Effects of paeonol, extractant of vegetable drug, on apoptosis of human colorectal cancer cells and controlling gene p53

Abstract:Aim: To investigate, the effects of paeonol (Pae), extractant of vegetable drug,on apoptosis of colorectal cancer cell line, and analyze its relationship with controlling gene p53. Methods: The experiment was conducted in the Department of Gastroenterology, People's Hospital of Wuhan University between September 2004 and July 2005. Colorectal cancer cell line HT-29 was routinely cultured in culture bottle, which were divided into 15.63 mg/ L, 62.5 mg/L,250 mg/L Pae solution groups and control group after entering into the logarithmic phase. 15.63 mg/L,62.5 mg/L and 250 mg/L of Pae solution (bought from Shanghai First Pharmaceutical Company) as well as same-volume of culture fluid were respectively added. The morphous changes were observed under inverted microscope by HE staining, and the apoptosis was detected with terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) assay and flow cytometry(FCM), so as to qualitatively and quantitatively analyze the relationship between Pae and apoptosis of colorectal cancer cells. The expression of apoptosis associated gene p53 was detected by immunocytochemical staining. Results: 1The apoptotic indexes of colorectal cancer cell line HT-29 in the 15.63 mg/L,62.5 mg/L and 250 mg/L Pae solution groups were respectively (8.16±2.24)%,(15.35±3.07)% and (23.70±3.42)%, which were significantly higher than the control group [(3.32±0.41)%(P<0.01)]. The apoptotic index was in a positive dose-dependent manner with Pae.2 The cell cycle distribution was obviously altered by Pae, representing as a increase of cells at S phase, while cells at G0/G1 phase and G2/M phase were decreased after treated by Pae.3The positive expression rates of p53 gene in colorectal cancer HT-29 cell line were (87.6±10.10)%, (83.7±9.43)% and (77.4±8.22)% respectively in 15.63 mg /L,62.5 mg/L and 250 mg/L Pae solution groups, which were significantly lower than the control group [(90.2±8.04)%(P < 0.05-0.01)], and the effect was in a negative correlation with the concentration of Pae. Conclusion: Pae can induce the apoptosis of colorectal cancer cells at certain concentration, which may be related with its effects on distribution of cancer cell cycle and the down-regulation of expression of p53, representing as a dose-dependent manner.
