Effect of Reactive Oxygen Species in Inducing Hepatic Apoptosis in Rats with Obstructive Jaundice

Abstract:[Objective] To investigate the role of reactive oxygen species(ROS) in obstructive jaundice. [Methods] Glutathione(GSH) level, glutathione peroxidase (GPx) activity, ROS level and hepatic apoptotic index(AI) were determined in cultured liver cells and/or in a rat model. [Results] In vitro experiments revealed that liver cells dying of apoptosis reached (43.50±7.09) % and (53.38±5.24) % after cocultured with 100μM glycine conjugates of chenodeoxycholic acid (GCDC) for 2 h and 4 h respectively. GSH was depleted in a time-dependent manner. In rat models, hepatic GSH level and GPx activity decreased and ROS level increased progressively in response to elevation of serum bile salt subseqent to ligation of bile duct. Tunel examination showed massive hepatic apoptosis. GSH level and GPx activity in rats treated with sodium selenite were higher, while ROS level and AI were lower than those who receiving only physiological saline. [Conclusions] ROS plays an essential role in obstructive jaundice-related hepatic injury. And the results present here indicate that sodium selenite has protected the cells against ROS-induced hepatic apoptosis .
