17Β-Estradiol Suppresses IL-17 Synthesis of Th17 Cells from Human Peripheral Blood

Abstract:This study is aimed to investigate the regulation effects of 17β-estradiol (E2) on key factors of Th17 cells in human peripheral blood.We detected IL-17A positive cells in human peripheral blood mononuclear cell (PBMC) by flow cytometry.PBMC was treated with E2 and its selective antagonists Tamoxifen and ICI 182.780,and then mRNA levels of the key factors of Th17 cells were measured with real time PCR.The results suggested that the percentages of IL-17A positive cells in PBMC varied from 0.2% to 1.5%;both low and high levels (1 nmol/L and 100 nmol/L) of E2 displayed significant suppression effects on expression of IL-17A and IL-17F,whereas medium level (10 nmol/L) did not.Furthermore,the suppression could not reversed by any antagonists.At the same time,the synthesis of ERα,ERβ,RORγt,RORα and IL-23R were not affected significantly.From these data,we speculated that Th17 cells in individuals with less estrogen level (such as male) may be not as active as that in ones with relative higher levels estrogen (such as female),which may partially explain the female tendency of many autoimmune diseases.
