Abstract:AIM:To investigate the change of cellular morphology,cell proliferative activity and cell dividing cycle in human retinal pigment epithelium (HRPE) cells after treatment of human vitreous conditional medium (VCM). METHODS:HRPE cells were treated with VCM including 100,250,500 and 1 000mL/L human vitreous respectively. Cellular morphology,cell proliferative activity and cell dividing cycle changes were observed by invert microscope,MTT assay and Flow Cytometry (FCM) assay respectively. RESULTS:HRPE cells showed polynucleation and pre-depigmenting,fibroblast-like phenotype. For HRPE cells proliferative activity,there was significant disparity between control and experimental group,especially for VCM with 250mL/L vitreous. VCM increased the HRPE cells' proportion of phase S (6%～13%)and heteroploid was found in VCM with 250 and 500 mL/L vitreous. CONCLUSION:VCM can increase the S phase proption in HRPE cells' cell dividing cycle,and increase their proliferative activity.
