Abstract:OBJECTIVE:To investigate the expression of B cell specific MLV integration site-1 (Bmi-1) in colorectal cancer (CRC) and its correlation to the clinicopathological features and prognosis of CRC. METHODS:Sixty CRC, 30 adenomas and 20 normal colorectal mucosal tissues were collected to detect the expression of Bmi-1 protein using immunohistochemistry, and the results were analyzed in comparison with the clinicopathological features and survival rate of patients. RESULTS:The positivity rate of Bmi-1 expression in CRC tissue was 51.7%. In CRC, the rate of Bmi-1 overexpression was 25.0%, significantly higher than that in the adenomas and normal colorectal mucosal tissues (6.67% and 0%, respectively, P<0.05). The overexpression of Bmi-1 protein in CRC was obviously associated with distant metastasis and the TNM stage (P<0.05), but not with gender, age, tumor size, tumor site, histological type, differentiation degree and lymph node metastasis (P>0.05). But logistic regression analysis showed that Bmi-1 protein overexpression in CRC was associated only with distant metastasis (P<0.01,OR>1); Kaplan-Meier survival analysis showed that the survival rate of the patients with high Bmi-1 expression was significantly lower than that in patients with low expression (P<0.05). CONCLUSION:The overexpression of Bmi-1 protein was significantly correlated to the tumorigenesis, metastasis and prognosis of CRC, and may serve as an indicator for evaluating the prognosis of CRC.
Matrix metalloproteinase-1 expression in breast carcinoma: a marker for unfavorable prognosis.

Abstract:Matrix metalloproteinase-1 (MMP1) is a member of the matrix metalloproteinases family, and its aberrant expression is implicated in tumor invasion and metastasis. However, the relationship between MMP1 abnormal expression and clinical outcome in breast cancer patients remains to be elucidated. To address this issue, we conducted immunohistochemistry in breast cancer and adjacent normal tissues, and mined the transcriptional and survival data of MMP1 in breast cancer patients through Oncomine, Kaplan-Meier Plotter, bc-GenExMiner, COSMIC and cBioPortal databases. First, we found that both protein and mRNA levels of MMP1 expression were significantly higher in breast cancer tissues. Second, high MMP1 mRNA expression correlated with worse overall survival among grade II (HR = 1.75; p = 0.011), nodal-negative (HR = 2.00; p = 0.00028), ER-positive (HR = 1.61; p = 0.00027) and HER2-negative (HR = 3.17; p = 0.029) patients with breast cancer by using Kaplan-Meier plotter database. Third, the overexpression of MMP1 was associated with unfavorable survival results including overall survival (HR = 1.6; p = 1.6e-05), relapse free survival (HR = 1.78; p < 1e-16) and distant metastasis free survival (HR = 1.65; p = 5.3e-05) in patients with breast cancer. Taken together, the expression status of MMP1 is a significant prognostic indicator and a potential drug target for breast cancer.
A Study on the Expression of MTSS1 in Hepatocellular Carcinoma

Abstract:Objective To explore the expression and significance of MTSS1 ( metastasis suppressor 1) in hepatocellular carcinoma.Methods MTSS1 expression was detected by immunohistochemistry in hepatocellular carcinoma, liver cirrhosis and normal liver tissues.Single-factor analysis was used to study the relationship with clinicopathological factor.Correlations between MTSS1 expression and TNM stage were analyzed with Spearman rank correlation analysis.Postoperative 5-year survival was evaluated using Kaplan-Meier survival curve analysis.Results The expression of MTSS1 in hepatocellular carcinoma was higher than normal liver tissue ( U = 168.000, P ＜ 0.05), and liver cirrhotic tissue ( U = 106.000, P ＜ 0.05); MTSS1 expression was correlated with TNM stage of hepatocellular carcinoma patients, lymph vascular invasion and tumor capsule ( separately U = 259.000, 258.500, 202.000, all P ＜ 0.05).MTSS1 expression in hepatocellular carcinoma was not correlated with patients age, gender, tumor size, AFP level, and hepatitis B surface antigen.MTSS1 expression and TNM stage of liver cancer patients was negatively correlated ( rs = - 0.383 , P ＜ 0.05 ).Postoperative 5-year survival of hepatocellular carcinoma patients with MTSS1 positive expression was significantly poorer than patients with negative and weakly positive expression (respectively 34.1% and 52.3% , x2 =6.386, P ＜ 0.05).Conclusions MTSS1 high expression may play an important role in the early hepatocellular carcinoma progression, indicating a poor prognosis.

Abstract:BACKGROUND:Metastasis-associated protein 1 (MTA1) plays an important role in tumourigenesis and progression of certain cancer types. In the current study, we analyzed the relationship between MTA1 expression and disease progression of colorectal cancer (CRC).MATERIALS AND METHODS:CRC tissues (n=93) and adjacent normal colorectal tissues (n=70) were analyzed by quantitative real-time polymerase chain reaction. MTA1 knockdown was established in RKO and HT115 cells using MTA1 siRNA.RESULTS:The expression of MTA1 was significantly increased in CRC tissues compared to paired normal colorectal tissues, but decreased expression of MTA1 was correlated with poor prognosis (higher lymph node involvement stage, TNM stage, local invasion and recurrence) that was associated with increased expression of VEGFC and -D and the receptor VEGFR3.CONCLUSION:MTA1 is up-regulated in CRC. MTA1 expression is inversely associated with lymphatic metastases and the expression of VEGFC, VEGFD and VEGFR3.

Abstract:BACKGROUND:The putative tumor metastasis suppressor 1(MTSS1) is an actin-binding scaffold protein that has been implicated to play an important role in carcinogenesis and cancer metastasis, yet its role in the development of gastric cancer has not been well illustrated. In this study, we detected MTSS1 expression and explored its clinical significance in gastric cancer.METHODS:Immunohistochemistry was performed using tissue microarrays containing gastric adenocarcinoma specimens from 1,072 Chinese patients with normal adjacent mucosa, primary gastric cancer and lymph node (LN) metastasis and specific antibody against MTSS1. MTSS1 mRNA and protein expression were detected by reverse transcription-polymerase chain reaction and Western blotting. The clinical follow-up was done in the 669 patients living in Shanghai that was chose from the 1072 cases.RESULTS:Complete loss of MTSS1 expression was observed in 751 cases (70.1%) of the 1,072 primary tumors and 103 (88%) of 117 nodal metastases; and loss of MTSS1 expression was significantly associated with poorly differentiated tumors, large tumor size, deep invasion level, the presence of nodal metastases and advanced disease stage. Moreover, multivariate analysis demonstrated that loss of MTSS1 expression correlated significantly with poor survival rates (RR = 0.194, 95% CI = 0.144-0.261, P < 0.001).CONCLUSIONS:MTSS1 expression decreased significantly as gastric cancer progressed and metastasized, suggesting MTSS1 may serve as a useful biomarker for the prediction of outcome of gastric cancer.
Expression of MTSS1 in cervical cancer tissue and its clinical significance

Abstract:Objective To detect the expression of metastasis sappressor 1(MTSS1) gene in cervical cancer tissue and to clarify its association with cervical cancer.Methods Totally 103 cases of cervical tissue were collected between Dec 2011 and Dec 2014 and classified according to biopsy and stage .Q-PCR and Western blotting were used to detect the expression of MTSS1 in normal cervical tissue and in different clinical stages of cervical cancer tissue .Results The expression of MTSS1 inⅡB-Ⅳstages of cervical cancer tissue was significantly higher than that of normal tissue or Ⅰ-ⅡA stages through q-PCR (P=0.000).Western blotting results showed that MTSS1 was positively expressed in normal cervical tissue at a rate of 23.3% or 53.3% in cervical cancer tissue.Moreover, the expression of MTSS1 was poorly correlated with age, tumor differentiation and lymphnode metastasis in cervical cancer tissue (P>0.05).The protein level of MTSS1 expressed in ⅡB-Ⅳ stages was significantly higher than that ofⅠ-ⅡA stages(P=0.005).Conclusion The expression of MTSS1 indicates the clinical stage of cervical cancer , suggesting that MTSS1 may play an important role in the development of cervical cancer .
Prognostic Correlation Between MTA2 Expression Level and Colorectal Cancer.

Abstract:OBJECTIVES:Association of MTA2 expression with presence, development, metastasis and prognosis of colorectal cancer (CRC) was investigated. METHODS:90 CRC-related cases with follow-up information were made into tissue microarrays according to the paired principle of cancer tissues and the adjacent tissues. Subsequently, the expression of MAT2 was detected with immunohistochemical analysis and SPSS software was finally utilized to analyze the relationships between experimental data and clinical indicatives. RESULTS:Expression of MTA2 in CRC tissues were notably higher than their adjacent tissues (P < 0.001) and showed significant positive correlation with tumor grade (r(2) > 0, P < 0.01). Moreover, survival analysis indicated that MTA2 expression in cancer tissues, serving as an independent correlation factor, was significantly correlated with poor prognosis (P = 0.004). CONCLUSIONS:MTA2 is a crucial biomarker that is closely related with prognosis of CRC and also a potential molecular target for evaluating the prognosis and treatment of CRC.
Role of Metastasis-Associated Protein 1 in Prognosis of Patients with Digestive Tract Cancers: A Meta-Analysis

Abstract:OBJECTIVES:Metastasis-associated protein 1 (MTA1) is a transcriptional regulator and significantly associated with prognosis of patients with cancer. However, its role as a potential prognostic marker in digestive tract cancer (DTC) is controversial. In this study, a meta-analysis was conducted to evaluate the MTA1 expression as a predictor of clinicopathology and survival of patients with DTC.METHODS:We searched PubMed, Ovid, Web of Science and Cochrane databases using multiple search strategies for eligible studies. STATA 11.0 software was used to pool the data and analyze the association, odds ratios (ORs) and 95% confidence intervals (CIs) were used to measure the strength of the association. Furthermore, the Newcastle-Ottawa scale was used to evaluate the quality of eligible studies.RESULTS:MTA1 overexpression was strongly associated with depth of invasion (OR = 1.88, 95%CI: 1.05-3.37, P = 0.03), lymph node metastasis (OR = 2.30, 95%CI: 1.76-3.01, P<0.001), vascular invasion (OR = 2.02, 95%CI: 1.40-2.91, P<0.001) and TNM stage (OR = 2.78, 95%CI: 1.63-4.74, P<0.001), and was related to 1- (RR = 1.84, 95%CI: 1.18-2.89, P = 0.008), 3- (RR = 1.74, 95%CI: 1.32-2.30, P<0.001) and 5-year (RR = 1.64, 95%CI: 1.18-2.27, P = 0.003) OS. Further, MTA1 was associated with 1- (RR = 4.16, 95%CI: 1.35-12.81, P = 0.01), 3- (RR = 1.90, 95%CI: 1.02-3.53, P = 0.04) and 5- (RR = 2.17, 95%CI: 1.41-3.32, P<0.001) year DFS. In subgroup analyses based on study quality and tumor type, MTA1 overexpression was obviously related to clinical parameters, such as lymph node metastasis and TNM stage, and was also associated with prognosis of patients with gastrointestinal or esophageal cancer.CONCLUSIONS:MTA1 expression is strongly correlated with metastasis-related variables, and represents a promising prognostic factor in DTC.

Abstract:OBJECTIVE:To investigate the expression of miR-23a and metastasis suppressor 1 (MTSS1) and their clinical significance in colon carcinoma.METHODS:A total of 92 cases of colon carcinomas were collected with both the tumor and paired normal tissue samples for the study. The miR-23a targeting MTSS1 was evaluated by luciferase reporter vector. Cell invasion potential was evaluated by trans-well invasion assay. In-situ hybridization and immunohistochemistry were used to detect miR-23a and MTSS1 expression.RESULTS:MiR-23a downregulated the expression of MTSS protein and enhanced the invasiveness of colon carcinoma. The expression rates of miR-23a and MTSS1 were 87.0% (80/92) and 17.4% (16/92) in colon carcinoma cases, respectively (P < 0.01). The up-regulation of miR-23a expression was associated with an advanced clinical stage (P = 0.029) and depth of invasion (P = 0.000). The expression of miR-23a was higher in the tumors with lymph node metastasis than those without (P = 0.041). Down-regulation of MTSS1 expression was associated with an advanced clinical stage (P = 0.027) and depth of invasion (P = 0.017). The expression of MTSS1 was lower in the tumors with lymph node metastasis than those without (P = 0.009). The expression of miR-23a had significantly negative correlation with that of MTSS1 (r = -0.594, P = 0.013).CONCLUSIONS:MiR-23a expression promotes colon carcinoma cell growth, invasion and metastasis through inhibition of MTSS gene. Both the low expression of MTSS1 and high expression of miR-23a may serve as important biological markers for the malignant phenotypes of colon cancer, such as invasion and metastasis.
[Relationship Between Metastasis-Associated Protein 1 Expression and Metastasis and Prognosis in Non-Small Cell Lung Cancer].

Abstract:BACKGROUND:Metastasis is the main cause of the death of lung cancer patients. Much attention has been pain to the research of lung cancer metastasis. Metastasis-associated protein 1 (MTA1) is one member of metastasis associated protein family. Its overexpression is correlated with metastasis of esophageal carcinoma and breast cancer, but the generality of its expression in cancer and the significance for judging biological behaviors of tumor and evaluating prognosis of patients is to be investigated. The aim of the study is to study the relationship between MTA1 expression and clinicopathological factor regarding metastasis and prognosis of human non-small cell lung cancer (NSCLC).METHODS:The expression of MTA1 was detected in 101 parafin-embedded specimens by immunohistochemistry method, as well as in 35 freshly-taken NSCLC tissues by Western blot.RESULTS:There were 56 cases (55.4%) of NSCLC with yellow or even brown particles in nucleus of tumor cell among 101 cases, and MTA1 protein showed negative expression in epithelia of bronchi or alveoli in neighboring noncancerous tissue. Western blot analysis showed the level of MTA1 in NSCLC tissues was remarkably higher than that in normal tissues (t=3.953, P=0.000). Expression of MTA1 was remarkably higher in tumor with metastasis than that in tumor without metastasis (t=4.057, P=0.000). Expression of MTA1 significantly correlated with differentiation (Chi-square=10.131, P=0.006), lymphatic metastasis (Chi-square=8.535, P=0.003) and p-TNM stage (Chi-square=17.419, P=0.000). The survival time of pa-tients with negative MTA1 expression was (44.866±12.946) months, which was significantly higher than that of patients with positive MTA1 expression [(23.714±7.498) months] (Chi-square=10.006, P=0.002). In multivariate analysis, only lymphatic metastasis and TNM stage could be considered as independent prognostic factors.CONCLUSIONS:MTA1 might play an important role in the development and metastasis of NSCLC. Patients with MTA1 expression have a greater chance of metastasis and a poorer prognosis. However, MTA1 expression is not an independent prognosis factor.
Overexpression Of Nicotinamide N-Methyltransferase Mainly Covers Stroma Of Colorectal Cancer And Correlates With Unfavorable Survival By Its Product 1-Mna

Abstract:Background: Accumulating evidence indicates that Nicotinamide N-methyltransferase (NNMT) is abnormally expressed in tumor tissues of several cancers including colorectal cancer (CRC) and associated with cancer progression. However, the distribution characteristics and the clinical value of each part of NNMT expression in CRC are still not fully understood. The purpose of this study is to determine the distribution of NNMT expression and its association with survival in CRC.Methods: By using the cancer genome atlas (TCGA) and clinical proteomic tumor analysis consortium (CPTAC), we firstly analyzed the difference of gene and protein levels of NNMT between CRC and normal colorectal tissue. Then, NN MT protein expressions were detected in 18 intraepithelial neoplastic samples and 177 CRC tumor samples through immunohistochemistry in our study cohort. Furthermore, the relationship between NNMT expression and clinicopathological characteristics, overall survival (OS) and disease-free survival (DFS) of CRC patients were analyzed by Pearson chi(2) test and log-rank test, respectively, in public datasets and our study cohort. Lastly, the function of NNMT and its product 1-methyl-nicotinamide (1-MNA) on migration and invasion in colorectal cancer cells was analyzed by wound healing assay and transwell assay.Results: We determined that higher NNMT expression in CRC tissues than normal tissues in both gene and protein level in TCGA and CPTAC datasets (all p < 0.05). In addition, the strong relationships of NNMT expression with stromal cells were found in the TCGA cohort. Fortunately, our cohort could validate that the expression of NNMT in tumor stroma cell was significantly higher than that in tumor cell (p < 0.0001), and both of them were significantly higher than that in adjacent normal tissue (ANT) (p < 0.0001 and p < 0.0001, respectively). Furthermore, the positive NNMT expression in tumor cell and stromal cell were associated with series of unfavorable clinical characteristics, including advanced TNM stage, lymph node metastasis, distant metastasis (all p < 0.05). Also, higher NNMT was associated with unfavorable survival both in our study and public datasets, including TCGA and two Gene Expression Omnibus (GEO) datasets (GSE33113 and GSE17538). Moreover, the functional experiments showed that stromal cells with high NNMT expression can secret 1-MAN to promote migration and invasion of CRC cells in vitro.Conclusions: In CRC, NNMT is overexpressed in tumor cells and stroma cells, and then mainly expressed in tumor stroma cells. Overexpression of NNMT in tumor cell and stroma cell both are associated with metastasis and unfavorable survival. Besides, stromal cells with high NNMT expression secrets 1-MAN to promote migration and invasion of CRC cells. Therefore, NNMT may be a potential prognostic indicator in CRC patients.
Downregulation of long non-coding RNA MT 1 DP correlates with poor prognosis in colorectal cancer

Abstract:Background: Accumulated evidence has shown that long non-coding RNAs (lncRNAs) are emerging as key molecules in human malignancies. The lncRNA metallothionein 1D, pseudogene (MT1DP) was recently identified as a tumor suppressor in liver cancers. However, its expression pattern and clinical performance in colorectal cancer (CRC) have not been addressed. Methods: Our investigation was carried out on 78 cases of CRC tissues and matched adjacent normal tissues. The expression levels of MT1DP were determined by quantitative real-time PCR. Subsequently, its clinical values in CRC were further evaluated. Results: MT1DP expression was significantly downregulated in 55 of 78 CRC tissues (70.5%, P<0.001). The area under curve (AUC) of the ROC was 0.714; 95% confidence interval (CI) was 0.636-0.783, the sensitivity was 88.46% and the specificity was 51.28%. Moreover, low level of MT1DP expression in CRC was closely correlated with TNM stage (P=0.020) and distant metastasis (P=0.005). In addition, low MT1DP expression had a poorer overall survival (OS) and disease-free survival (DFS) rates in CRC patients (P=0.002 and 0.006, respectively). Additionally, univariate and multivariable Cox regression analyses further identified that downregulated MT1DP might act as an independent prognostic factor for OS and DFS of CRC patients. Conclusions: Low expression of MT1DP is involved in CRC progression and could be a novel biomarker of diagnostic and prognostic evaluation of CRC patients.

Abstract:Objective Cancer metastasis involves multiple genes and their products.Metastasis-associated-gene 1(MTA1) is a newly discovered cancer metastasis-related gene.This study was to explore the relationship of MTA1 expression in non-small cell lung cancer(NSCLC) with the clinicopathological profiles of the patient and determine its value in predicting the metastasis potential and clinical prognosis of NSCLC.Methods Using immunohistochemical staining,we detected the expression of MTA1 in the NSCLC tissue samples of 96 patients with complete follow-up data.Meanwhile,we analyzed the clinicopathological profiles and evaluated their correlation with metastasis potential and prognosis of NSCLC.Results The expression of MTA1 exhibited significant differences in NSCLC among tumors of different diameters(d)(d≤3 cm: 54.5%,d≤5 cm: 93.3%,d 5 cm: 81.8%,χ2 = 13.898,P = 0.001),TNM stages(stageⅠ: 53.8%,stage Ⅱ: 46.7%,stage Ⅲ: 89.1%,stage Ⅳ: 81.8%,χ2 = 15.348,P = 0.002),differentiation degrees(high and moderate: 61.5%,low and anaplastic: 81.4%,χ2 = 4.117,P = 0.042),and between lymph node metastasis and non-lymph node metastasis(59.1% vs 81.1%,χ2 = 4.501,P = 0.034),but not among different age and gender groups and pathological types(P 0.05).The Kaplan-Meiert survival curve showed the 2-year the progress-free survival(PFS) rates to be 8.2% and 30.4% in the NSCLC patients with highly and lowly expressed MTA1,respectively(P = 0.007).COX multivariate analysis indicated that high MTA1 expression,TNM staging and lymph node metastasis were independent risk factors for the prognosis of NSCLC.Conclusion MTA1 is highly expressed in NSCLC,and is closely associated with the metastasis and prognosis of lung cancer.

Abstract:Objective To study the expression of metastasis associated gene 1(MTA1)and its association with prognosis in bladder cancer.Methods The expression of MTA1 was investigated by Western blot,real time PCR and immunohistochemical staining in bladder cancer tissues and adjacent tissues.The relationship between expression of MTA1 and clinical features,including age,tumor size,T stage,lymph node metastasis and lymphatic invasion,were analyzed.And then the MTA1 influence on prognosis was explored.Results MTA1 expression was significantly higher in bladder cancer tissues than in adjacent tissues by western blot and PCR.The positive rate of MTA1 was 63.79% in bladder cancer tissues,and there was no significant association between MTA1 and clinical features except for lymph node metastasis(P=0.035)and lymphatic invasion(P=0.001).The 5-year survival rate of patients with strong MTA1 expression was lower than that of patients with negative expression.MTA1 expression was an independent prognostic factor for overall survival of bladder cancer.Conclusion Expression of MTA1is significantly high in bladder cancer and has critical roles in tumor metastasis,which might provide a potential prognostic and therapeutic marker for patients with bladder cancer patients.

Abstract:BACKGROUND Metastasis suppressor 1 (MTSS1) is proposed to function as a cytoskeletal-associated protein, which may play a role in the aggressiveness of cancer cells. Recent studies have demonstrated a clinical significance of MTSS1 in certain types of cancer, but its role in bladder cancer remains unknown. We investigated the expression of MTSS1 in normal and malignant human bladder tissues and its molecular interaction within bladder cancer cells. MATERIALS AND METHODS The expression of MTSS1 in human bladder tissues and bladder cancer cell lines was assessed at both the mRNA and protein levels using reverse transcription-PCR (RT-PCR) and immunohistochemistry, respectively. Full-length MTSS1 cDNA was amplified from normal mammary tissues. The effect of MTSS1 overexpression on cellular functions was examined in bladder cancer cells using a variety of in vitro assays. RESULTS Transitional cells of bladder tissues stained positively for MTSS1. However, cancer cells from tumour tissues did not stain for MTSS1. Similarly, in bladder cancer cell lines, MTSS1 was almost absent from T24 and RT112 cells, and the EJ138 cell line expressed a very low level of MTSS1. Overexpression of MTSS1 reduced the growth and adhesion of bladder cell lines in vitro. However, overexpression of MTSS1 had no bearing on the invasion and migration of bladder cell lines in vitro. CONCLUSION MTSS1 is expressed at low levels or is absent from human bladder cancer cells. MTSS1 levels are inversely correlated with the growth and adhesion of bladder cancer cells in vitro. MTSS1 appears to be a potential tumour suppressor in human bladder cancer.

Abstract:Background Metastasis suppressor-1 (MTSS1) has been proposed to function as a cytoskeletal protein with a role in cancer metastasis. Recent studies have demonstrated the clinical significance of MTSS1 in certain type of cancers, yet the clinical relevance of MTSS1 in oesophageal squamous cell carcinoma (ESCC) has not been reported. Methods In this study, we assessed the expression levels of MTSS1 in tumours and its matched adjacent non-tumour tissues obtained from 105 ESCC patients. We also used ESCC cells with differing MTSS1 expression and assessed the influence of MTSS1 on ESCC cells. Results Down-regulation of MTSS1 expression was observed both in oesophageal tumour tissues and ESCC cancer cell lines. We also reported that MTSS1 expression was associated with tumour grade (p = 0.024), lymph node metastasis (p = 0.010) and overall survival (p = 0.035). Patients with high levels of MTSS1 transcripts had a favorable prognosis in comparison with those who had reduced or absent expression levels. Using over-expression and knockdown approach, we created sublines from ESCC cells and further demonstrated that MTSS1 expression in ESCC cells significantly influenced the aggressiveness of the oesophageal cancer cells, by reducing their cellular migration and in vitro invasiveness. Conclusion MTSS1 serves as a potential prognostic indicator in human ESCC and may be an important target for cancer therapy.

Abstract:Objectives: Association of MTA2 expression with presence, development, metastasis and prognosis of colorectal cancer (CRC) was investigated. Methods: 90 CRC-related cases with follow-up information were made into tissue microarrays according to the paired principle of cancer tissues and the adjacent tissues. Subsequently, the expression of MAT2 was detected with immunohistochemical analysis and SPSS software was finally utilized to analyze the relationships between experimental data and clinical indicatives. Results: Expression of MTA2 in CRC tissues were notably higher than their adjacent tissues (P 0, P < 0.01). Moreover, survival analysis indicated that MTA2 expression in cancer tissues, serv- ing as an independent correlation factor, was significantly correlated with poor prognosis ( P = 0.004). Conclusions: MTA2 is a crucial biomarker that is closely related with prognosis of CRC and also a potential molecular target for evaluating the prognosis and treatment of CRC.
Overexpression of MSK1 is associated with tumor aggressiveness and poor prognosis in colorectal cancer

Abstract:Background/aims: Mitogen- and stress-activated protein kinase 1 (MSK1) has recently been implicated in cell proliferation and neoplastic transformation. However, the involvement of MSK1 in colorectal cancer (CRC) has not been addressed. This study aimed to evaluate the expression and potential functions of MSK1 in CRC. Methods: The MSK1 expression was investigated by immunohistochemistry, western blot and reverse transcription-polymerase chain reaction. The associations between clinicopathological characteristics and MSK1 expression were assessed. Kaplan-Meier analysis and Cox regression models were carried out. CRC cells with MSK1 knockdown or overexpression were generated. A range of experiments were performed to demonstrate MSK1's role in CRC. Results: MSK1 was overexpressed in 148 out of 329 CRC patients. CRC patients with high MSK1 expression had shorter overall survival than those with low MSK1 (P=0.033), especially among patients with stage III tumors (P=0.005). Knockdown of MSK1 in CRC cells suppressed cell proliferation, anchorage-independent growth, migration and invasion, and promoted 5-fluorouracil chemosensitivity and intracellular NADP+/NADPH ratio. However, overexpression of MSK1 had the opposite effects. Conclusions: Overexpression of MSK1 is associated with poor prognosis in CRC and is connected to tumor aggressiveness. MSK1 is a potential target for new therapies and a candidate of biomarker for prognosis.

Abstract:Metastasis suppressor 1 (MTSS1) is a potential metastasis suppressor gene involved in the regulation of cytoskeleton dynamics and subsequently in cell motility. MTSS1 expression is frequently reduced in a variety of cancer cells and tissues and this loss may account for increased invasive traits in cancer cells. The present study aimed to assess the role of MTSS1 in epithelial ovarian cancer (EOC) cells. Expression of MTSS1 in human ovarian cells was assessed at both the mRNA and protein levels using reverse transcription-PCR (RT-PCR) and immunohistochemistry, respectively. Full-length MTSS1 cDNA expression vector was used to generate MTSS1 overexpressing cells. The effect of MTSS1 overexpression on cellular functions was examined in EOC cells using a variety of in vitro assays. MTSS1 expression was observed both in ovarian cancer tissues and EOC cells. Over-expression of MTSS1 protein reduced the growth, invasion, adhesion and migration of EOC cell lines in vitro. The present study revealed that MTSS1 plays an essential inhibitory role in the development and progression of ovarian cancers. MTSS1 overexpression is intimately related to migration and metastasis, suggesting that MTSS1 is a potential prognostic marker and therapeutic molecular target in human ovarian cancer.
