Abstract:Density functional theory (DFT) calculations were performed to investigate the origins of enantioselectivity in benzotetramisole (BTM)-catalyzed dynamic kinetic resolution of azlactones. The transition states of the fast-reacting enantiomer are stabilized by electrostatic interactions between the amide carbonyl group and the acetate anion bound to the nucleophile. The chiral BTM catalyst confines the conformation of the α-carbon and the facial selectivity of the nucleophilic attack to promote such electrostatic attractions.

Abstract:Talaromyces flavus containing a constitutive epoxide hydrolase (EH) resolved racemic benzyl glycidyl ether and nine derivatives into their ( R )-enantiomers. After optimization of the fermentation conditions, the specific EH activity and biomass concentration were improved from 13.5 U/g DCW and 14.8 g DCW/l to 26.2 U/g DCW and 31.3 g DCW/l, respectively, with final values for e.e. s of 96 % and E of 13 with ( R )-benzyl glycidyl ether. Substituents on the phenyl ring, however, gave low enantioselectivities.

Abstract:A family of well-defined octahedral cationic chiral-at- cobalt(III) catalysts based on ( R , R )-1,2-cyclohexanediamine and ( R , R )-1,2-diphenylethylenediamine has been examined in the dynamic kinetic resolution of an azlactone derived from N -benzoyl- tert -leucine. The reactions catalyzed by 10 mol% of Co III complexes in the presence of 1 M aqueous NaOH solution under phase-transfer alcoholysis afforded the corresponding benzyl ester of tert -leucine with up to 76% yield and up to 66% enantioselectivity ( ee ).

Abstract:Catalytic asymmetric dynamic kinetic resolution of configurationally labile bridged biaryls is emerging as a powerful strategy for atropisomer synthesis. However, the reported examples suffer from an inherent challenge as the reactivity is highly dependent on the torsional strain of the biaryl substrates, which significantly narrows down the scope and hampers the application. Herein, we report our discovery and development of a torsional strain-independent reaction between biaryl thionolactones and activated isocyanides. By employing auto-tandem silver catalysis, a universal synthesis of both tri- and tetra-ortho-substituted thiazole-containing biaryls was realized in high yields with high enantioselectivities. In addition, these products could be facilely converted to a novel type of bridged biaryls bearing an eight-membered lactone. Mechanistic studies were carried out to elucidate the cause of this unusual torsional strain-independent reactivity.

Abstract:A new method for catalytic nonenzymatic kinetic resolution of alpha-substituted alkanoic acids has been developed, which relies on their activation with DCC followed by enantioselective alcoholysis of the intermediate symm-anhydrides in the presence of the amidine-based catalyst homobenzotetramisole (HBTM). Moderate to excellent selectivity factors (s = 5-96) have been obtained in the case of several classes of substrates, namely, alpha-aryl-, alpha-aryloxy/alkoxy-, alpha-halo-, alpha-azido-, and alpha-phthalimido-alkanoic acids. Under similar conditions, alpha-(arylthio/alkylthio)-alkanoic acids undergo dynamic kinetic resolution providing corresponding esters in up to 92% ee and up to 93% yield.

Abstract:The development of methods to allow the selective acylative dynamic kinetic resolution (DKR) of tetra-substituted lactols is a recognised synthetic challenge. In this manuscript, a highly enantioselective isothiourea-catalysed acylative DKR of tetra-substituted morpholinone and benzoxazinone-derived lactols is reported. The scope and limitations of this methodology have been developed, with high enantioselectivity and good to excellent yields (up to 89%, 99:1 er) observed across a broad range of substrate derivatives incorporating substitution at N(4) and C(2), di- and spirocyclic substitution at C(5)- and C(6)-position, as well as benzannulation (>35 examples in total). The DKR process is amenable to scale-up on a 1 g laboratory scale. The factors leading to high selectivity in this DKR process have been probed through computation, with an N-C=O•••isothiouronium interaction identified as key to producing ester products in highly enantioenriched form.

Abstract:A convenient method for the enantioselective synthesis of β′-hydroxy-β-amino esters was developed through intensive investigations of the reaction conditions. Dichloromethane (DCM)/2,2,2-trifluoroethanol (TFE) and 1,2-dichloroethane (DCE)/TFE combinations were found to be the appropriate co-solvents for the reaction. In this mixed solvent system, [Ru(cymene)Cl2]2–Sunphos exhibited unusually higher activity and selectivity than the corresponding [Ru(cymene)I2]2–Sunphos. In situ generated catalyst could be used for this transformation. Asymmetric hydrogenation of a series of β′-keto-β-amino esters derivatives using Ru–Sunphos complexes via dynamic kinetic resolution led to the corresponding β′-hydroxy-β-amino esters with ee up to 99.6% and de up to 98.8% (S/C=1000).

Abstract:Enantiomerically pure (R)-1-aminotetralin was prepared by dynamic kinetic resolution using a system of novel racemization catalyst combined with Novozym 435. The autocatalytic amidation reaction in the process would reduce the eeP value of the reaction. The autocatalytic amidation reaction was suppressed by changing the structure of acyl donor. The influence of acyl donor with different acid moieties and different alcohol moieties was investigated. As acyl donor became complicated,the dynamic kinetic resolution result of 1-aminotetralin could also become better. While using 4-chlorophenyl valerate as acyl donor,dynamic kinetic resolution could reach the best result with eeP value>99%and conversion>99%.

Abstract:An efficient rhodium-catalyzed asymmetric transfer hydrogenation of β-cyano α-ketoesters via dynamic kinetic resolution has been developed. Despite the challenge posed by multiple functional groups, the reaction proceeded smoothly under mild conditions, generating versatile synthons with two adjacent stereocenters in high yields with excellent enantio- and diastereoselectivities. Furthermore, the power of this strategy is highlighted by the scale-up reaction and the follow-up synthesis of cytoxazone and paclitaxel intermediates.

Abstract:We report herein an unprecedented atroposelective dynamic kinetic resolution of Bringmann's lactones with C-nucleophiles. By the use of activated isocyanides as the reagent, a wide range of novel axially chiral oxazole-substituted biaryl phenols were accessed in high yields with high enantioselectivities. Abstract:A new and efficient dynamic kinetic resolution (DKR) process of secondary aromatic alcohols by using long carbon-chain esters as acyl donors has been developed. During the process, the transesterification catalyzed by CD8604 was found to be the main reason for the decrease in enantiomeric excess (ee). Using complex acyl donors, such as 4-chlorophenyl valerate, we could effectively inhibit the resin-catalyzed transesterification, and an excellent ee value (>99%) at high yield (>99%) was achieved. The mechanism for the inhibition of resin-catalyzed transesterification is believed to be the formation of micro-micelles in the pores of CD8604. It is noteworthy that the system can be reused more than 20 times without a loss of yield or ee value.

Abstract:Dynamic kinetic resolution of various α-amido-β-keto phosphonates via asymmetric hydrogenation proceeded efficiently to give the corresponding β-hydroxy-α-amido phosphonates in high diastereo- and enantioselectivities (up to 99:1 syn/anti, 99.8% ee). The addition of catalytic amounts of CeCl(3)·7H(2)O is necessary to achieve both good selectivity and catalytic efficiency under mild reaction conditions.

Abstract:Herein we report a highly enantioselective kinetic resolution of tertiary benzyl alcohols via palladium/chiral norbornene cooperative catalysis. With simple aryl iodides as the resolution reagent, a wide range of readily available racemic tertiary benzyl alcohols are applicable to this method. Both chiral tertiary benzyl alcohols and benzo[c]chromene products are obtained in good to excellent enantioselectivities (selectivity factor up to 544). The appealing synthetic utility of the obtained enantioenriched tertiary alcohols is demonstrated by the facile preparation of several valuable chiral heterocycles. Preliminary mechanism studies include DFT calculations to explain the origin of enantiodiscrimination and control experiments to uncover the formation of a transient axial chirality during the kinetic resolution step.

Abstract:Dynamic kinetic resolution (DKR) has been applied to primary amines utilizing a ruthenium catalyst and an (R)-selective lipase to provide the corresponding amides in up to 95% isolated yield and up to >99% ee. Further investigations of the chemoenzymatic DKR of amines show that it is general and can be applied to a wide range of amines. The protocol was found to be applicable with both isopropyl acetate and dibenzyl carbonate as acyl donors. In the latter case, release of the free amine from the carbamate products was carried out under very mild conditions. A new route to prepare norsertraline, an antidepressant, utilizing DKR as one of the key steps has also been developed.Further investigations of the DKR of 1-phenylethylamine led to the development of a protocol that could be used on gram scale. Several parameters were studied including catalyst loading, acyl donor used, and concentration of the reaction. The Ru-catalyst loading was decreased from 4 mol% to 1.25 mol% and the enzyme catalyst loading was decreased from 40 mg/mmol substrate to 10 mg/mmol substrate.The racemization of pipecoloxylidide, an intermediate in the synthesis of commonly used anesthetics, was carried out. This racemization method was developed for its potential use in an integrated process that combines enantiomer separation techniques and racemization of the undesired enantiomer. The integration of racemization of the undesired enantiomer would increase the overall yield of the desired enantiomer and thus make the procedure highly efficient.DKR has also been applied to allylic alcohols utilizing a ruthenium catalyst and either an (R)-selective lipase or an (S)-selective protease to provide the corresponding allylic esters in high yield and high ee. A copper-catalyzed allylic substitution was then applied to provide the corresponding alkenes with inversion of stereochemistry. Subsequent C-C double bond cleavage afforded pharmaceutically important a-methyl substituted carboxylic acids in high ee.

Abstract:A new and efficient dynamic kinetic resolution (DKR) process of 1-phenylethanol was developed with acid resin as racemization catalyst and lipase as resolution catalyst Acid resin CD550 was shown to have excellent racemization activity and could racemize 100 mmol/L (S)-1-phenylethanol in 1 5 h at 60 degrees C When employing CD550 and complex acyl donors such as 1,2-diacetoxybenzene, 1,3-diacetoxybenzene and 3,5-dimethyl phenyl acetate instead of vinyl acetate as racemization catalyst and acyl donor respectively to the DKR of 1-phenylethanol, enantiomerically pure (R)-1-phenylethanol acetate could be obtained with excellent yield and ee values During processing, the conversion and the selectivity of the DKR were successfully enhanced by using more complex acyl donors to inhibit the acid resin-catalyzed unslective trans-esterification For instance, when 1,2-diacetoxybenzene as acyl donor, the conversion and the ee(p) could reach above 99% and 90 5% after 10 h, respectively
