Abstract:Objective To explore the inhereditary background of HLA in the Han Nationality of Hunan Province for other associated studies, we observed the distribution of HLA-A locus alleles in population of Hunan Hans. Methods The polymorphism of HLA-A locus allele was determined with ARMS/PCR-SSP method for DNA typing. Direct amplification from genomic DNA and after electrophoresis the result reading was carried out with the method provided by 12th WS(International Histocompatibility Workshop and Conference). Results Our DNA typing data, HLA-A locus allele polymorphism of 66 normal Hunan Hans cases was submitted to and corrected by the central database of 12th WS. The results showed that the allele frequencies of A * 11G1 (27. 10%), A * 02(21.60%), A * 33(12.40%), and A * 24G1 (10.40%) were higher than those of other alleles. G (group) suffix means a set of alleles which cannot be split using the current typing system. Of seven A * 02 subtypes, A * 02 G3(9. 30%), A * 02G1 (8. 20%) and A * 0203(4. 10%) were found in Hunan Hans. Conclusions HLA class I DNA typing was used officially for the first time in 12th WS. The ARMS/PCR-SSP DNA typing can identify directly the subtypes of alleles from genomic DNA and find the single base difference between alleles. When the frequency of each allele in Hunan Hans was compared with the combined world frequency of that allele on 2471 individuals in 45 populations, it could be found that the allele frequencies of A * 02G3, A * 11G1 (P < 0. 001) and A * 33 (P < 0.01) in Hunan Hans were higher significantly than that in combined world, but the allele frequencies of A1 and A3 were lower significantly and that of A11, A24 and A33 higher remarkably in Hunan Hans than that in Swedish. All of these results indicate that there is a distinct inhereditary background among the Han Nationality of Hunan Province, so this result will be the basic and reference data in Hunan Hans.
