Abstract:Zinc (Zn) has antibacterial and antifungal properties, so, it is being widely used in food and pharmaceutical industries. Transition metal especially Zn deficiency has deleterious effects all over the world due to wide consumption of Zn deficient food byproduct. Several studies have reported that transition elements especially Zinc deficiency affects the endocrine and reproductive system. This overview comprises the information about negative effects of Zn deficiency on reproductive system of male rat and their possible outcomes. Scientific studies indicated that Zn deficiency severely affects the reproductive physiology of male rats. Zn deficiency suppresses the function of male rat testes inclosing spermatogenesis and steroidogenesis by reducing the production of androgen hormone. In addition, Zn deficient rats also indicated testicular atrophy, primary and secondary spermatogonial stem cells disruption, lower number of sperm cells count, sperm quality, motility, viability, enhance in oxidative stress, inhibition of Testosterone (T) level which, in turn may cause infertility. The different consequences in animal models by zinc application are dose and duration dependent. Several studies propose that both higher and lower Zn concentration can effects the rat reproductive function. Therefore, the available outcomes revealed that Zn deficiency might have significant influence on luteinizing hormone and follicle stimulating hormone affecting the sperm physiology and histology in certain tissues.
Effects of Zinc on Growth Performance and Biochemical Parameters of Piglets

Abstract:The effects of zinc on growth performance, activities of enzymes, and concentrations of hormones were investigated in this study. Crossbred (Duroc x Landrace x Large White) piglets (initial body weight: 26.66 +/- 2.45 kg) were randomly assigned into 4 groups receiving a basal diet and were supplemented with 60, 300, 1000, or 3000 mg/kg of zinc (as ZnSO4 center dot 7H2O). The results showed that supplementation of zinc at 3000 mg/kg increased (P < 0.05) the average daily feed intake, weight gain, gain/feed, and relative organ weight, and promoted the activities of alkaline phosphatase and Cu-Zn superoxide (P < 0.05) by 84.17% and 49.86%, respectively, when compared with the control group. It also led to higher hepatic metallothionein content and tissue zinc concentration (P < 0.05) than in the control group. Furthermore, the concentrations of insulin and neuropeptide Y (NPY) in the 3000 mg/kg group were increased (P < 0.05) by 55.19% and 26.53%, respectively, compared to the control group. The present study suggests that pharmacological dietary supplementation of zinc increased growth performance, activities of enzymes, tissue zinc concentrations, and plasma insulin and NPY concentrations.

Abstract:An experiment was conducted to investigate the effects of zinc glycinate (Zn-Gly) supplementation as an alternative for zinc sulphate (ZnSO4) on productive and reproductive performance, zinc (Zn) concentration and antioxidant status in broiler breeders. Six hundred 39-week-old Lingnan Yellow broiler breeders were randomly assigned to 6 groups consisting of 4 replicates with 25 birds each. Breeders were fed a basal diet (control group, 24 mg Zn/kg diet), basal diet supplemented with 80 mg Zn/kg diet from ZnSO4 or basal diet supplemented with 20, 40, 60 and 80 mg Zn/kg diet from Zn-Gly. The experiment lasted for 8 weeks after a 4-week pre-test with the basal diet, respectively. Results showed that Zn supplementation, regardless of sources, improved (P < 0.05) the feed conversion ratio (kilogram of feed/kilogram of egg) and decreased broken egg rate, and elevated (P < 0.05) the qualified chick rate. Compared with the ZnSO4 group, the 80 mg Zn/kg Zn-Gly group significantly increased (P < 0.05) average egg weight, fertility, hatchability and qualified chick rate, whereas it decreased (P < 0.05) broken egg rate. The Zn concentrations in liver and muscle were significantly higher (P < 0.05) in 80 mg Zn/kg Zn-Gly group than that in ZnSO4 group. Compared with ZnSO4 group, 80 mg Zn/kg Zn-Gly group significantly elevated (P < 0.05) the mRNA abundances of metallothionein (MT) and copper-zinc superoxide (Cu-Zn SOD), as well as the Cu-Zn SOD activity and MT concentration in liver. Moreover, the 80 mg Zn/kg Zn-Gly group had higher (P < 0.05) serum T-SOD and Cu-Zn SOD activities than that in the ZnSO4 group. This study indicated that supplementation of Zn in basal diet improved productive and reproductive performance, Zn concentration and antioxidant status in broiler breeders, and the 80 mg Zn/kg from Zn-Gly was the optimum choice for broiler breeders compared with other levels of Zn from Zn-Gly and 80 mg/kg Zn from ZnSO4.

Abstract:Infertility has become one of the most common chronic diseases among reproductive- aged individuals, and male factors account for about 50 %. Zinc is a trace element that is essential for sperm quality and male reproductive system. Although several current studies with small sample sizes have investigated this relationship, the conclusions have been still controversial. Here, using a large population sample involved 25,915 participants from Changzhou Maternity and Child Health Care Hospital, we revealed an inverted "U"-shaped trend between seminal plasma zinc concentrations and sperm motility PR/PR + NP (PR, progressive motility; NP, non-progressive motility). The results showed that the highest values of sperm PR/PR + NP observed in the group with intermediate concentrations of zinc (Group 2, 0.25-2.11 mmol/L). The mean values were 43.17 ± 19.03 % and 56.64 ± 20.28 %, respectively. And the lowest values came out in the highest zinc levels group (Group 4, > 3.04 mmol/L). In vitro cell experiments also showed that zinc caused dose-dependent cytotoxicity for GC-2 cells at a threshold value. Furthermore, RNA-seq analysis demonstrated that high concentrations of zinc exerted toxic effects on GC-2 cells through immune injury. Taken together, our findings suggested that moderate amounts of zinc are crucial for human reproduction and excessive concentrations may have adverse effects on male fertility.
Effect of Dietary Zinc Level on Production Performance and Semen Quality of Roosters

Abstract:45 Yisha roosters of parental generation in ten weeks were allotted at random into 3 groups with 3 replicates,5 roosters in each replicate.They were fed the basic diet and the basic diet added 60 mg/kg and 180mg/kg of Zn to studg the in fluence of dietary Zn level on the production per formance and semen quality of roosters.The results showed that supplementing Zinc in basal diet influenced semen quality and production performance to different degrees.There was no significant influence on the body weight and feed to weight of roosters at 31 weeks and 34 weeks(P>0.05),in addition,the difference of each groups at different weeks were all not significant(P>0.05);the difference of weight gain at 31 stweek between trial Ⅱ group and control group was significant(P<0.05);the differeme between trial group Ⅱ and trial group Ⅰat 32nd week was extremely significant(P<0.01);the difference between trialgroup Ⅱ and control group at 33rd weed week was also extremely significant(P<0.01).There was no significant difference on survival tate and live index of semen(P>0.05);the density of sperm in trial group Ⅰwas significantly lower than that of the trial group Ⅱ(P<0.01)and the control(P<0.05);the number of available sperm were very close between trial group Ⅰ was significantly higher than that of the coutrol(P<0.05);the pH value for semen in the control was extremely higher than that of the trial group Ⅰ and Ⅱ(P<0.01).

Abstract:In recent years, the quality of male sperm has shown a downward trend year by year. The male infertility rate in China continues to increase. It has been found that the impacts of environmental factors on male semen quality are mainly negative. Inorganic metallic elements as environmental contaminants have become a class of chemicals that cannot be ignored, and their health impacts on human reproductive systems have been received widespread attention and research. They certainly play a significant role in impairing male reproductive ability and are relative to the lower and lower semen quality. This review focused on the relationship between exposure to environmental metallic elements and semen quality of humans and animals, as well as summarized specific results from epidemiological studies, animal trials, or molecular experiments to provide a theoretical basis for protecting male reproductive capacity.

Abstract:To study effect of diet supplemented with different zinc levels on production performance,and reproductive hormone in laying hen,66 ISA brown laying hens in eighteen-week old were divided into control,treatment 1 and treatment 2 groups with the basal diet supplemented with different zinc levels at the dosage of 0 mg/kg,60 mg/kg,180 mg/kg,respectively.The results showed that supplemental zinc had not significant effect on egg production,feed conversion ratio and feed consumption.In compared with control group,percentage of eggs with defective shells was decreased in treatment groups,but it was no signi-ficant change(P0.05).Egg weight decreased significantly(P0.05).E2 concentration in serum was higher at treatment 1 group than that at control group(P0.05),and no difference with treatment 2 group(P0.05);while P4 concentration in serum was no obviously difference at all groups(P0.05).

Abstract:Zinc (Zn) is an essential trace element; it exhibits a plethora of physiological properties and biochemical functions. It plays a pivotal role in regulating the cell cycle, apoptosis, and DNA organization, as well as in protein, lipid, and carbohydrate metabolism. Among other important processes, Zn plays an essential role in reproductive health. The ZIP and ZnT proteins are responsible for the mobilization of Zn within the cell. Zn is an inert antioxidant through its interaction with a variety of proteins and enzymes to regulate the redox system, including metallothioneins (MTs), metalloenzymes, and gene regulatory proteins. The role of Zn in the reproductive system is of great importance; processes, such as spermatogenesis and sperm maturation that occur in the testicle and epididymis, respectively, depend on this element for their development and function. Zn modulates the synthesis of androgens, such as testosterone, for these reproductive processes, so Zn deficiency is related to alterations in sperm parameters that lead to male infertility.

Zinc Ameliorates Tripterygium Glycosides-Induced Reproductive Impairment in Male Rats by Regulating Zinc Homeostasis and Expression of Oxidative Stress-Related Genes

Abstract:Tripterygium glycosides (TG) can seriously damage male reproductive function, and the reproductive system is difficult to restore after stopping the administration of TG in male rats. Zinc (Zn) is one of the most important trace elements in the human body and plays an important role in maintaining male fertility. The aim of this study was to investigate whether zinc supplementation could improve the testicular reproductive damage induced by TG toxicity in rats and to investigate its mechanism of action. The results showed that zinc sulfate (ZnSO4) could improve testicular tissue structure and semen parameters, promote testosterone synthesis, increase zinc-containing enzyme activity, increase zinc concentration in serum and testicular tissues, and maintain zinc homeostasis in male rats induced by TG toxicity. Zinc supplementation activated relevant signalling molecules in the KEAP1-NRF2/ARE pathway and alleviated TG-induced oxidative stress. Therefore, this study concluded that zinc supplementation could improve reproductive damage by regulating zinc homeostasis and the expression of genes related to oxidative stress.

Abstract:Zearalenone (ZEA) mainly injures the reproductive system of mammals. In the present study, we aimed to explore the mechanism by which zinc inhibits ZEA-induced reproductive damage in KK-1 cells for the first time. The results shown that both zinc sulfate and zinc gluconate addition increased the intracellular zinc concentration and influenced the expression of zinc transporters (Slc30a1 and Slc39a1) in a time-dependent manner. Co-incubation of zinc with ZEA significantly reduced the ZEA-induced reactive oxygen species and malondialdehyde elevation by promoting the transcription of Mtf1 and Mt2. Meanwhile, two different zincs inhibited the ZEA-induced loss of mitochondrial membrane potential and elevation of late-stage apoptosis via activating the mitochondrial apoptotic pathway by recovering the mRNA and protein expression of pro-apoptotic genes (Bax, Casp3, Casp9). Zinc also recovered cells from S-phase cell cycle arrest. In addition, both of them promoted the ZEA-induced estrogen production but regulated the expression of steroidogenic enzymes (Star, Cyp11a1, Hsd3b1, Cyp17a1) in different way. All these results indicated that zinc could inhibit the reproductive toxicity of ZEA.
The Role of Zinc in Poultry Breeder and Hen Nutrition: an Update

Abstract:Zinc (Zn) is an essential trace mineral in breeder hen diets and functions in diverse physiological processes, including reproduction, immunity, antioxidant ability, and epigenetic processes. In this paper, five main aspects of Zn nutrition in poultry breeder birds and hens, including semen quality, molting, egg production and egg quality, hatchability and embryonic development, and offspring performance, are reviewed. Zn deficiency in poultry breeder birds led to lower semen quality (reducing around 10% sperm motility) and egg production (lowering 3-10 g/day/bird egg mass) as well as poor offspring development and growth performance (increasing 9-10% weak chick ratio and 10% mortality of progeny). Adequate maternal or higher Zn supplementation was adopted not only to induce molting with a greater postmolt performance (rising 4-7% laying rate) but also to enhance progeny immune response and antioxidant ability via epigenetic mechanisms. Therefore, it is necessary to reevaluate the optimal Zn requirement for egg production as well as the embryonic development and offspring chick performance of breeder hens. In the last 10 years, greater attention has been focused on the effectiveness of organic Zn for improving the reproductive performance of breeders and progeny viability and immune status. In fact, organic Zn sources are not always beneficial to the above aspects. So far, it has been very important to know the exact mechanisms of greater bioavailability and the epigenetic role of organic Zn sources in the augmentation of immune status and antioxidant abilities in poultry breeder birds and offspring. Therefore, a comprehensive analysis of these key points will not only aid in maintaining the beneficial effects of Zn nutrition for breeders and their progeny under stable conditions but will also support birds under stressful conditions such as disease as well as provide a better understanding of the integrated nutrition of breeder-offspring.

Protective Effects of Zinc on Rat Sperm Head-tail Connection

Abstract:ve: To study effects of zinc on male rat sperm head-tail connection. Methods: Rat spermatozoa from epididymis caput and cauda and testis were studied with phase contrast microscopy by exposing them in the detergent sodium dodecyl sulfate(SDS) or SDS containing EDTA including the effects of zinc on their stable percentage. Results: All these spermatozoa had different stable percentage while those from testis had the highest tendency to detach. Treatment with EDTA enhanced the proportion of detachment of spermatozoa. Still a certain percentage was resistant to EDTA treatment and this proportion increased upon 24 h of saline incubation. Zinc could reversibly inhibit such detachment. Conclusion: Zinc has protec-tive effects on sperm head-tail connection, the mechanism of such effects was probably relative to zinc-thiol interaction in nuclear chromatin.

Abstract:Zinc (Zn) is an essential nutrient for the human body. This nutrient is involved in numerous physiological functions and plays an important role in spermatogenesis. Zn-enriched yeast (ZnY) is considered a Zn supplement with high bioavailability and is widely used as a functional food. However, the effect of ZnY on male reproductive function remains unclear. This study aimed to investigate the beneficial effects of ZnY on the treatment of male spermatogenesis disorders. The spermatogenic dysfunctional mice were established by using cyclophosphamide (CP). CP was administered in saline at a dose of 50 mg/kg bw/day for 5 days by intraperitoneal injection (i.p.). Then, ZnY was orally supplemented at the dose levels of 2, 4, and 8 mg Zn/kg bw/day for 30 days. CP significantly decreased the sperm density and viability, testicular marker enzymes, serum testosterone, follicular stimulating hormone (FSH), and luteinizing hormone (LH). ZnY supplementation significantly improved these sperm parameters and hormone levels. Additionally, ZnY decreased the CP-induced lipid peroxidation and increased the glutathione levels. Moreover, ZnY increased the gene expression of anti-apoptotic proteins and steroid synthetase in mouse testes. The low-dose ZnY supplementation has a better effect on improving spermatogenesis, while the other two groups are less beneficial roles possibly due to excessive Zn intake. The present results suggest that appropriate ZnY can act as an accessory factor to improve steroid production and antioxidant levels in spermatogenic dysfunction mice.

Abstract:One of the effective methods for the long-term preservation of mammalian genetic resources is the cryopreservation of semen. However, a number of parameters, including diluents, the rate of freezing and thawing, cryoprotectants, etc., can easily alter the survival of frozen-thawed sperm. Numerous studies have documented the addition of a variety of zinc compounds, to the diluents used to cryopreserve sperm. The primary objective of this review is to briefly describe that adding zinc to diluents as an antioxidant significantly enhances frozenthawed sperm quality. Second, a summary of the present understanding of zinc's molecular mechanism on semen cryopreservation is provided. Thirdly, this study addresses that nanoparticles of zinc can offer suggestions for raising cryopreservation effectiveness.
Zinc Supplementation Reduces Testicular Cell Apoptosis in Mice and Improves Spermatogenic Dysfunction Caused by Marginal Zinc Deficiency

Abstract:Zinc (Zn) is an important trace element in the human body and plays an important role in growth, development, and male reproductive functions. Marginal zinc deficiency (MZD) is common in the human population and can cause spermatogenic dysfunction in males. Therefore, the aim of this study was to investigate methods to improve spermatogenic dysfunction caused by MZD and to further explore its mechanism of action. A total of 75 4-week-old male SPF ICR mice were randomly divided into five groups (control, MZD, MZD + ZnY2, MZD + ZnY4, and MZD + ZnY8, 15 mice per group). The dietary Zn content was 30 mg/kg in the control group and 10 mg/kg in the other groups. From low to high, the Zn supplementation doses administered to the three groups were 2, 4, and 8 mg/kg·bw. After 35 days, the zinc content, sperm quality, activity of spermatogenic enzymes, oxidative stress level, and apoptosis level of the testes in mice were determined. The results showed that MZD decreased the level of Zn in the serum, sperm quality, and activity of spermatogenic enzymes in mice. After Zn supplementation, the Zn level in the serum increased, sperm quality was significantly improved, and spermatogenic enzyme activity was restored. In addition, MZD reduced the content of antioxidants (copper-zinc superoxide dismutase (Cu–Zn SOD), metallothionein (MT), and glutathione (GSH) and promoted malondialdehyde (MDA) production. The apoptosis index of the testis also increased significantly in the MZD group. After Zn supplementation, the level of oxidative stress decreased, and the apoptosis index in the testis was reduced. Furthermore, quantitative real-time polymerase chain reaction (qRT–PCR) showed that the expression of B-cell lymphoma-2 (Bcl-2) mRNA and Bcl-2/BCL2-associated X (Bax) in the control group decreased in testicular cells, and their expression was restored after Zn supplementation. The results of this study indicated that Zn supplementation can reduce the level of oxidative stress and increase the ability of testicular cells to resist apoptosis, thereby improving spermatogenic dysfunction caused by MZD in mice.

Effects of Zinc-Glycine Supplement on the Serum Hormone Level,Corpus Luteum Count and Conception Rate of Small-tail Han Ewes

Abstract:In order to ascertain the effects of zinc-Glycine supplemented in diet on the reproduction effect of receptor sheep,60 two-years old Small-tail Han ewes managed under stall-feeding condition were divided into treatment and control groups randomly.The control group was fed with basal diet,and the treatment group was fed with basal diet plus 90mg/kg of Zn-Glycine.The quality and quantity of corpus luteum of small-tail Han ewes were evaluated when the embryos were transferred into them,the serums were prepared to determine the levels of P4,E2,FSH,LH,FT3 and FT4 at 25th-day after treatment.The results showed that the treatment group had 2.6 red plump corpus luteums in average,compared to an increase of 1.2 for the control group.The concentrations of serum P4 and E2 in treatment group were 7.03 μg/L and 10.82 ng/L respectively,both of them were significantly more than in the control group(P<0.05).The levels of serum FSH and LH in treatment group were 4.97 IU/L and 4.30 IU/L,there was no obvious difference between two groups.The serum FT3 and FT4 levels were 4.98 pmol/L and 10.86pmol/L respectively,the FT3 level was significantly lower than that of control group(P<0.05).Hence,the quantity of available corpus luteums and serum P4 and E2 levels of small-tail Han sheep used for receptor were improved by Zn-Glycine supplemented in diet.

Abstract:Selenium is one of the essential trace elements for animals,dietary selenium can effectively improve reproductive performance of sows.Studies have shown that selenium improve antioxidant capacity of sow through selenoprotein and synergy with vitamin E,and regulate reproductive performance of sow through metabolic regulation of deiodinase and pancreatic digestive enzymes.In this paper,the effects of selenium on the reproductive performance of sows and its possible mechanism were reviewed,and aimed at providing reference for related research and reasonable application of selenium in sows.
