Effect of inhibiting NgR expression on growth of neurons cultured in inhibitive microenvironment

Abstract:Objective To detect the effect of inhibiting Nogo receptor(NgR) expression on axon growth of neurons cultured in inhibitive microenvironment.Methods Myelin membrane proteins extracted from rat brain were used as the culture substrate of cortical neurons.Cells were divided into four groups of commonly cultured(group A),myelin membrane proteins(group B),siRNA plus myelin membrane proteins(group C) and scrambled oligonucleotide plus myelin membrane proteins(group D).Cells were transfected with NgR siRNA and then immunostained with monoclonal antibody recognizing the neuron-specific βⅢ-tubulin every day.The length of the longest neurite of each positive neuron was measured.Results During the early days after transfection(24h and 48h),the growth of the cells in group A was the best with the length of the longest neurite of (22.5±12.2)μm and (59.1±23.6)μm,respectively.That in group B was the second with the length of (17.1±10.2)μm and (44.7±22.5)μm.The growth of the cells in groups of B and D were obviously inhibited \[(8.1±5.8)μm and (27.7±13.7)μm for group B and (7.3±5.8)μm and (24.9±12.4)μm for group D).At the time of 96 h after transfection,the differences among the four groups tended to disappear.Conclusion Myelin membrane proteins have obvious inhibitive effect on the growth of the neurite of cortical neurons and the inhibition could be relieved by transfecting the cells with NgR siRNA in vitro.
