Effects of L-NAME on blood-brain barrier injury and brain edema in severely burned rats

Abstract:Objective:To explore whether nitric oxide (NO) mediates blood-brain barrier (BBB) injury at early stage of severe scald by observing the effects of N_ω-nitro-L-arginine methyl ester (L-NAME) on the permeability of BBB in post-scald rats with severe burn.Methods:Healthy Sprague-Dawley rats were separated into three groups:normal control,scald and L-NAME,and the later two groups were divided into five post-scald groups:1 h,3 h,6 h,12 h, 24 h.The changes in BBB permeability were determined by detection of Evans blue (EB) content in the rat brains.To measure water content of the brain,the uhrastructural changes of BBB in the brain were observed.Results.Compared with the control group,a significant increase of EB content was found in the untreated brains with L-NAME.Howev- er,EB extravasations were significantly restricted in the all subgroups of treated rats with L-NAME under transmis- sion electron microscope,the vacuolated,swollen endothelium and abnormity of capillary basement membranes in BBB of the burned rats were observed.The changes of BBB of the brains with severe scald nearly returned to normal after treatment with L-NAME.Conclusion:The BBB permeability may increase after acute severe scald,which can be sig- nificantly restricted by pretreatment with L-NAME at the early stage.
