Abstract:Background and purpose:Melanoma-associated antigens(MAGE) are a group of well-characterized members of the cancer/testis antigens(CTA) family.They are rarely expressed in normal adult tissues except for testis,but highly expressed in various forms of cancer.So they are considered as the tumor-specific antigen(TSA).MAGE-A genes consist of 12 family members,including MAGE-A1-MAGE-A12.The aim of this study was to investigate the regulative effects of 4 kinds of MAGE-A9 deletion mutants pcDNA3.1 MAGE-A9(1-29),pcDNA3.1 MAGE-A9(1-114),pcDNA3.1 MAGE-A9(115-285) and pcDNA3.1 MAGE-A9(286-315) on the proliferation of MCF-7 cells.Methods:Four MAGE-A9 deletion mutants were constructed and transfected into MCF-7 cells.RT-PCR and Western blot were performed to examine the expression of these plasmids.MTT and colony formation assay was performed to detect the effect of MAGE-A9 deletion mutant on the proliferation of MCF-7 cells.Results:pcDNA3.1 MAGE-A9(1-29),pcDNA3.1 MAGE-A9(1-114) and pcDNA3.1 MAGE-A9(286-315) had no significant effect on the proliferation of MCF-7 cells(P>0.05).pcDNA3.1 MAGE-A9(115-285) significantly increased the proliferation of MCF-7 cells(P<0.05).Conclusion:pcDNA3.1 MAGE-A9(115-285) could increase the proliferation of MCF-7 cells,suggesting that MAGE-A9(115-285) could be the functional domain of MAGE-A9.

Abstract:Melanoma-associated antigens (MAGEs) are a group of well-characterized members of the cancer/testis antigen (CTA) family, which is one of the largest groups of human tumor-associated antigens. MAGE-A9 is a particular member in the context of the MAGE-A gene family and was defined as presenting prognostic relevance in certain type of human cancer. However, the expression of MAGE-A9 in invasive ductal breast cancer (IDC) and the relationship with the clinical attributes of IDC, especially prognostic characteristic, remain poorly understood. In this present study, one-step quantitative reverse transcription polymerase chain reaction (18 fresh-frozen IDC tissues and corresponding non-cancerous tissues) and immunohistochemistry by tissue microarrays (82 IDC tissue samples and 70 matched tumor-adjacent non-cancerous tissue samples) were performed to characterize expression of the MAGE-A9 gene in IDC. Kaplan-Meier survival and Cox regression analyses were performed to evaluate the prognosis of IDC. The results of qPCR and IHC analysis showed that the expression of MAGE-A9 in IDC was significantly higher than that in corresponding non-cancerous tissue. Moreover, the expression level of MAGE-A9 protein in IDC was significantly related to histological grade (p = 0.011) and distant metastasis (p = 0.019). Multivariate analysis with the Cox regression model showed that MAGE-A9 protein expression (p = 0.009), histological grade (p = 0.014), lymph node metastasis (p = 0.012) and distant metastasis (p = 0.011) were independent prognostic factors for overall survival of IDC patients. The data suggest that MAGE-A9 expression is correlated with malignant attributes of IDC and it may serve as a novel prognostic factor and an ideal candidate for targeted therapy in IDC treatment.

Abstract:Background Cancer/testis antigens (CTAs) are a special type of tumor antigen and are believed to act as potential targets for cancer immunotherapy. Methods In this study, we first screened a rational CTA MAGE-A1 for lung adenocarcinoma (LUAD) and explored the detailed characteristics of MAGE-A1 in LUAD development through a series of phenotypic experiments. Then, we developed a novel MAGE-A1-CAR-T cell (mCART) using lentiviral vector based on our previous MAGE-A1-scFv. The anti-tumor effects of this mCART were finally investigated in vitro and in vivo. Results The results showed striking malignant behaviors of MAGE-A1 in LUAD development, which further validated the rationality of MAGE-A1 as an appropriate target for LUAD treatment. Then, the innovative mCART was successfully constructed, and mCART displayed encouraging tumor-inhibitory efficacy in LUAD cells and xenografts. Conclusions Taken together, our data suggest that MAGE-A1 is a promising candidate marker for LUAD therapy and the MAGE-A1-specific CAR-T cell immunotherapy may be an effective strategy for the treatment of MAGE-A1-positive LUAD.

Abstract:Purpose: The possibility of tumor rejection antigen, encoded by the MAGE-1 gene, being a target for immunotherapy in hepatocellular carcinoma (HCC) patients and the cloning of the full-length cDNA of this gene for subsequent studies were explored with the aim of discovering new immunotherapeutic strategies for HCC. Methods: Expression of the MAGE-1 gene in HCC specimens and HCC cell lines was examined by the reverse transcription/polymerase chain reaction (RT-PCR) with a pair of specific primers, which gave a 421-bp fragment. Another pair of primers were then used to amplify the full-length MAGE-1 cDNA by the same method. The PCR products were then digested by restriction endonucleases and inserted into the plasmid PUC19. After primary selection of the recombinants by endonuclease digestion, the sequences of the inserted gene fragments were confirmed by DNA sequence analysis. Results: In 45 HCC patients, MAGE-1 mRNA was expressed in 27 tumor tissues (60%) and 5 paratumor tissues (11.1%). All the four HCC cell lines positively expressed the MAGE-1 gene. Owing to the high homology of the MAGE gene family, we obtained three clones of different MAGE genes using the same pair of cloning primers. The three clones were confirmed to be a full-length MAGE-1 gene, a 750-bp fragment of the MAGE-3 gene and a fragment highly homologous to MAGE-6 and MAGE-12 but not identical to any known MAGE genes. Conclusion: The high expression frequency of MAGE-1 gene in HCC suggests the possibility of using it as a target for immunotherapy in HCC patients. Some MAGE genes other than MAGE-1 may also be expressed in HCC together with an unknown MAGE gene that might introduce new targets for immune attacks. The three gene clones obtained in this study can be used in further investigations and especially in the development of new liver cancer vaccines.

Abstract:Objective To explore protein expression and significance of MAGE genes in colorectal carcinoma(CRC)tissues.Methods The expression of MAGE genes were studied by using tissue chip and immunochemistry methods in primary CRC tissue and paired adjacent tissue samples in 97 cases.Data were analyzed with x2-test by SPSS 16.0 software.Results The protein expression of MAGE-A3,MAGE-A4 and MAGE-A10 genes were 57％(56/97),63％(61/97)and 28％(27/97)respectively in 97 cases of primary adenocarcinoma.The protein expression frequency of MAGE-A3 in poor colorectal adenocarcinomas was significantly higher than in well-and moderately disfferentiated adenocarcinomas(x2 =9.133,P =0.010).MAGE-A10 in poor colorectal primary adenocarcinomas was significantly higher than in well and moderately adenocarcinomas(x2 =15.280,P =0.000); MAGE-A10 protein expression was significantly higher in stage TNM Ⅲ + Ⅳ than in stage TNM Ⅰ + Ⅱ(x2 =4.227,P=0.040); MAGE-A10 gene expression was higher in metastasis lymphoid node than in no metastasis lymphoid node(x2 =5.557,P =0.018),and the expression level was higher in primary lesion with the increasing of the numbers of lymphoid node metastasis(x2 =7.296,P =0.026).Conclusions The protein expression of MAGE genes is associated with the tumor differentiation,TNM stage and lymphoid node metastasis.MAGE-A3 and MAGE-A10 genes are the possible prognosis marker and potential target of immunotherapy of CRC.

Abstract:The melanoma antigen (MAGE) protein family contains more than 25 members that share a conserved MAGE homology domain (MHD). Type I MAGE genes exhibit cancer/testis-specific expression patterns and antigenic properties which render them ideal candidates for cancer immunotherapies. Maged1, a type II MAGE gene, is ubiquitously expressed and has been previously shown to play an important role in neuronal apoptosis during development. Recent studies have expanded the functional tissues and processes in which Maged1 activity is important and uncovered interacting partners of MAGED1 protein, adding novel layers to Maged1 functions. Maged1 plays a role in anti-tumorigenesis in a variety of cell types, and the down-regulation of MAGED1 has been observed in tumor cells. Moreover, MAGED1 can interact with a specific group of nuclear members and regulate circadian clock functions. These newly identified functions will enrich the molecular and clinical studies of the MAGE family of proteins.

Abstract:AIM:To detect the expression of melanoma antigen-1 and -3 genes (MAGE-1 and MAGE-3) and explore its significance in immunotherapeutic application.METHODS:The expression of MAGE-1 and MAGE-3 in gastric cancer, para-tumorous normal mucosa, and gastric biopsy tissues were detected by RT-PCR. The target genes in RT-PCR product were verified by means of DNA sequencing.RESULTS:MAGE-1 expression was positive in 26 out of 36 (72.22%) cases of gastric cancer, MAGE-3 positive in 25 out of 36(69.44%), and both MAGE-1 and MAGE-3 positive in 19 out of 36 (52.78%). Positive expression rates of MAGE-1, MAGE-3 and both MAGE-1 and MAGE-3 were 47.22%, 44.44% and 30.55%, respectively in para-tumorous normal mucosa, while 29.73%, 27.03% and 8.1% in gastric biopsy tissues The positive rates of MAGE-1 and MAGE-3 gene expressions in tumor tissues was significant higher than those its parallelism mucosae and biopsy tissues from chronic gastritis. The expressions of MAGE-1 and MAGE-3 genes in tumor tissues were relevant to intruded depth and the differentiated degree of tumor cells (P<0.05).CONCLUSION:MAGE-1 and MAGE-3 protein may be used as targets molecules of immunotherapy for gastric cancer and indexes for follow-up study of chronic gastritis as well.

Abstract:To date, dozens of melanoma-associated antigens (MAGEs) have been identified and classified into 2 subgroups, I and II. Subgroup I consists of antigens which expression is generally restricted to tumor or germ cells, also named as cancer/testis (CT) antigen. Proteins and peptides derived from some of these antigens have been utilized in promising clinical trials of immunotherapies for gastrointestinal carcinoma, esophageal carcinoma, pulmonary carcinoma and so on. Various MAGE family members play important physiological and pathological roles during embryogenesis, germ cell genesis, apoptosis, etc. However, little is known regarding the role of MAGE family members in cell activities. It is reasonable to speculate that the genes for subgroup I MAGEs, which play important roles during embryogenesis, could be later deactivated by a genetic mechanism such as methylation. In the case of tumor formation, these genes are reactivated and the resultant proteins may be recognized and attacked by the immune system. Thus, the subgroup I MAGEs may play important roles in the immune surveillance of certain tumor types. Here, we review the classifications of MAGE family genes and what is known of their biological functions.

Abstract:PD-L1 programmed death ligand 1/CD274 has been shown to be an excellent immune therapy target for many tumors. Currently, studies have shown PD-L1in combination with other immune therapy targets can improve patient outcomes generating the need for new targets. One candidate is the cancer testis antigen Melanoma Antigen Gene Family (MAGE-A) because these genes have limited expression in normal tissues and high expression in cancer. Clinical trials have already begun targeting MAGE-A3 and MAGE-A4 proteins in multiple tumor types. Protein assessment of individual MAGE-A family members has been challenging due to sequence homology as high as ninety-five percent between members. Previously, with the use of CytoSections two highly specific antibodies for immunohistochemistry to MAGE-A3 (clone OTI1H1) and MAGE-A4 (clone OTI2C1) were found. In this study, the MAGE-A3 and MAGE-A4 antibodies were screened with PD-L1 on colon, lung, and bladder cancers. Results show detection of both MAGE-A3 and MAGE-A4 in 25% of colon; 30% of lung; and 20% of bladder cancers. More than 70% of the lung and bladder cancers were positive for MAGE-A4 but only six of twenty-four colon cancers were positive for MAGE-A4. The results show 60 percent of the positive for MAGEA3 or MAGEA4 in colon, lung, and bladder cancers were also positive for PD-L1. This study suggests that MAGE-A3 and MAGE-A4 have tumor specific expression patterns and a potential to be a co-immunotherapy target with PD-L1. [Expression of MAGE-B Genes in Hepatocellular Carcinoma].

Abstract:OBJECTIVE:To investigate the expression of MAGE-B genes in hepatocellular carcinoma (HCC) in order to find new targets for immunotherapy. METHODS:The expression of MAGE-B1, B2, A1 and A3 mRNA was detected using RT-PCR in HCC tissues and the corresponding adjacent non-HCC tissues from 47 HCC patients, 30 samples of cirrhosis and normal liver tissues. Four samples selected randomly from MAGE-B1 or B2 with positive RT-PCR results were sequenced to confirm the results of RT-PCR. The relationship between the expression of MAGE-B and some clinicopathological parameters was analyzed. RESULTS:MAGE-B1 mRNA and MAGE-B2 mRNA were detected in 44.7% (21/47) and 61.7% (29/47) of HCC samples, respectively, while neither MAGE-B1 nor MAGE-B2 could be detected in the corresponding adjacent non-HCC liver tissues. In addition, none of 30 samples of cirrhosis and normal liver tissues was shown to express both MAGE-B genes. The DNA sequence confirmed that the RT-PCR products were truly target cDNA. The frequency of the expression of MAGE-A1 and A3 was 74.5% (35/47) and 44.7% (21/47), respectively. There was significant correlation between the expression of MAGE-B and MAGE-A (P < 0.05). However, the positive expression of MAGE-B was observed in 5 out of 12 HCC tissues without expression of MAGE-A1 and/or A3. When all four MAGE genes were examined, the positive rate of expression of one, two, three and four genes was 83.0% (39/47), 55.3% (26/47), 48.9% (23/47), and 38.3% (18/47) of 47 HCC tissues, respectively. No correlation was found between the expression of MAGE-B and clinical parameters such as age, sex, tumor size, degree of tumor differentiation, serum alpha-fetoprotein level and hepatitis B virus or hepatitis C virus infection (P > 0.05). CONCLUSION:MAGE-B genes are expressed with relatively high frequency and specificity in HCC. Most HCC patients with positive expression of at least one member of MAGE-B or MAGE-A gene family are adequate candidates to receive specific immunotherapy. Frequent co-expression of multiple members of MAGE-B and MAGE-A subfamilies provides the possibility of using polyvalent vaccines to achieve more effective immunotherapeutic results.

Abstract:BACKGROUND:To investigate the expressions of MAGE-1,-2,-3 and -4 in lung cancer at mRNA level.METHODS:Using a reverse transcription-polymerase chain reaction assay (RT-PCR), the expressions of MAGE-1,-2,-3 and -4 in 35 lung cancer samples and non-tumorous lung tissues were examined.RESULTS:Of the 35 tumor samples, the MAGE-1,-2,-3 and -4 were expressed at mRNA level in 22.9% (8/35), 62.9% (22/35), 37.1% (13/35) and 77.1% (27/35) respectively. At least one of these genes was expressed in 85.7% (30/35) of the samples. Two or more of these genes were expressed in 71.4% (25/35) of the samples. None of the non-tumorous lung tissue was positive for these genes. No significant difference of the frequency of MAGE gene expression was found between adenocarcinoma and squamous cell carcinoma, between different tumor stages, and between groups with or without lymph node metastasis (P > 0.05).CONCLUSIONS:MAGE-1,-2,-3 and -4 are expressed at a high percentage in lung cancer. All of these genes are negative in the adjacent non tumorous lung tissue. These results indicate the possibility of a future specific immunotherapy for lung cancer based on these MAGE antigens.
Expression of the MAGE-1 Gene in Human Hepatocellular Carcinomas

Abstract:OBJECTIVE To further investigate the expression of MAGE-1 gene in hepatocellular carcinoma (HCC). METHODS The tumors and adjacent liver tissue from 45 HCC patients and liver tissue from 28 non-HCC patients (16 with liver cirrhosis and 12 with normal liver) were characterized by RT-PCR. A 421 bp PCR product from a cDNA fragment spanning exons 1, 2 and 3 was sequenced. The HLA type was assayed by standard ELISA in 43 HCC patients. RESULTS Thirty-two of 45 tumor tissues from HCC patients expressed MAGE-1 mRNA (71.1%). In contrast, MAGE-1 mRNA was not detected in adjacent tissues. Three were found to have point mutations at 3 identical sites resulting in the substitution of two amino acid residues. The most frequent HLA types in 43 HCC patients were: HLA-A2, 53.5%; A11, 25.6%; A24, 20.9%; A33, 20.9%; HLA-B13, 28.3% and B35, 23.2%. Expression of HLA-A33 (20.9%) was higher in HCC patients than that predicted in the normal Chinese population (8.8%). There was no discemable correlation between MAGE-1 expression and alpha-FP level, tumor size and hepatitis B or C virus infection. The identification of peptides which are restricted by haploptypes other than A1 should increase the opportunity for peptide based immunotherapy. CONCLUSIONS This study shows that MAGE-1 mRNA is highly expressed in HCC tumor tissue in Chinese patients. Previously unreported point mutations in the MAGE-1 gene are described and may also provide additional opportunities for immunotherapy.

Abstract:Melanoma-associated antigen (MAGE) is expressed on the surface of multiple tumor cell types and is a promising target of biotherapeutic drug delivery via the anti-MAGE-A1 antibody. In this study, a human single-chain variable fragment (scFv) antibody phage library was generated and applied to recombinant MAGE-A1-coated immunotubes by phage display technology. The soluble anti- MAGE-A1 scFv was expressed and purified by immobilized metal-chelated affinity chromatography (IMAC). The anti-MAGE-A1 scFv could bind native MAGE-A1 confirmed by enzyme-linked immunosorbent assay (ELISA), dot blot, and immunoprecipitation (IP) analysis. The immunotoxin was expressed and purified by IMAC successfully. The results indicated that the human anti-MAGE-A1 immunotoxin could provide a valuable drug for clinic cancer therapy.
Global analysis of HLA-A2 restricted MAGE-A3 tumor antigen epitopes and corresponding TCRs in non-small cell lung cancer

Abstract:Background: Advanced non-small cell lung cancer (NSCLC) is the most common type of lung cancer with poor prognosis. Adoptive cell therapy using engineered T-cell receptors (TCRs) targeting cancer-testis antigens, such as Melanoma-associated antigen 3 (MAGE-A3), is a potential approach for the treatment of NSCLC. However, systematic analysis of T cell immune responses to MAGE-A3 antigen and corresponding antigen-specific TCR is still lacking. Methods: In this study, we comprehensively screened HLA-A2 restricted MAGE-A3 tumor epitopes and characterized the corresponding TCRs using in vitro artificial antigen presentation cells (APC) system, single-cell transcriptome and TCR V(D)J sequencing, and machine-learning. Furthermore, the tumor-reactive TCRs with killing potency was screened and verified. Results: We identified the HLA-A2 restricted T cell epitopes from MAGE-A3 that could effectively induce the activation and cytotoxicity of CD8+ T cells using artificial APC in vitro. A cohort of HLA-A2+ NSCLC donors demonstrated that the number of epitope specific CD8+ T cells increased in NSCLC than healthy controls when measured with tetramer derived from the candidate MAGE-A3 epitopes, especially epitope Mp4 (MAGE-A3: 160-169, LVFGIELMEV). Statistical and machine-learning based analyses demonstrated that the MAGE-A3-Mp4 epitope-specific CD8+ T cell clones were mostly in effector and proliferating state. Importantly, T cells artificially expressing the MAGE-A3-Mp4 specific TCRs exhibited strong MAGE-A3+ tumor cell recognition and killing effect. Cross-reactivity risk analysis of the candidates TCRs showed high binding stability to MAGE-A3-Mp4 epitope and low risk of cross-reaction. Conclusions: This work identified candidate TCRs potentially suitable for TCR-T design targeting HLA-A2 restricted MAGE-A3 tumor antigen.

Tissue Expression Profile of Human MAGE-D1, the New Member of Melanoma Antigen-encoding Gene Family

Abstract:Members of MAGE(melanoma antigen-encoding gene) family are expressed in a wide variety of tumors but not in normal cells, with the exception of the male germ cells, placenta, and possibly, cells of the developing fetus. However, the discovery of MAGE-D1 has broken this discipline since it is ubiquitously expressed in normal tissues rather than strictly in cancer/testis. Moreover, the knowledge about the MAGE-D1 gene is far from being understood. The tissue expression profile of human MAGE-D1 using Northern blot and Multiple-Tissue-RNA Dot blot techniques were reported.The results demonstrated that human MAGE-D1 was extensively expressed in cancer cell lines and ubiquitously expressed in many normal tissues. In forty-eight kinds of tumors detected, expression of human MAGE-D1 were significantly up-regulated in thirteen and down-regulated in seven , implicating potential diagnostic value of MAGE-D1 in clinical application. Moreover, the expression level of MAGE-D1 in fetal tissues was higher than that in adult ones. However, although MAGE-D1 does not encode a tumor-special antigen, the biological function of MAGE-D1 and the relationship with tumor need much more pathological study.
Prediction, Synthesis and Identification of HLA-A2-restricted Cytotoxic T Lymphocyte Epitopes of the Tumor Antigen MAGE-n

Abstract:OBJECTIVE:To predict, synthesize, and identify HLA-A2-restricted cytotoxic T lymphocyte (CTL) epitopes of the tumor the novel antigen MAGE-n. METHODS:Long-distance prediction system SYFPEITHI combined with polynomial method was used to predict the HLA-A2-restricted CTL epitopes of the tumor antigen MAGE-n. The candidate epitopes were synthesized with solid phase strategies, purified with reverse phase high-performance liquid chromatography and identified by mass spectrometry, The binding affinity and biding stability of the synthesized peptides were examined by cellular competition-based HLA-A2 peptide binding assay, T2 peptide stabilization assay, and peptide-major histocompatibility complex dissociation assay. RESULTS:Five HLA-A2-restricted CTL epitopes of MAGE-n were selected: The epitopes QLVFGIEVV (159-167), IMPKTGGLI (195-203), and FLIIVLMI (201-209) with high HLA-A2 binding affinity (LC(50) < 15 micro mol/L) and binding stability (DT(50) > 6 h) were selected as candidate epitopes for further study in immunotherapy for tumor. CONCLUSION:Epitope prediction combined with epitope reconstruction improves the study of HLA-A2-restricted CTL epitopes of the tumor antigen MAGE-n. The selected epitopes of MAGE-n may be used in the design of therapeutic peptide vaccine for hepatocellular carcinoma.
Expression and Prognostic Value of MAGE-A9 in Laryngeal Squamous Cell Carcinoma.

Abstract:BACKGROUND:Melanoma-associated antigen (MAGE) family genes are reported to play important roles in the development of human cancers. However, the relationship between the expression of MAGE-A9 and clinicopathological characteristics in human laryngeal carcinoma remains unclear. This study aimed to examine the expression of MAGE-A9, and to evaluate the clinical significance of its expression in human laryngeal squamous cell carcinoma (LSCC).METHODS:Quantitative real-time reverse transcription-PCR (qPCR) and immunohistochemistry (IHC) were performed to characterize the expression of MAGE-A9 in LSCC tissues and tumor-adjacent normal tissues. Kaplan-Meier survival and Cox regression analyses were performed to evaluate the prognosis of patients with LSCC.RESULTS:The expression of MAGE-A9 was significantly higher in LSCC than in tumor-adjacent normal tissues. Cytoplasmic expression of MAGE-A9 was detected in 70 of 123 (56.9%) LSCC specimens. Levels of MAGE-A9 in LSCC were related to histopathological grade (P = 0.024). Kaplan-Meier survival and Cox regression analysis revealed that MAGE-A9 expression level and lymph node metastasis were independent prognostic factors of LSCC (P = 0.005; P = 0.001, respectively).CONCLUSIONS:Our study suggests that MAGE-A9 expression is a prognostic biomarker for LSCC patients. High expression of MAGE-A9 indicates unfavorable survival outcome in LSCC patients.

Abstract:To date, dozens of melanoma-associated antigens (MAGEs) have been identified, and classified into subgroups I and II. Subgroup I, also named as cancer/testis (CT) antigen, consists of antigens whose expression is generally restricted to tumor cells, or germ cells. Proteins and peptides derived from some of these antigens have been used in promising clinical trials for cancer immunotherapies. Although the role of MAGEs in cell activities is unclear, various MAGEs were noticed to function physiologically and pathologically during embryogenesis, germ cell genesis, apoptosis, etc. It is reasonable to speculate that the genes of subgroup I of MAGEs, after playing important roles during embryogenesis, could be deactivated by genetic mechanisms, such as methylation. In the case of tumor formation, these genes are reactivated, and the resulted proteins may be recognized and attacked by immune system. Thus, subgroup I of MAGEs may play important roles in immune surveillance of certain tumor types. Here, we have sought to review the classifications and biological functions of MAGE family genes.
