Synthesis and Antiinflammatory Activities of NO Releasing-Flobufen Derivatives

Abstract:Aim To search for a novel nonsteroidal antiinflammatory drug with high potency and low undesirable effects.Methods Coupling flobufen,a new chemical entity in phase Ⅲ clinical trials,with NO donors having different NO releasing mechanisms,in which NO synthase(NOS)substrate analogues,nitrates and furoxans are included.Evaluating antiinflammatory activities against the xylene-induced mice ear swelling and carrageenan-induced rat paw edema,observing side effects in the rat gastrointestinal(GI)tract and assessing NO releasing ability.Results Nineteen target compounds were synthesized,their structures were determined by MS,IR, 1H-NMR and elemental analysis.The preliminary pharmacological tests showed that 10 target compounds(Ⅰ_1,Ⅰ_ 3～5 ,Ⅰ_8,Ⅰ_ 12 ,Ⅱ_2,Ⅱ_3,Ⅱ_6,Ⅱ_7 )displayed relatively high antiinflammatory activities,among which 5(Ⅰ_1,Ⅰ_3,Ⅰ_4,Ⅱ_3,Ⅱ_6 )possessed activity comparable to flobufen( P 0.05).All the active compounds showed low GI side effects,among which GI lesions of Ⅰ_1,Ⅰ_3,Ⅰ_5 and Ⅱ_7 were significantly less than that of flobufen( P 0.05).Ⅰ_1 and Ⅰ_3 showed comparable antiinflammatory activites to flobufen,while their GI side effects were significantly lower than that of flobufen.Conclusion Compounds Ⅰ_1 and Ⅰ_3 ,which were on the whole superior to flobufen,are worthy to be intensively studied.
