Abstract:Electrostatic self-assembled films of deoxyribonucleic acid (DNA) and diazoresin (DAR) were fabricated on substrates. Under irradiation, photoreaction occurring between adjacent layers was confirmed by UV-Vis and Fourier transform infrared(FTIR) spectra, which covalently attached DNA to DAR in the multilayer. The stability of the multilayer was greatly improved, which was confirmed by etching in NaCl solution and monitored by UV-Vis and scanning force microscopy (SFM).

Abstract:Poly(N-isopropylacrylamide) (PNIPAAm) grafted with single-stranded (ss) DNA conjugate (PNIPAAm-g-DNA) self-assembles above its lower critical solution temperature to form colloidal particles. When the ssDNA within the particle hybridizes with its complementary DNA, the particles aggregate above a certain threshold of salt concentration with drastically increased turbidity in solution. Detailed structural information of the particle was obtained mainly by small-angle X-ray scattering. The influence of copolymer composition on the morphology of particle and non-crosslinking aggregation was examined. The particle consists of hydrophobic PNIPAAm core surrounded by hydrophilic DNA strands. The increase in DNA fraction brought about a significant decrease in core size, whereas the shell thickness little changed and corresponded to the length of DNA. A structural model with a sticky potential was applied to the analysis of particle aggregate. This analysis provided that the particles aggregate while the coronal layers interpenetrate each other. The interaction between the particles was quantified in terms of the sticky potential and showed a trend to be influenced by the particle size rather than the graft density of DNA strands on the particle.

Abstract:It is reported on the multilayer architecture containing a photo-reactive polyelectrolyte diazo-resins (DAR) as polycation and poly(4-styrene sulfonate) (PSS) as polyanion, held together by electrostatic interaction. Upon UV irradiation, the adjacent interfaces of the multilayer reacted to form a covalently crosslinking structure which greatly improved the stability of the films. These changes were confirmed by UV-VIS and FTIR spectroscopy. The thickness of the covalently attached films was characterized with X-ray diffraction, results from which showed that the thickness of the films can be adjusted in a nanometer scale by adding salts or diluting the solution of DAR. Atomic force microscopy (AFM) experiments showed that both the films before and after UV irradiation were closely compacted as well as smooth, with no obvious changes in morphology to be observed during the photoreaction. It was confirmed that this was an effective way to fabricate photosensitive and then stable layered nanoarchitecture.

Abstract:We reported on the multilayer architecture containing diazo-resins (DAR) as polycations and polyaniline poly(aniline-co-N-propanesulfonic acid aniline) (PAPSAH) as polyanions held together by electrostatic interaction. Upon UV irradiation, the adjacent interfaces of the multilayer reacted to form a covalently crosslinking structure which greatly improved the stability of the films as confirmed by solvent etching experiments. These changes were confirmed by UV-Vis and FTIR spectroscopy. The thickness of the covalently attached films were characterized with small angle X-ray diffraction (SAXD) and a value of 30.0 Å per bilayer was obtained. This type of film was further characterized by cyclic voltammetry which showed that the electroactive property of PAPSAH was still kept in the films after photoreaction.

Abstract:Thin films of alternating DNA and rare earth ion Eu3+ layers from dilute aqueous solutions were fabricated onto quartz substrates and silicon wafers through the layer-by-layer (LbL) self-assembly technique. UV-visible spectroscopy shows that a uniform layer of DNA can be fully adsorbed onto each alternate Eu3+ layer. Microscopic FTIR spectra show Eu3+ interacts with both the phosphate groups and nitrogenous bases of DNA, and the formation of {DNA/Eu}(n) films induces a change of the conformation of the DNA secondary structure to a certain extent. Various parameters affecting the DNA or Eu3+ loading into the composite film were investigated with emphasis on the effect of the pH and ionic strength of the DNA solution used for the film preparation. Atomic force microscopy was utilized to observe the morphologies of the DNA in the films obtained at two different pH values. Small molecules, such as alpha-tenoyltrifluoroacetone (TTA), Hoechst 33258 (Hoe), and ethidium bromide (EB), are all observed to interact with Eu3+ or DNA in the {DNA/Eu}(n) films. The {DNA/Eu}(n) films incorporated with these molecules show different fluorescent characteristics, and the fluorescence intensity of the films versus the bilayer number has a good linear relationship, confirming the potential for creating a different luminescence ability of the multilayer by controlling the number of DNA/Eu bilayers.

Abstract:We describe the formation and structure of nucleolipid/dendrimer multilayer films controlled by non-covalent interactions to obtain biomaterials that exhibit molecular recognition of nucleic acids. Layers of cationic poly(amidoamine) (PAMAM) dendrimers of generation 4 and the anionic nucleolipids 1,2-dilauroyl-sn-glycero-3-phosphatidylnucleosides (DLPNs) based on uridine (DLPU) and adenosine (DLPA) were first formed at the silica-water interface. The PAMAM/DLPN layers were then exposed to short oligonucleotides, polynucleotides and single stranded DNA (ssDNA). The interfacial properties were characterized using quartz crystal microbalance with dissipation monitoring, attenuated total reflection Fourier transform infrared spectroscopy and neutron reflectometry. Both types of DLPN were found to adsorb as aggregates to preadsorbed PAMAM monolayers with a similar interfacial structure and composition before rinsing with pure aqueous solution. Nucleic acids were found to interact with PAMAM/DLPA layers due to base pairing interactions, while the PAMAM/DLPU layers did not have the same capability. This was attributed to the structure of the DLPA layer, which is formed by aggregates that extend from the interface towards the bulk after rinsing with pure solvent, while the DLPU layer forms compact structures. In complementary experiments using a different protocol, premixed PAMAM/DLPN samples adsorbed to hydrophilic silica only when the mixtures contained positively charged aggregates, which is rationalized in terms of electrostatic forces. The PAMAM/DLPA layers formed from the adsorption of these mixtures also bind ssDNA although in this case the adsorption is mediated by the opposite charges of the film and the nucleic acid rather than specific base pairing. The observed molecular recognition of nucleic acids by dendrimers functionalized via non-covalent interactions with nucleolipids is discussed in terms of biomedical applications such as gene vectors and biosensors.

Abstract:Through electrostatic layer-by-layer (LbL) assembly, negatively charged calf thymus double stranded DNA (CTds-DNA), and positively charged Zr4+ ions were alternately deposited on gold substrate modified with chemisorbed cysteamine. Thus-prepared three-dimensional DNA networks were characterized by surface plasmon resonance (SPR) spectroscopy, X-ray photoelectron spectroscopy (XPS) and infrared reflection–absorption spectroscopy (IR-RAS). SPR spectroscopy indicates that the effective thickness of DNA monolayer in the (DNA/Zr4+)1 bilayer was 1.5±0.1nm, which corresponds to the surface coverage of 79% of its full packed monolayer. At the same time, a linear increase of film thickness with increasing number of layers was also confirmed by SPR characterizations. The data of XPS and IR-RAS show that Zr4+ ions interact with both the phosphate groups and nitrogenous bases of DNA and load into the framework of DNA. Furthermore, the interactions between this composite film and heme protein cytochrome c (Cyt c) were investigated by SPR spectroscopy and electrochemistry. Compared with the adsorption of Cyt c on DNA monolayer, this composite multilayer film can obviously enhance the amount of immobilized Cyt c confirmed by SPR reflectivity-incident angle (R-θ) curves. Cyclic voltammetry (CV) indicates the Cyt c adsorbed on the composite film is electroactive, and the enhancement of peak current in CV indirectly verifies the increase of the amount of immobilized Cyt c.

Abstract:In this work, we illustrate a simple chelation-based strategy to trigger DNA release from DNA-incorporated multilayer films, which were fabricated through the layer-by-layer (LbL) assembly of DNA and inorganic zirconium (IV) ion (Zr4+). After being incubated in several kinds of chelator solutions, the DNA multilayer films disassembled and released the incorporated DNA. This was most probably due to the cleavage of coordination/electrostatic interactions between Zr4+ and phosphate groups of DNA. Surface plasmon resonance (SPR), UV–vis spectrometry and atomic force microscopy (AFM) were used to characterize the assembly and the disassembly of the films. By incorporating plasmid DNA (pDNA) into this controllable disassembly system, the multilayer films sustained the consecutive DNA release. The released pDNA retained its integrity and transcriptional activity, and also expressed enhanced green fluorescent protein (EGFP) after being transfected into HEK 293 cells. Besides the simplicity and cost efficiency of this method, the most advantage of this route was that the release of DNA from the films could be modulated by various external conditions, such as the chelator and ionic strength. The Zr4+/DNA multilayer films with the ability to precisely control the release rate of DNA might serve as an alternative localized gene delivery system in the perspective of biomedical applications.
The Assembly of DNA Amphiphiles at Liquid Crystal-Aqueous Interface

Abstract:In this article, we synthesized a type of DNA amphiphiles (called DNA-lipids) and systematically studied its assembly behavior at the liquid crystal (LC)—aqueous interface. It turned out that the pure DNA-lipids at various concentrations cannot trigger the optical transition of liquid crystals from planar anchoring to homeotropic anchoring at the liquid crystal—aqueous interface. The co-assembly of DNA-lipid and l-dilauroyl phosphatidylcholine (l-DLPC) indicated that the DLPC assembled all over the LC-aqueous interface, and DNA-lipids prefer to couple with LC in certain areas, particularly in polarized and fluorescent image, forming micron sized net-like structures. The addition of DNA complementary to DNA-lipids forming double stranded DNA-lipids caused de-assembly of DNA-lipids from LC-aqueous interface, resulting in the disappearance of net-like structures, which can be visualized through polarized microscope. The optical changes combined with DNA unique designable property and specific interaction with wide range of target molecules, the DNA-lipids decorated LC-aqueous interface would provide a new platform for biological sensing and diagnosis.

Abstract:The layer-by-layer (LbL) self-assembly of poly-l-lysine (PLL) and deoxyribonucleic acid (DNA) was used to construct the enzymatic biodegradable multilayered films. The LbL build up of DNA multilayers was monitored by UV–vis spectrometry, and atomic force microscopy (AFM). AFM, UV–vis spectrometry and fluorescence spectrometry measurements indicated that 90% of DNA within the films was released almost linearly under 5UmL–1 α–chymotrypsin in PBS at 37°C in 35h. TEM and zeta potential experiments revealed that the released DNA molecules were condensed into the slight positive complexes with size from 20 to several hundred nanometers. The well-structured, easy processed enzymatic biodegradable multilayered film may have great potential for gene applications in tissue engineering, medical implants, etc.

Abstract:DNA-mediated interactions present a significant opportunity for controlling colloidal self-assembly. Using microcontact printing to achieve spatial control of DNA-surface patterning and DNA-functionalized polystyrene colloids, we report that DNA hybridization can be utilized for sequence-specific reversible selfassembly of well-ordered 2D colloidal arrays. Two essential indicators of DNA-hybridization mediated assembly were confirmed: thermal reversibility and sequence specificity. The arrays melted at 50 degrees C and reassembled when introduced to fresh colloid suspension, and sequence specificity with < 1 % nonspecific binding was confirmed using fluorescent polystyrene colloids. The real-time assembly of the colloids onto the periodically patterned substrate was monitored by simple laser diffraction to obtain assembly kinetics. Maximum surface coverage of DNA-mediated assembly was determined to be 0.593 for DNA-functionalized 100 nrn polystyrene colloids, and 90% of the assembly was complete after 6.25 h of hybridization in 50 mM NaCl Tris buffer. We also demonstrate that DNAzymes, catalytic DNA molecules, can be incorporated into the design, and in the presence of 10 mu M Pb2+, the hybrid ization-incluced array assembly can be disrupted via DNAzyme activity.
DNA Assembly on 2-Dimensional Array of Colloidal Gold

Abstract:The primary object of this contribution is to self-assemble DNA on 2-dimensional array of 1-dodecanethiol-encapsulated colloidal gold (2-DACG). The 2-DACG film was deposited by Langmuir-Blodgett (LB) technique. Through phase transfer of aqueous colloidal gold particles into ethanol solutions containing 1-dodecanethiol, the hydrophobic gold colloidal nanoparticles were accomplished. In the presence of traces of 1-dodecanethiol, the gold nanoparticels were transferred via LB technique to a freshly cleaned gold electrode and 2-DACG of close-packed, ordered monolayer was obtained. The 2-DACG was then used as a substrate for the immobilization of DNA under a controled potential of + 0.20 V vs. saturated calomel electrode (SCE). The properties of 2-DACG before and after the DNA assembly were investigated by means of transmission electron microscopy (TEM), atomic force microscopy (AFM), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The results indicated that 2-DACG deposited by LB technique was suitable as a substrate for DNA self-assembly.

Abstract:Studies of the deoxyribonucleic acid (DNA) molecule at surfaces are driven by both the inherent interest in understanding different aspects of this molecule and its importance in biomimetic materials science and molecular electronics. In this paper, we fabricate binary DNA/surfactant-modified DNA arrays on a heterogeneous patterned surface through twice controlled condensation and dewetting processes. Through the muCP technique, we created a hydrophobic/hydrophilic pattern, using a condensed microdroplet array as a template to fabricate a porous film of polystyrene (PS). With similarity to the "membrane-based patterning" method, this PS porous film was used as a mask, exposed hydrophilic regions inducing DNA to form as an array. After the PS porous films are peeled off, blocked surface sites emerged for the subsequent adsorption with other species. Through another condensation process, where a DNA aqueous solution droplet array could be used as a template, surfactant-modified DNA was sited in the ordered hydrophobic regions through a second dewetting process and hydrophobic interaction, and we fabricated binary arrays on the same surface. At the same time, the double-helical structure and function of DNA still remained. This provides a simple, flexible, and efficient patterning technique for biomolecules.
Connection of ssDNA to Silicon Substrate Based on a Mechano-Chemical Method

Abstract:A novel fabrication process to connect single-stranded DNA (ssDNA)to a silicon substrate based on a mechano-chemical method is proposed. In this method, the single crystal silicon substrate was mechanically scribed in a diazonium solution of benzoic acid using a diamond tip which formed silicon free radicals. These combined covalently with organic molecules of diazonium benzoic acid contained in the solution to form self-assembled films (SAMs). The SAMs were characterized and analyzed by AFM, X-ray photoelectron spectroscopy and infrared spectroscopy. The results showed that the self-assembled films were covalently connected to the silicon substrate by Si-C. In this way, a nano-level benzoic acid coupling layer was self-assembled on the scribed area of the silicon substrate. The ssDNA was further covalently connected to the silicon surface by the coupling layer. Fluorescence microscopy showed that ssDNA had been connected, and the influence of ssDNA concentration on the fixation effect was studied. The fluorescence brightness gradually increased with the gradual increase in ssDNA concentration from 5 μmol/L to 15 μmol/L, indicating that the fixed amount of ssDNA increased. However, when the concentration of ssDNA increased from 15 μmol/L to 20 μmol/L, the detected fluorescence brightness decreased, indicating that the hybridization amount decreased. The reason may be related to the spatial arrangement of DNA and the electrostatic repulsion between DNA molecules. It was also found that ssDNA junctions on the silicon surface were not very uniform, which was related to many factors, such as the inhomogeneity of the self-assembled coupling layer, the multi-step experimental operation and the pH value of the fixation solution.
Synthesis and Self-Assembly of Dna-Aliphatic Polyether Dendron Hybrids

Abstract:A new kind of amphiphilic DNA-aliphatic polyether dendron hybrids consisting of a flexible hydrophobic polyether dendron and a single stranded DNA are synthesized, which are characterized by MALDI-TOF mass spectroscopy, HPLC and polyacrylamide gel electrophoresis (PAGE). In aqueous solution, as DNA length shortens from 24 mer to 18 mer, to 12 mer, to 6 mer, the hydrophilic DNA content in the DNA-aliphatic polyether dendron hybrid decreases, the morphology of the aggregates change from spherical micelles to nanofibers, and to irregular clusters. These different assemblies from DNA-aliphatic polyether dendron hybrids in aqueous solution are depended on the hydrophobic/hydrophilic ratio between the polyether dendron skeleton and DNA strand. However, when adding 1/10 (VIV) organic solvents such as dichloromethane (DCM), diethyl ether (EtOEt) or tetrahydrofuran (THF) into aqueous solution and after the assembling process that the sample solution is heated to 90 degrees C for 30 min and subsequently cooled to room temperature overnight, the third generation dendron conjugated 18 mer DNA hybrid could assemble into nanofibers. Meanwhile, in the THF/H2O (1: 10, V/Y) mixed solvents, with the same assembling process, as different dendron generations (the second or third generation) and different DNA lengths (6 mer, 12 mer, 18 mer or 24 mer) in the hybrids, all these hybrids could assemble into long nanofibers. The assembled structures have been characterized by transmission electron microscopy (TEM), atomic force microscope (AFM), dynamic light scattering (DLS) and fluorescent experiments. Subsequently, we verified the assembling mechanism that the spherical micelles and nanofibers contain a hydrophobic dendron core and a hydrophilic DNA shell by hydrophobic fluorescent molecule Nile Red encapsulation experiment and precise DNA hybridization to load gold nanoparticles at a size of 10 nm. The hybridization property of DNA at the shell associated with the encapsulation ability of dendron at the inner core indicated the potential application in gene or drug delivery and nanotechnology.

The stretched DNA molecules were visualized by the fluorescent microscope. The stretched DNA was also evidenced by the silver reduction along DNA backbones with hydroquinone in both alkaline and acidic conditions. The scanning electron microscope (SEM) shows that the unstretched DNA molecules result in random silver coils, whereas the stretched DNA molecules give the straight silver wires of more than 10 mu m long. The molecular combing technique was successfully demonstrated in bridging single DNA molecule over the gold microelectrodes, which is observed by the fluorescent microscope. The DNA anchoring on the gold surface was enhanced by the aminoethanethiol modification and by oligonucleotide hybridization. The aminoethanethiol modification of the electrode surface was analyzed by a Physical Electronics PHI 7200 ToF-SIMS Spectrometer and a reflectron ToF analyzer. A more sophisticated DNA 1-D structure was also achieved by the DNA self-assembly and visualized by transmission electron microscope (TEM) with the negative staining. These achievements suggest a useful approach of converting biological macromolecules to potential applications in micro electronic industry.
Creation of Quasi‐DNA Origami at the Air/Water Interface Using the Collapsed Mechanism of Langmuir Monolayer

Abstract:It has created quasi‐DNA origami by utilizing the disintegration behavior of monolayer at the air/water interface of a diamino‐s‐triazine‐based polymers. Langmuir monolayer of polymers with a single layer of DNA adsorbed was collapsed by overcompression, forming a folded structure. The prepared quasi‐DNA origami was characterized for its morphology and molecular arrangement. It has achieved the preparation of quasi‐deoxyribonucleic acid (DNA) origami using the techniques of organized molecular films. After the adsorption of DNA from the subphase onto a Langmuir monolayer of polymers containing diamino‐s‐triazine rings, the monolayer collapses owing to high surface pressure, resulting in the formation of a three‐dimensional folded structure of DNA. A homopolymer featuring a diamino‐s‐triazine ring and a copolymer containing 20 % long alkyl chains are used to form the Langmuir monolayer. A‐T base pairs of salmon testis‐derived DNA from the subphase are hydrogen‐bonded and adsorbed onto each Langmuir monolayer. Following the collapse of the Langmuir monolayer, the film height almost doubles, indicating the formation of a bilayer structure consisting of a bioadsorbed polymer monolayer, especially evident in copolymer‐collapsed films. During this period, the bioconjugated bilayer structure adopts a mesoscopic linear morphology within the plane. In this quasi‐DNA origami structure, the chirality in the DNA molecule is maintained, as evidenced by the circular dichroism (CD) spectra displaying a positive Cotton effect. These results correspond to the proposal of a new DNA morphology control technology.
