Abstract:Chemical investigation on the aerial parts of Euphorbia fischeriana resulted in the identification of eight abietane-type diterpenoids including three undescribed ones, named 3α-acetoxy-14-hydroxy-ent-abieta-8(9),13(15)-dien-16,12-olide (1), 3α,7β-dihydroxy-ent-abieta-11(12),13(15)-dien-16,12-olide (2), and 2β-hydroxy helioscopinolide B (3). Compound 1 exhibited moderate cytotoxic activities against the HL-60, SMMC-7721, and SGC-7901 cancer cells.

Abstract:Two new acetophenone derivatives and a new sesquiterpene, along with four known acetophenone compounds, were isolated from the roots of Euphorbia ebracteolata. The structures of the new compounds were identified as 2,2',4,4'-tetrahydroxy-6,6'-dimethoxy-3,3'-dimethyl-7,5'-bisacetophenone, 2,4-dihydroxy-6-methoxy-3-methylacetophenone 4-O-alpha-L-arabinofuranosyl-(1-->6)-beta- D-glucopyranoside and decahydro-1a,4a,7,7-tetramethyl-1H-cycloprop[e]azulene-2,5-diol (1a R,2 R,4a S,5 S,7a R,7b R) using 1D and 2D NMR spectroscopy and X-ray crystallography.

Abstract:Two new triterpenoids, namely 24-methylene-5,24-dien-19(10→9)- abeo -8 α ,9 β ,10 α -eupha-3 β -ol ( 1 ) and 24-methyl-5,23-dien-19(10→9)- abeo -8 α ,9 β ,10 α -eupha-3 β -ol ( 2 ) were isolated from the stems of Euphorbia royleana , together with three known analogs. The structures of the new compounds were elucidated by extensive 1 H NMR, 13 C NMR, HSQC, HMBC, 1 H- 1 H COSY, ROESY and HR-MS spectroscopic analyses.

Abstract:Two new ent-abietane diterpenoids, euphcopenoids A (1) and B (2), along with six known analogues (3 - 8), were isolated from the whole plants of Euphorbia helioscopia. Their structures were determined by various spectroscopic methods (MS, UV, IR, and NMR), and the absolute configurations of compounds 1 and 2 were determined by quantum chemical ECD calculations. All compounds were screened for the cytotoxicity against A549 and HEPG2 cancer cell lines, but were inactive. [GRAPHICS]

Moreover. revised structures are proposed for decipinone B (1) and C (2) on the basis of X-ray-diffraction analyses. Abstract:Phytochemical investigation of the roots of Euphorbia ebracteolata Hayata resulted in the isolation of three new rosane diterpenoids, euphebracteolatins C-E (1-3), along with fourteen known analogs (4-17). Their structures were determined on the basis of extensive spectroscopic analysis including HR-ESI-MS, 1D and 2D NMR. Euphebracteolatin C (1) contains a C-1/C-10 double bond and a keto group at C-7, and euphebracteolatins D and E (2-3) possess an aromatic ring-A in their skeleton. The plausible biogenetic pathways of all the isolates were also proposed. Furthermore, compounds 1 and 9 showed selective cytotoxicity against HepG2 cells with IC50 values of 14.29 and 12.33 mu M, respectively, and 2-3 displayed moderate cytotoxicity against three human cancer lines, with IC50 values ranging from 23.69 to 39.25 mu M.

Abstract:Two new ent -labdane diterpenes, ent -3 α ,16-dihydroxylabda-8(17),12( E ),14-triene ( 1 ) and ent -14( S ),15-dihydroxylabda-8(17)-12( E )-dien-18-oic acid ( 2 ), were isolated from the roots of Euphorbia yinshanica . Their structures were elucidated on the basis of spectroscopic methods including HR-ESI-MS and 1D and 2D NMR. The cytotoxic activities against five tumor cell lines was also evaluated, but there was no significant activity.

Abstract:Thirteen diterpenoids (1-13), including two new norditerpene lactones (1-2) and eight new rosane diterpenoids (3-10), were isolated from the roots of Euphorbia ebracteolata. The structures were determined by 1D and 2D NMR, HRESIMS, and electronic circular dichroism (ECD). The ECD-based empirical rule for α,β-unsaturated-γ-lactones was applied to determine the absolute configurations of 1 and 2. Compounds 7, 10, and 13 exhibited significant inhibition of nitric oxide production in RAW 264.7 lipopolysaccharide-induced macrophages, with IC50 values of 2.44, 2.76, and 1.02 μM, respectively.

Abstract:Euphnerins A (1) and B (2), two extremely modified diterpenoids possessing an unprecedented 5/6/6 rearranged spirocyclic carbon skeleton, and a biosynthetically related known diterpenoid (3) were purified from the stems of Euphorbia neriifolia. Their structures were identified by NMR experiments and X-ray diffraction analysis, as well as experimental and calculated electronic circular dichroism data comparison. A putative biosynthetic relationship of 1 and 2 with their presumed precursor 3 is proposed. Compound 1 showed NO inhibitory effects in lipopolysaccharide-stimulated BV-2 cells with an IC50 value of 22.4 μM.
