Abstract:A general one-step synthesis for tris N-alkylated cyclens with good yield and unprecedented selectivity is presented. Tris and 1,4-bis N-alkylated cyclens, as the only two major products are isolated. Furthermore, according to the single crystal X-ray structures of tris and 1,4-bis N-alkylated cyclen 1 and 1a, one nitrogen atom on the cyclen ring can be protonated under this reaction condition, which prevents its further alkylation, and gives rise to the regioselectivity ultimately.

Abstract:A sustainable, cyanide-free synthesis of alkyl nitriles via the aerobic oxidative deconstruction of unstrained cycloalkanones with ammonium salts has been developed. Using inexpensive and stable ammonium salts as the nitrogen source, a variety of alkyl nitriles containing a distal carbonyl group were obtained in good yields under visible-light-promoted iron catalysis. This protocol is characterized by mild conditions, abundant and environmentally benign materials, and high atom and step economy with minimal waste generation. The primary mechanism study revealed that 1O2 is likely to be involved in this reaction.

Abstract:A novel and convenient one-step method for the synthesis of 1,3,4,-Triaryl-substituted-3,4,5,6-tetrahydropyrimidin-2(1H)-ones (3) was developed by cyclization of aromatic isocyanates (1) with 3-arylamino-1-phenylpropan-1-ones (2) under refluxing of toluene. Effects of reaction conditions on the converting ratio and yield were investigated. The method offered several advantages including simple experimental procedure, easily available starting materials and excellent yields (79%similar to 96%).

Abstract:Enantioselective synthesis of (spiro)cyclobutane derivatives poses significant challenges yet holds promising applications for both synthetic and medicinal chemistry. We report here a nickel-catalyzed asymmetric syn-hydrometalative 4-exo-trig cyclization of alkynones to synthesize alkenyl cyclobutanols with a tetrasubstituted stereocenter. This strategy features a broad substrate scope, delivering a variety of trifluoromethyl-containing rigid (spiro)carbocycle skeletons in good yields and high enantioselectivities (up to 84% yield and 98.5:1.5 er). The synthetic utility is demonstrated through stereospecific transformations into fused spirocycles. Experimental and computational mechanistic studies indicate that the reaction is initiated by an active Ni–H species, with carbonyl-directed hydrometalation as the key for regioselective control. This catalytic method provides a general solution for regioselective hydrofunctionalization of alkynes and represents an efficient reaction pattern for assembling highly strained enantioenriched bioisosteres.

Abstract:A convenient synthetic route to 1,4-bis(tert-butoxycarbonylmethyl)tetraazacyclododecane (cyclen) (1) with high yield and excellent regioselectivity is described. Compound 1 reacted with a range of functionalized alkyl halides under two reaction conditions to give mono-N-alkylated 1,4-bis(tert-butoxycarbonylmethyl)tetraazacyclododecane (2-9) in good yield.

Abstract:Herein, we firstly demonstrate an exclusive approach for the preparation of alkynyl cyclopropa[c]coumarin derivatives with high-yield utilizing propargyl sulfonium salts as C1 synthon. Compared with Br-, ClO4- used as the counter anion dramatically improved the yield due to its less nucleophilic ability. This method features mild reaction conditions and a broad substrate scope with good diastereoselectivity.

Abstract:A highly efficient, chemo-, and regioselective approach has been developed for the switchable synthesis of tetrasubstituted alkenyl sulfones and naphthyl sulfones from homopropargylic alcohols via sulfonylation/1,4-aryl migration and sulfonylation/cyclization. The present switchable processes are characterized by mild and metal-free conditions, high selectivities, good functional group tolerance, the use of inexpensive and easily handled sulfonyl hydrazides as sulfonyl sources, and the release of the nontoxic byproducts H2O and N-2.

Abstract:We herein develop a highly diastereoselective synthesis of cyano-substituted cyclopropanes via palladium-catalyzed direct cyanoesterification of cyclopropenes, which features mild reaction conditions, good functional group compatibility, and simple operation. This transformation represents a stepwise, highly atom economic, and scalable protocol for obtaining synthetically useful cyclopropanecarbonitriles.

Abstract:In this paper, we report a facile synthesis of 2-carbamoyl-2-cyanocyclopropanecarboxylates through a tandem iodosobenzene/tetrabutylammonium iodide-induced oxidative cyclization and a subsequent neighboring group-assisted decarboxylation of the Michael adducts of 2-cyanoacetamides with α,β-unsaturated malonates. This method affords the desired highly functionalized cyclopropanes in moderate to good yields and with excellent diastereoselectivities. In addition, the reaction proceeds smoothly under mild conditions and with good functional group tolerance.

Abstract:The double ring-closing metathesis reaction of N-alkynyl-N-(1,omega)-alkadienyl acrylamides 1 using first- or second-generation Grubbs' catalyst afforded, in a highly selectively manner, the fused bicyclic quinolizidine alkaloid derivatives and their analogues bearing a 1,3-diene moiety, which may further undergo a Diels-Alder reaction with a dienophile to afford N-containing polycyclic compounds. The excellent selectivity of fused/dumbbell-mode cyclization has been realized by the higher reactivity of the electron-rich C=C bond or carbon-carbon triple bond combined with the lower reactivity of the electron-deficient C=C bond toward metallocarbenes and the thermodynamically more stable nature of fused bicyclic compounds 3 vs dumbbell-type bicyclic compounds 4.

Abstract:Ir-catalyzed asymmetric tandem allylation/iso-Pictet-Spengler cyclization of arylidenaminomalonates with indolyl allylic methyl carbonates was successfully developed, which provided a direct and practical approach to access synthetically useful and biologically active tetrahydro-γ-carboline derivatives bearing multiple functional groups and stereogenic centers in good to high yields and excellent stereoselective control (44%-96% yields, >20:1 dr, 94% → 99% ee). A wide range of substrate generality, easily available substrates, and simple chiral catalytic system displayed great potential practicality of this efficient protocol.

Abstract:Selective synthesis of [6]-cyclometaphenylene ([6]-CMP) was realized through a one-pot method utilizing a confined nanoreactor. The yield was up to 15%, which was 275% higher than that of homogeneous catalyst. The preliminary derivatization of [6]-CMP led to the production of amphiphilic macrocycles capable of self-assembly in solution, thereby expanding the scope of CMP synthesis and applications.

Abstract:An four-component cascade cyclization reaction between readily available 2,4-pentanediones, primary amines and sodium sulfinates is reported. Highly functionalized 5-alkylidene-3-pyrrolin-2-ones were efficiently synthesized in moderate to excellent yields under mild conditions. This regioselective cascade cyclization was achieved by direct transformation of C(sp3)−H bonds at two weak nucleophilic sites in the enaminones structure, and two strong nucleophilic sites of the structure were not involved.

Abstract:4-Aryl-3,4-dihydrocoumarins are one of the most important structural motifs. Herein, we disclose an enantioselective N -Heterocyclic Carbene catalyzed β-arylation/cyclization of α-bromoenals with 3-aminophenols under mild conditions. The protocol allows for the rapid preparation of 4-aryl-3,4-dihydrocoumarins in acceptable yields with good enantioselectivities. The products of this reaction could be converted into chiral diarylpropanoic acid derivatives without erosion of the enantioselectivity.

Abstract:An efficient asymmetric approach to access functionalized pyrido- and pyrrolo [1,2-c] [1,3]oxazin-1-ones has been developed through a nucleophilic addition-cyclization process of N,O-acetal with ynamides. A number of substituted ynamides 8a-8o and 3-silyloxypyrrolidine or piperidine N,O-acetals 6a, 7 were amenable to this transformation, and the desired products 9a-9o, 10a-10m were obtained with excellent regioselectivities and outstanding diastereoselectivities. Moreover, chiral ynamides 14a-14f could also experience this addition-cyclization process to afford products 15a-15f in excellent yields.
