Abstract:[GRAPHICS]A highly convergent total synthesis of the natural products epothilone B and D is described. The route is highlighted by efficient generation of a C12-C13 trisubstituted olefin which exploits a sequential Nozaki-Hiyama-Kishi coupling and a stereoselective thionyl chloride rearrangement.

Abstract:Echinopines A and B [(+)-1 and (+)-2], two naturally occurring compounds characterized with a unique [3.5.5.7] carbon framework, have been synthesized in both enantiomeric and racemic forms. Their total synthesis involves a novel intramolecular rhodium-catalyzed cyclopropanation (4 -> 16) and a samarium diiodide-mediated ring closure (3 -> 37).

Abstract:Ring-closing metathesis (step a in the scheme below), deprotection (b), Stille coupling (c), and epoxidation (d) are the key steps in the first total synthesis of the naturally occurring epothilone E (1). An advanced intermediate, the product of the olefin metathesis, allows access to a range of epothilone E analogues having different aromatic moieties in the side chain. TBS = tBuMe2Si.

Abstract:The total synthesis of epothilone A is described by the coupling four segments 4-7a. Three of the segments, 4, 5 and 7a, have only one chiral center; all other chiral centers were introduced by simple asymmetric catalytic reactions. The key steps are the ring opening of epoxide 5 with acetylide 8 for the construction of the C12-C13 cis double bond and a practical hydrolytic kinetic resolution (HKR) developed by Jacobsen group for the introduction the chiral center at C3. Especially, the stereospecific epoxidation of 3-O-PMB epothilone C 3b through long-range effect of 3-O-PMB protecting group gave high yields of the C12-C13 a-epoxide for the synthesis of target molecule.

Abstract:The total syntheses of epothilones A (1) and B (2) and several analogues thereof are described. The reported strategy relies on a macrolactonization approach and features selective epoxidation of the macrocycle double bond in precursors 3 and 4 (Scheme 1), respectively, as well as high convergency and flexibility. Building blocks 9−12 and 15 were constructed by asymmetric processes and coupled via Wittig, aldol, and macrolactonization reactions to afford the basic skeleton of epothilones and that of several of their analogues by a relatively short route. The utilization of intermediate 14, obtained via a stereoselective Wittig reaction and its Enders coupling to SAMP hydrazone 13 (Scheme 8), in combination with a stereoselective aldol reaction with the modified substrate 69 (Scheme 10) improved the stereoselectivity and efficiency of the total synthesis of these new and highly potent microtubule binding antitumor agents.

Abstract:The concise synthesis of the novel telomerase inhibitors dictyodendrins B and E was completed in only 9 and 11 steps (longest linear sequence). The highly convergent strategy employed a palladium-catalyzed Larock indole synthesis and a palladium-mediated one-pot consecutive Buchwald-Hartwig amination/C-H activation reaction as key steps. The present synthesis exhibits respectable levels of atom-, redox-, and step-economy.

Abstract:The first asymmetric total syntheses of (+)-dihydrolyfoline and (-)-5-epi-dihydrolyfoline have been achieved in five and six steps with 4.6% and 14% overall yields, respectively, in which the chiral biaryl axes were constructed in a highly regioselective and stereoselective manner via a biogenetic enzymatic oxidative couplings of phenols, and the requisite quinolizidinone cores were prepared by an enzymatic Mannich reaction.

Abstract:A Stille coupling strategy has been utilized to complete a total synthesis of epothilone E from vinyl iodide 7 and thiazole-stannane 8h. The central core fragment 7 and its trans-isomer 11 were prepared from triene 15 using ring-closing metathesis (RCM), and were subsequently coupled to a variety of alternative stannanes to provide a library of epothilone analogues 18a-o and 19a-o. The Stille coupling approach was then used to prepare epothilone B analogues from the key macrolactone intermediate 24 which was itself synthesized by a macrolactonization based strategy.

Abstract:The first asymmetric total synthesis of bioactive diterpenoid hypoestin A with an unprecedented [5-8-5-3] tetracyclic skeleton is accomplished in 15 steps from commercially available (R)-limonene. Furthermore, the second asymmetric total syntheses of sesterterpenoids albolic acid and ceroplastol II in 21 steps are also reported. The synthetically challenging and highly functionalized [X-8-5] (X = 5 or 7) tricarbocyclic ring systems found in hypoestin A, albolic acid, ceroplastol II, and schindilactone A, as well as other natural products, are efficiently and directly constructed via a unique intramolecular Pauson-Khand reaction of an allene-yne. This work represents the first reported use of the Pauson-Khand reaction to access synthetically challenging eight-membered-ring systems in natural product synthesis.

Abstract:For structure-activity relationship studies, two series of epothilone A (1) analogues have been designed and synthesized, one containing an oxazole moiety instead of the thiazole heterocycle and the other containing a spirocyclopropane moiety in place of the gem-dimethyl group at position C-4 (4,4-ethano-epothilones). The olefin metathesis strategy in solution was utilized for the chemical synthesis of these compounds starting with key building blocks 7-9 for the oxazole series (compounds 2, 14-18, 21-26) and building blocks 8, 30, and 31 for the 4,4-ethano series (compounds 3, 39-43, 46-51), The convergent strategy towards the designed epothilone A series involved a) an aldol condensation reaction, b) an esterification reaction, c) an olefin metathesis reaction catalyzed by [RuCl2(=CHPh)(PCy3)(2)], and d) epoxidation of the macrocycle double bond.

Abstract:In order to probe structure-activity relationships in the epothilone area, two series of epothilone B analogues have been designed and synthesized. The first series containing an oxazole moiety in place of a thiazole on the side chain was constructed by utilizing key intermediates 7-9 or 10, 12, and 13 (Scheme 1), whereas the second series containing an ethano group instead of the gem-dimethyl group at position 4 was synthesized from fragments 42 and 43. A Yamaguchi-type macrolactonization reaction was used to construct the macrocycle from a seco-acid, which was assembled, in both cases, by means of a) an aldol reaction, b) an Enders alkylation, and c) a Wittig-type reaction. This convergent strategy provided access to oxazole analogues 2, 4, 29-32 and 4,4-ethano derivatives 3, 40, 60-63 for biological studies.

Abstract:An efficient total synthesis of (±)-13-epineostenine (2) has been achieved in 15 steps and 17% overall yield. This approach involved the key alkylation/Michael additions of the central 1,4-cyclohexanedione monoethylene acetal and all of the stereocenters on central cyclohexane moiety were generated in highly stereoselectivity.

Abstract:The first total synthesis of both C-11 epimers of 13-hydroxy-α-eudesmol 1a and 1b by the use, as a key reaction, of the Sharpless asymmetric dihydroxylation of alkene 7 is presented. The absolute configuration of natural 13-hydroxy-α-eudesmol is established through comparison of the 1H NMR spectrum of natural diol and synthetic diols. In our synthesis another natural product (+)-α-selinene 2 has also been accomplished.

Abstract:The total synthesis of endolides A and B has been achieved in a concise and highly stereoselective fashion (12 steps; 16.2 and 16.0% overall yield, respectively). Key features of the route include a modified Negishi coupling between 3-bromofuran and an organozinc reagent derived from an iodoalanine derivative for the synthesis of a 3-(3-furyl)alanine derivative, and a judicious choice of reaction conditions to overcome the conformational constraints placed by converting a linear peptide into the corresponding macrocycle.

Abstract:A seven-step total synthesis of (+)-cylindricine D and epi-cylindricine D is described. The efficiency relies on the highly diastereoselective reductive bis-functionalization of N,O-protected (S)-pyroglutaminol by our recently improved protocol to build the N-α-tert-alkylamine motif, and a telescoped one-step four-reaction protocol to build the fused bicyclic ring system. (+)-Cylindricine C and its 2-epimer were synthesized by essentially the same sequence in eight steps. Acetylation of the latter then delivered (+)-cylindricine E and its 2-epimer, respectively.
