Abstract:Structured lipids have attracted significant interest in industries, such as food, pharmaceuticals, and cosmetics, because they can be alternatives to traditional lipids. Enzymatic synthesis techniques utilizing lipases have gained attention for structured lipids synthesis because of their site and substrate specificity. However, most lipases discovered to date exhibit sn-1,3 regiospecificity or nonspecificity, limiting the selective range of natural oils and fats that can be used as raw materials. In this review, we provide an overview of the current synthesis methods and applications of structured lipids and the limitations of existing lipases in producing structured lipids. In addition, we discuss innovative approaches, including metagenomics, and machine learning, to discover, and classify new lipases and the use of gene editing technologies for lipase engineering. These efforts aim to overcome the limitations of existing lipases and expand the range of natural oils and fats that can be used for the production of structured lipids. Therefore, this study aims to promote a better understanding of structured lipids as a material with high-added value and provides insight into ongoing research efforts in the field.

Abstract:To introduce the liposomes preparation and targeting in recent advances. The method of preparing liposomes comprises conven-tional methods(e. g., thin lipid film hydration method, reverse phase evaporation method, ethanol injection method) and novel methods (e. g.,micro hydrodynamic focusing, supercritical reverse phase evaporation method, freeze drying method), the novel methods for preparing liposomes having high encapsulation efficiency, uniform particle size, with no residual organic solvent. Compared with ordinary liposomes, targeted liposomes(e. g.,long-circulating liposomes, physical chemistry target liposomes, ligand targeting liposomes) can specifically target tumor tissue, improve drug efficacy, and decrease drug toxicity.

Abstract:Liposomes (or lipid vesicles) are a versatile platform as carriers for the delivery of the drugs and other macromolecules into human and animal bodies. Though the method of using liposomes has been known since 1960s, and major developments and commercialization of liposomal formulations took place in the late nineties (or early part of this century), newer methods of liposome synthesis and drug encapsulation continue to be an active area of research. With the developments in related fields, such as electrohydrodynamics and microfluidics, and a growing understanding of the mechanisms of lipid assembly from colloidal and intermolecular forces, liposome preparation techniques have been enriched and more predictable than before. This has led to better methods that can also scale at an industrial production level. In this review, we present several novel methods that were introduced over the last decade and compare their advantages over conventional methods. Researchers beginning to explore liposomal formulations will find this resource useful to give an overall direction for an appropriate choice of method. Where possible, we have also provided the known mechanisms behind the preparation methods.

Abstract:Lipids, the fundamental components of cell membrane, play important roles in the whole cycle of cell life, thus attracting worldwide attention, owing to their physicochemical property and extensive use in the applications based on lipid assemblies. Compared with liposomes, lipid nanotubes (LNTs) usually possess unique properties, such as highly ordered structure, precise molecular recognition, and the possibility of substance transport, thus providing more potential applications in different research fields. However, until now, there are still quite rare cases of LNTs successfully employed in practical applications. Bearing this in mind and based on our own experience in this field, we summarized and discussed the recent progress of the fabrication approaches and representative applications of the LNTs in the past decade, which would potentially provide basic understanding and guidance towards their future development.

Abstract:Liposomes are small spherical vesicles composed of phospholipid bilayers capable of encapsulating a variety of ingredients, including water- and oil-soluble compound, which are one of the most commonly used piggybacking and delivery techniques for many active ingredients and different compounds in biology, medicine and cosmetics. With the increasing number of active cosmetic ingredients, the concomitant challenge is to effectively protect, transport, and utilize these substances in a judicious manner. Many cosmetic ingredients are ineffective both topically and systemically when applied to the skin, thus changing the method of delivery and interaction with the skin of the active ingredients is a crucial step toward improving their effectiveness. Liposomes can improve the delivery of active ingredients to the skin, enhance their stability, and ultimately, improve the efficacy of cosmetics and and pharmaceuticals. In this review, we summarized the basic properties of liposomes and their recent advances of functionalities in cosmetics and and pharmaceuticals. Also, the current state of the art in the field is discussed and the prospects for future research areas are highlighted. We hope that this review will provide ideas and inspiration on the application and development of cosmetics and pharmaceuticals.

Abstract:In the last decades, pharmaceutical interested researches aimed to develop novel and innovative drug delivery techniques in the medical and pharmaceutical fields. Recently, phospholipid vesicles (Liposomes) are the most known versatile assemblies in the drug delivery systems. The discovery of liposomes arises from self-forming enclosed phospholipid bilayer upon coming in contact with the aqueous solution. Liposomes are uni or multilamellar vesicles consisting of phospholipids produced naturally or synthetically, which are readily non-toxic, biodegradable, and are readily produced on a large scale. Various phospholipids, for instance, soybean, egg yolk, synthetic, and hydrogenated phosphatidylcholine consider the most popular types used in different kinds of formulations. This review summarises liposomes composition, characterisation, methods of preparation, and their applications in different medical fields including cancer therapy, vaccine, ocular delivery, wound healing, and some dermatological applications.

Abstract:Liposomes, which possess the properties of nano-scale, biofilm similar structure, excellent biocompatibility, become more and more useful in the drug development as the delivery system. Liposomes are relatively stable, their aqueous phase could contain the hydrophilic drugs and their phospholipid bilayer should localize the lipophilic drugs. Moreover, their surface-modifiable characteristics have really extended the liposomes' application to targeting and environmental sensitive delivery system. In order to make the common liposome more fit the human and animal body's complex environment, the structural variation strategy in the head, tail and bond of lipid molecules have been employed to develop the different functionalized liposomes-based drug delivery system for the localizable relieve and organ/tissue targeting relieve. In this paper, we would like to summarize the recent development on the design and optimization of liposomes, including Long-circulation liposomes, Specific active targeting liposomes, Environmental sensitive liposomes, Multifunctional liposomes, and so on. And the liposome content selection and current status of clinical application are systematically discussed.

Abstract:Recently, owing to well-controlled release, enhanced distribution and increased permeability, nanocarriers used for alternative drug and food-delivery strategies have received increasingly attentions. Nanocarriers have attracted a large amount of interest as potential carriers of various bioactive molecules for multiple applications. Drug and food-based delivery via polymeric-based nanocarriers and lipid-based nanocarriers has been widely investigated. Nanocarriers, especially liposomes, are more and more widely used in the area of novel nano-pharmaceutical or food-based design. Herein, we aimed to discuss the recent advancement of different surface-engineered nanocarriers type, along with cutting-edge applications for food and nanomedicine and highlight the alternative of phytochemical as nanocarrier. Additionally, safety concern of nanocarriers was also highlighted.

Abstract:Over the past several decades, liposomes have been extensively developed and used for various clinical applications such as in pharmaceutical, cosmetic, and dietetic fields, due to its versatility, biocompatibility, and biodegradability, as well as the ability to enhance the therapeutic index of free drugs. However, some challenges remain unsolved, including liposome premature leakage, manufacturing irreproducibility, and limited translation success. This article reviews various aspects of liposomes, including its advantages, major compositions, and common preparation techniques, and discusses present U.S. FDA-approved, clinical, and preclinical liposomal nanotherapeutics for treating and preventing a variety of human diseases. In addition, we summarize the significance of and challenges in liposome-enabled nanotherapeutic development and hope it provides the fundamental knowledge and concepts about liposomes and their applications and contributions in contemporary pharmaceutical advancement.

Abstract:Microfluidics is an emerging technology that can be employed as a powerful tool for designing lipid nano-microsized structures for biological applications. Those lipid structures can be used as carrying vehicles for a wide range of drugs and genetic materials. Microfluidic technology also allows the design of sustainable processes with less financial demand, while it can be scaled up using parallelization to increase production. From this perspective, this article reviews the recent advances in the synthesis of lipid-based nanostructures through microfluidics (liposomes, lipoplexes, lipid nanoparticles, core-shell nanoparticles, and biomimetic nanovesicles). Besides that, this review describes the recent microfluidic approaches to produce lipid micro-sized structures as giant unilamellar vesicles. New strategies are also described for the controlled release of the lipid payloads using microgels and droplet-based microfluidics. To address the importance of microfluidics for lipid-nanoparticle screening, an overview of how microfluidic systems can be used to mimic the cellular environment is also presented. Future trends and perspectives in designing novel nano and micro scales are also discussed herein.

Abstract:Liposome-based drugs have emerged as highly successful nanomedicines for clinical applications owing to their excellent biocompatibility, biodegradability, and targeting effects. However, the lipids that play a crucial role in most liposome formulations are often engineered rather than natural phospholipids. From this perspective, we outlined the classification of engineered lipids based on their material chemistry, including charged, polymer-conjugated, ligand-conjugated, and choline phosphate lipids. We also discussed their drug, gene, protein, and probe delivery applications. With the increasing requirements for the innovation of nano drug excipients by the Food and Drug Administration, attention to drug efficacy and side effects has increased in clinics, and the development of superior engineered lipids has become urgent. Therefore, we aim to provide a new idea for developing engineered lipids in which lipids can be engineered with minimal chemical structure changes to achieve maximum functions enhancement, with choline phosphate lipids being a favorable choice. In the future, novel engineered lipids and liposomes should be developed with more powerful functions and superior therapeutic safety, which will allow their utilization in the diagnosis and treatment of major diseases and be valuable for more clinical applications.

Abstract:Drug delivery has always been a hot topic of concern for researchers. A good drug delivery system can not only enhance the efficacy of drugs, but also reduce their side effects. Liposome, a microscopic spherical particle similar to bio-membrane, is an ideal drug carrier being used during recent years for its biocompatibility, which has changed the traditional administration method. This article analyze different examples, refers to multiple literature and provides an overview of the birth and development of liposomes, as well as their latest types and applications. Through the research of many literatures, liposome carriers are now applied in many areas, such as cancer, antibacterial drug, wound healing and so on. It is clearly sure that with new polymer materials and different formulations, liposome carriers can handle the treatment of a sea of diseases.

Abstract:<p>Conventional liposomes still face many challenges associated with the poor physical and chemical stability, considerable loss of encapsulated cargo, lack of stimulus responsiveness, and rapid elimination from blood circulation. Integration of versatile functional biopolymers has emerged as an attractive strategy to overcome the limitation of usage of liposomes. This review comprehensively summarizes the most recent studies (2015–2020) and their challenges aiming at the exploration of biopolymer-liposome hybrid systems, including surface-modified liposomes, biopolymer-incorporated liposomes, guest-in-cyclodextrin-in-liposome, liposome-in-hydrogel, liposome-in-film, and liposome-in-nanofiber. The physicochemical principles and key technical information underlying the combined strategies for the fabrication of polymeric liposomes, the advantages and limitations of each of the systems, and the stabilization mechanisms are discussed through various case studies. Special emphasis is directed toward the synergistic efficiencies of biopolymers and phospholipid bilayers on encapsulation, protection, and controlled delivery of bioactives (<em>e.g.,</em> vitamins, carotenoids, phenolics, peptides, and other health-related compounds) for the biomedical, pharmaceutical, cosmetic, and functional food applications. The major challenges, opportunities, and possible further developments for future studies are also highlighted.</p>

Abstract:Lamellarins are polyaromatic alkaloids isolated from marine organisms, including mollusks, tunicates, and sponges. Currently, over 100 structurally distinct natural lamellarins have been reported, and most of them exhibit promising biological activities, such as topoisomerase inhibition, mitochondrial function inhibition, multidrug resistance reversing, and anti-HIV activity. There has also been a significant progress on the synthetic study of lamellarins which has been regularly updated by numerous medicinal chemists as well. This review provides a detailed summary of the synthesis, pharmacology, and structural modification of lamellarins over the past decades.

Abstract:Membraneless organelles (MLOs) formed by liquid-liquid phase separation (LLPS) have been extensively studied due to their spatiotemporal control of biochemical and cellular processes in living cells. These findings have provided valuable insights into the physicochemical principles underlying the formation and functionalization of biomolecular condensates, which paves the way for the development of versatile phase-separating systems capable of addressing a variety of application scenarios. Here, we highlight the potential of constructing synthetic MLOs with programmable and functional properties. Notably, we organize how these synthetic membraneless compartments have been capitalized to manipulate enzymatic activities and metabolic reactions. The aim of this review is to inspire readerships to deeply comprehend the widespread roles of synthetic MLOs in the regulation enzymatic reactions and control of metabolic processes, and to encourage the rational design of controllable and functional membraneless compartments for a broad range of bioengineering applications.

Abstract:Achieving superlubricity, a state of lubrication where friction nearly vanishes, has become one of the most promising approaches to combat friction-induced energy dissipation and medical device failure. Phospholipids are amphiphilic molecules comprising highly hydrophilic phosphatidylcholine head groups as well as hydrophobic hydrocarbon chains, When solubilized, phospholipids can readily self-assemble to form different structures such as bilayers and vesicles (liposomes). Recently, liposomes have been identified as excellent lubricants, especially in the boundary lubrication regime the most common lubrication status in the field of biotribology. In this review, we summarize recent progress in employing liposomes as key players for employing superlubricity in biomedical applications. The relationship between lipids and liposomes, manufacturing approaches, lubrication regimes, and regulation mechanisms of liposomes are discussed. Finally, we indicate possible future directions for the use of liposome-mediated superlubricity in biomedical applications.

Abstract:Liposomes are consolidated and attractive biomimetic nanocarriers widely used in the field of drug delivery. The structural versatility of liposomes has been exploited for the development of various carriers for the topical or systemic delivery of drugs and bioactive molecules, with the possibility of increasing their bioavailability and stability, and modulating and directing their release, while limiting the side effects at the same time. Nevertheless, first-generation vesicles suffer from some limitations including physical instability, short in vivo circulation lifetime, reduced payload, uncontrolled release properties, and low targeting abilities. Therefore, liposome preparation technology soon took advantage of the possibility of improving vesicle performance using both natural and synthetic polymers. Polymers can easily be synthesized in a controlled manner over a wide range of molecular weights and in a low dispersity range. Their properties are widely tunable and therefore allow the low chemical versatility typical of lipids to be overcome. Moreover, depending on their structure, polymers can be used to create a simple covering on the liposome surface or to intercalate in the phospholipid bilayer to give rise to real hybrid structures. This review illustrates the main strategies implemented in the field of polymer/liposome assembly for drug delivery, with a look at the most recent publications without neglecting basic concepts for a simple and complete understanding by the reader.

Abstract:Ocular barriers and the poor water solubility of drug candidates present a number of problems for the development of ocular drug delivery systems. Recently, the emergence of lipid-based nanocarriers has provided a viable means of enhancing the bioavailability of ophthalmic formulations. A number of these formulations have been found to be clinically active and several others are currently undergoing clinical trials. In this review, the advantages of lipid-based nanocarriers as non-invasive topical ocular drug delivery systems are presented. Many systems, including emulsions, liposomes, cubosomes, niosomes and other lipid-based nanocarriers, are reviewed.
