matrix, enhancing the lifetime and efficiency performance over other material systems and motors. These motor functional coatings offer great versatility as they can be coated on a wide array of substrates and materials across length scales, with opportunities as modular power sources for microrobots and small-scale devices. The synergy between the protein motor matrix and the chemical fuel enables the wider design of self-powered surface microrobots without previous limitations in their fabrication and performance, including the new design of hybrid microrobots with protein functional coatings as a modular power source.

As the scale decreases, molecular motors can be helpful as they are not limited by increased viscous resistance. Moreover, molecular motors can move against diffusion gradients and are naturally fitted for nanoscale transportation. Among motor proteins, kinesin has particular potential for lab-on-a-chip applications. It can be used for sorting, concentrating or as a mechanical sensor. When bound to a surface, kinesin motors propel microtubules in random directions, depending on their landing orientation. In order to circumvent this complication, the microtubule motion should be confined or guided. To this end, dielectrophoretically aligned multi-walled-carbon nanotubes (MWCNT) can be employed as nanotracks. In order to control more precisely the spatial repartition of the MWCNTs, a screening method has been implemented and tested. Polygonal patterns have been fabricated with the aim of studying the guiding and the microtubule displacement between MWCNT segments. Microtubules are observed to transfer between MWCNT segments, a prerequisite for the guiding of microtubules in MWCNT circuit-based biodevices. The effect of the MWCNT organization (crenellated or hexagonal) on the MT travel distance has been investigated as well.
