Abstract:OBJECTIVE To study the chemical constituents of the whole plant of Potentilla discolor Bunge. METHODS Repeated chro-matographies of silicagel column, highly porous resin R-A,and Sephadex LH - 20 were employed for the isolation and purification. The structures of the constituents were elucidated by the spectral analysis.RESULTS Six compounds were identified as ursolic acid (Ⅰ) ,2α,3β-di-hydroxyl-urs-12-en-28-oic acid(Ⅱ), euscaphic acid(Ⅲ), tormentic acid(Ⅳ),β-sitosteriol-3-O-β-D-glucopyranoside(Ⅴ) and β-sitosterol (Ⅵ) .CONCLUSION All compounds are isolated from this plant for the first time.

Abstract:Context: Potentilla discolor Bge. is a plant extensively utilized in Traditional Chinese Medicine (TCMs). Nevertheless, our knowledge of the chemical components and metabolic complexities in its extracts is still quite limited. Objective: The study aimed to thoroughly analyze the flavonoid metabolism in Potentilla discolor Bge. extracts and conduct a pharmacodynamic evaluation. Methods: In this study, we investigated the metabolism of flavonoids in Potentilla discolor Bge. extracts using SD rats. The research adopted the strategy of “in vivo metabolism assessment, basic study of pharmacodynamics and preliminary evaluation of molecular docking” to systematically investigate the pharmacodynamic substances and targets of action of Potentilla discolor Bge.. Results: The results of the study showed that the main metabolic forms of Potentilla discolor Bge. in vivo include hydroxylation, methylation, and glycosylation. Among them, luteolin in the total flavonoids of Potentilla discolor Bge. has the strongest antimicrobial and antioxidant ability. Outcomes of molecular docking experiments indicated that the glycosylated metabolite of luteolin had a significant advantage in acting with Glp-1 (Glucagon-like peptide-1). Conclusion: The present study revealed the metabolic pathways of total flavonoids in Potentilla discolor Bge. It not only effectively screened out their pharmacodynamic substances and targets but also provided a theoretical basis for the application of TCMs into a systematic application and also provided ideas for the direction of drug optimization in the future. However, the sample range of this study is limited, and it generalizability needs to be investigated.

Abstract:Potentilla species have been used for treatment of several ailments (ulcer, respiratory problems or diarrhea) in different countries. The different solvent extracts (ethyl acetate, methanol, and water) of P. anatolica were studied for antioxidant properties, enzyme inhibitory activity, and phytochemical profile. Phytochemical profile was evaluated as total phenolic, flavonoid, saponin, triterpenoid content, and fatty acid composition. The ethyl acetate extract contained the richest concentration of total phenolics, saponins and triterpenoids. Methanol extract contained high levels of flavonoids. Ethyl acetate and methanol extracts had higher levels of total triterpenoids. The P. anatolica oil contained palmitic acid (29.21%) as major fatty acid, followed by oleic acid (21.33%), and stearic acid (10.79%). The antioxidant properties of extracts were also evaluated by using free radical scavenging(DPPH and ABTS), reducing power (FRAP and CUPRAC), metal chelating activity and phosphomolybdenum assays. All extracts exhibited antioxidant capacity. The best results of P. anatolica were noted in ethyl acetate extract (except total flavonoids, CUPRAC, metal chelating activity). The extracts of P. anatolica demonstrated potent inhibitory activity against α-amylase and α-glucosidase, AChE and tyrosinase and no activity against BChE. P. anatolica could be a potential natural source of antioxidant.

Abstract:An abietane diterpenoid, triptobenzene Y and six sesquiterpene polyol esters, wilforsinines C–H, together with 14 known compounds, have been isolated from the roots of Tripterygium wilfordii. The structures of the compounds were elucidated on the basis of spectroscopic analyses. The quinone reductase (QR) induction assay indicated that two compounds showed moderate QR-inducing activities at concentrations of 25μM and 50μM, respectively.

Abstract:Ultraviolet-B (UV-B) radiation is harmful to plants and human beings. Many secondary metabolites, like flavonoids, alkaloids, and lignin, are UV-B absorbing compounds, which can protect the genetic material of plants. Furthermore, they are active components of herbal drugs. UV-B radiation can activate the self-protective secondary metabolism system. The results of this paper provide a method to induce bioactive secondary metabolites from mulberry leaves (Morus alba L.) by UV-B irradiation in vitro. Five significantly different chromatographic peaks were found by HPLC fingerprint after induction, from which two active compounds were identified: One was chalcomoracin, a natural Diels-Alder type adduct with antibacterial activity; the other one was moracin N, which is a precursor of chalcomoracin. Their contents were 0.818 mg/g and 0.352 mg/g by dry weight, respectively.

Abstract:Marine-derived fungi of the genus Penicillium represent a huge potential for synthesizing the secondary metabolites with structural and bioactive uniqueness and diversity. In this study, six previously undescribed compounds peniciphenalenins A-F and four known compounds (+)-sclerodin, (+)-scleroderolide, (+)-sclerodione, and physcion were isolated from the culture of a marine-derived fungus Penicillium sp. ZZ901. Structures of the isolated compounds were elucidated by a combination of extensive NMR spectroscopic analysis, HRESIMS data, optical rotation value, ECD calculation, and single crystal X-ray diffraction. Peniciphenalenins A-C are the second examples of the type of neoherqueinones. The possible biosynthetic route of nine phenalenone derivatives has been suggested. The known (+)-scleroderolide showed both antiproliferative activity against glioma cells with IC50 values of 23.24-37.26 mu M and antibacterial activity in suppressing the growth of methicillin-resistant Staphylococcus aureus and Escherichia call with MIC values of 7.0 and 9.0 mu g/mL, respectively. Abstract:Background Potentilla discolor Bunge (PDB) is a plant of the Rosaceae family. A wide range of pharmacological activities, such as antibacterial, hypoglycemic, and others, can be achieved through its chemical components, which include terpenes, steroids, polyphenols, and flavonoids. However, the effects of the herb on the heart are unclear. Purpose To explore the effect of the main components of PDB on myocardial cell membranes. Materials and Methods We investigated the role of different concentrations of PDB in cardiomyocytes by evaluating myocardial electrical activity. We used high-performance liquid chromatography to analyze the components. Patch-clamp experiments were conducted to detect the drugs’ impact on the ion channels of cardiomyocytes. Results PDB regulated 14 potential metabolites in mice. Interestingly, quercetin exerts certain effects on the heart by acting on the inositol phosphate metabolism pathway. Quercetin is a monomer compound that can enhance the activity of IK1 on the myocardium membrane. Conclusion This study showed that monomeric compounds of PDB can significantly enhance IK1 activity and influence ion channels in a concentration-dependent manner.

Abstract:Five new compounds, including two sesquiterpenoid glycosides ( 1 and 2 ), two monoterpenoid glycosides ( 3 and 4 ), and a quinovose ester ( 5 ), together with four known compounds ( 6–9 ) were isolated from branches and leaves of Pittosporum pulchrum Gagnep. Their structures were established by 1D and 2D NMR, HR-ESI-MS, IR and UV spectral analyses. This is the first time to investigate the chemical constituents of P. pulchrum . Subsequently, the anti-inflammatory and antioxidant activities of different solvent fractions of ethanol extract and isolated compounds were evaluated. Dichloromethane and ethyl acetate fractions dramatically inhibited the production of NO in a concentration-dependent manner in LPS-induced RAW264.7 cells. Ethyl acetate and n -butanol fractions showed excellent DPPH radical scavenging activities with IC 50 values of 24.31 μg/mL and 27.81 μg/mL, respectively. Compounds 7 and 8 might be potential natural antioxidants with IC 50 values of 16.13 μM and 24.81 μM, respectively.

Abstract:Three new sorbicillinoids sorbicatechols E -G ( 1 – 3 ), along with seven known compounds 4 – 10 , were obtained from the ethanol extract of Penicillium sp. HS-11, a fungal endophyte of the medicinal plant Huperzia serrata . The structures of 1 – 3 were established by detailed interpretation of the spectroscopic data and their absolute configurations were established by comparative analyses of the ECD spectra. Sorbicatechol G ( 3 ) represented the first hybrid sorbicillinoid bearing a tetralone skeleton. In the in-vitro bioassay, trichodimerol ( 5 ) exhibited moderate inhibitory activity against the Escherichia coli β -glucuronidase (EcGUS) with an IC 50 value of 92.0 ± 9.4 μM.

Abstract:Four new compounds ( 1 – 4 ) along with twelve known compounds ( 5 – 16 ) were isolated from the fermentation broth of two desert endophytic fungi Neocamarosporum sp. HM 175 and Alternaria sp. HM 213. The structures of the compounds were elucidated by ESI-MS and NMR data. The PTP1B inhibitory activity results showed that compounds 2 , 5 , 11 , 12 , and 13 exhibited certain inhibitory activity on PTP1B. The activity test of these compounds provided potential drug lead compounds for the research and development of new PTP1B inhibitors.

Abstract:Chemical examination of an unidentified sponge associated marine fungus Penicillium terrestre resulted in the isolation of eight compounds. Their structures are identified as cyclo (glycyl-D-proline) (1), cyclo (L-prolyl-L-alanine) (2), cyclo (L-prolylD-alanine) (3), cyclo (L-leucyl-trans-4-hydroxy-L-proline) (4), cyclo (L-leucyl-cis-4-hydroxy-D-proline) (5), cyclo (L-trans-(4-hydroxyprolinyl)-L-phenylalanine) (6), hexylitaconic acid (7), and hexylitaconic methylate (8), based on spectroscopic analysis (1D, 2D NMR). Compounds 7, 8 were obtained from the genus Penicillium for the first time, while compounds 1-6 were isolated from the fungus Penicillium terrestre for the first time.

Abstract:Herbaceous peony (Paeonia lactiflora Pall.) is a traditional flower, famous in China, which owns highly ornamental value due to its various colors, especially double-colored species. However, the researches on color formation of double colored are rare reported. In this study, the P. lactiflora variety ‘Hebao Jinlian’ with red-yellow double colored petals was used as materials. The anatomical structure and flavonoid components were observed and determined. It was found that colored cells were mainly distributed in the upper and lower epidermis, six kinds of flavonoid components were identified. The total amount of anthocyanins and anthoxanthins decreased gradually with the growth of flowers, and the content in the outer petals was higher than that in the inner petals. Furthermore, by transcriptome sequence, the differential expression genes (DEGs) of outer and inner petals were enriched in flavonoid biosynthesis, phenylpropane biosynthesis and so on at four developmental stages. The accuracy of transcriptome sequence was verified by the expression pattern of DEGs detected by quantitative real-time polymerase chain reaction (qRT-PCR). Combined the analyses of flavonoid assay with transcriptome sequence, the synergistic effect of chalcone synthase (CHS), flavonoid 3-hydroxylase (F3H) and anthocyanindin synthase (ANS) may be the main reason for the formation of red outer petals, while the expression of chalcone isomerase (CHI) and flavonol synthase (FLS) were conducive to the formation of yellow inner petals. The results will lay a foundation for clarifying the color mechanism of double colored petals of P. lactiflora, and also provide a reference for the study of flower color in other ornamental plants.

Abstract:Populus trees meet continuous difficulties from the environment through their life cycle. To warrant their durability and generation, Populus trees exhibit various types of defenses, including the production of secondary metabolites. Syntheses derived from the shikimate-phenylpropanoid pathway are a varied and plentiful class of secondary metabolites manufactured in Populus. Amongst other main classes of secondary metabolites in Populus are fatty acid and terpenoid-derivatives. Many of the secondary metabolites made by Populus trees have been functionally described. Any others have been associated with particular ecological or biological processes, such as resistance against pests and microbial pathogens or acclimatization to abiotic stresses. Still, the functions of many Populus secondary metabolites are incompletely understood. Furthermore, many secondary metabolites have therapeutic effects, leading to more studies of secondary metabolites and their biosynthesis. This paper reviews the biosynthetic pathways and therapeutic impacts of secondary metabolites in Populus using a genomics approach. Compared with bacteria, fewer known pathways produce secondary metabolites in Populus despite P. trichocarpa having had its genome sequenced.

Abstract:Two new flavonoids, Alpinetin-7-O-[3''- O -galloyl-4'',6''-hexahydroxydiphenoyl]- β - D -glucose ( 1 ), Pinocembrin-7- O -[4''- O -galloyl]- β - D -glucose ( 2 ), and three known compound ( 3-5 ) were isolated from Penthorum chinense Pursh (Saxifragaceae). The structures of all compounds were deduced from their comprehensive spectroscopic analysis including IR, HR-ESI-MS, 1 H NMR, 13 C NMR, DEPT, COSY, HMBC and HMQC. Molecular docking model was used to test the anti-cholestatic liver activities of the isolated compounds, and compounds 1 and 2 showed higher docking scores (-9.90 and −11.27 kcal/mol, respectively) binding with FXR than 3 , 4 and 5 (-8.00, −9.77, −9.10 kcal/mol, respectively), suggesting 1 and 2 exhibited potential anti-cholestatic activities. The present results show that P. chinense is a potential source of 2 new lead compounds that can be utilised to produce therapeutic drugs for liver diseases upon further studies.

Abstract:Eight new compounds (1-8), along with three known related compounds (9-11) were isolated from the leaves of Sauropus spatulifolius Beille. Their structures and configurations were elucidated by means of spectrometric and the modified Mosher's method. Among the new compounds, compounds 1 and 2 were identified as ethyl 3, 6-anhydro-2-deoxy-β-D-arabino-hexofuranoside (1) and ethyl 3, 6-anhydro-2-deoxy- hexofuranoside (2). Compounds 3-5 were the 2-acetylpyrrole derivatives and identified as 2-(2-acetyl-1H-pyrrol-1-yl)-4-hydroxybutyric acid (3), methyl 4-(2-acetyl-lH-pyrrol- 1-yl) butanoate (4) and 1, 4-bis (2-acetyl-1H-pyrrol-1-yl) butane (5), respectively. Compound 6 was elucidated as 7-megastigmane-3, 8, 9-triol. Compounds 7, 8 were identified as kaempferol-3-O-2-deoxy-β-D-glucoside (7) and kaempferol-3-O-β-D- glucopyranosyl-(1-6)-2-deoxy-β-D-glucoside (8). In addition, the cytotoxic activities of all the compounds were also evaluated, where compounds 3, 5, 7, 9\10 and 11 exhibited the magnificent inhibition activity on lung fibroblast differentiation induced by TGF-β1with low toxicity against the RLE-6TN cell.

Abstract:The secondary metabolites of the endophytic fungus Talaromyces malicola hosted in the arthropod Armadillidium vulgare were separated by silica gel column chromatography, gel column chromatography, and semi-preparative high-performance liquid chromatography. Eleven compounds(1-11) were obtained from the ethyl acetate fraction of the fermentation broth of T. malicola, and their structures were identified by NMR, HR-ESI-MS, UV, IR, and ECD. The 11 compounds were talarosesquiterpene A(1),(3β,5α,6α,15α,22E)-5,6-epoxyergosta-8(14),22-diene-3,7,15-triol(2), vermistatin(3), hydroxyvermistatin(4), bercheminol A(5), penicillide(6), lunatinin(7), penipurdin A(8), emodin(9), BE-25327(10), and(-)-regiolone(11). Compound 1 was a new diaporol-type sesquiterpene. Compounds 2, 4-5, and 7-11 were isolated from Talaromyces for the first time.

Abstract:A joint phytochemical investigation on the MeOH extracts of the twigs and needles of two endangered Pinaceae plants endemic to the Chinese Qinling Mountains, Picea neoveitchii (an evergreen spruce) and Larix potaninii var. chinensis (a deciduous larch), led to the isolation and characterization of 34 and 24 structurally diverse terpenoids, respectively. Among them, seven are previously undescribed, including a picane-type [i.e., 14(13 → 12)abeo-12αH-serratane] (neoveitchin A) and a serratane-type (neoveitchin B) triterpenoids, and an abietane-type (neoveitchin C) as well as four labdane-type (potalarxins A–D) diterpenoids. Their structures and absolute configurations were established by extensive spectroscopic methods and/or X-ray diffraction analyses. All isolates were evaluated for their inhibitory activities against the human protein tyrosine phosphatase 1B (PTP1B). Serrat-14-en-3α,21β-diol, betulinic acid, 3β-hydroxy-11-ursen-13(28)-olide, ursolic acid, and oleanolic acid were found to have considerable inhibitory effects against PTP1B, with IC50 values ranging from 1.1 to 18.1 μM. The interactions of the bioactive triterpenoids with PTP1B were thereafter performed by employing molecular docking studies. In addition, 7-oxo-dehydroabietic acid (an abietane-type diterpenoid) and mangiferonic acid (a cycloartane-type triterpenoid) inhibited acetyl-coenzyme A carboxylase 1 (ACC1), with IC50 values of 3.4 and 6.6 μM, respectively.
