Study on Mode of Cultured Hippocampal Neurons' Death Induced by Β-Amyloid Protein in Vitro

Abstract:Objective To investigate the modes of hippocampal neurons'death induced by β-amyloid protein(Aβ).Methods The toxicity of Aβ_(1-40) to neurons was observed at different concentrations for different time-periods in vitro.Neuron death was analyzed by electron microscopy,DNA agarose gel electrophoresis and fluorescence staining.Results At 12 h after treating with 5,10,20 μmol/L Aβ_(1-40),apoptosis and oncosis rates of neurons were significantly higher than those of the control group.At 6 h,12 h and 24 h following adding 10 μmol/L Aβ_(1-40) to the culture medium,apoptosis and oncosis rates of neurons were significantly higher than those of the control group.Conclusion Certain concentrations of Aβ_(1-40) showed cytotoxic effect on cultured neurons and induced the neuron necrosis in both dose-and time-dependent manners.
