The study on mutation and MSI of LGI 1 in glioma

Abstract:Objective To detect mutations of LOH and MSI in the encoding regions of LGI 1 in human glioma specimens and glioma cell lines.Methods The brain tissue specimens of 30 glioma, 2 para-tumor, 2 meningioma ,2 craniocerebral injury from cranialcerebral injury patients with intracranial decompression and 5 glioma cell lines were used in present study. Genomic DNA was isolated from all specimens. Primers were designed for amplifying each exon of LGI 1 with PCR. The PCR products were analyzed with silver staining method for single-strand conformation polymorphism(SSCP). Primers were designed for amplifying microsatellite sequence of LGI 1 with PCR; The PCR products were detected with the methods of electrophoresis and silver staining. If any mutation or LOH was found in banding patterns, the DNA sequence analysis was made directly.Results (1) PCR-SSCP analysis: No LGI 1 exon 1a in PCR products was detected in one of gradeⅡglioma specimen and no LGI 1 exon 8c was detected in one of grade Ⅱand Ⅲ glioma specimens respectively by agar-gel electrophoresis. No alternation in banding patterns was observed in 30 glioma specimens and 4 glioma cell lines. LGI 1 exon 5 in banding patterns was observed in cell line TJ905, Mutations were substantiated by DNA sequencing. (2) The MSI and LOH were not found in 30 glioma specimens and 5 glioma cell lines.Conclusions It appears that mutation of LOH or MSI of LGI 1 is not a common event in gliomagenesis and malignant development.
