Effect of humanin and [Gly14]-humanin on expression of hippocampal glial fibrillary acidic protein in mice with Alzheimer disease

Abstract:Objective To investigate neuroprotective effect and probable mechanism of humanin (HN) and its derivative [Gly14]-humanin(HNG) in experimental mice with Alzheimer's disease (AD).Methods Eighty-four healthy male mice were randomly divided into 4 groups:sham operated group(9 mice),Aβ model group(15 mice),HN treatment group(30 mice) and HNG treatment group(30 mice). Thirty mice in HN treatment group were randomized into two subgroups: one with 10μmol/L HN and the other with 100μmol/L HN; and those in HNG treatment group were randomized into two subgroups :10 nmol/L HNG subgroup and 100 nmol/L HNG subgroup. Changes in expression of glial fibrillary acidic protein(GFAP) in hippocampus were observed by immunohistochemical method on days 3,7,14. Results The number of cells expressing GFAP in hippocampus decreased in HN treatment group and HNG treatment group on days 3,7,14. The number of cells expressing GFAP in hippocampus in 100 μmol/L HN group was less than that in 10 μmol/L HN group on days 3 and 14;the number of cells expressing GFAP was less in 100 nmol/L HNG group on days 3,7,14 compared with 10 nmol/L HNG group. The number of GFAP positive cells was less in 100 nmol/L HNG group than in 100μmol/L HN group on 3~(rd) day (P<0.05). Conclusion Both HN and HNG can time- and dose-dependently decrease the expression of GFAP caused by the toxicity of Aβ_(25-35),indicating that both HN and HNG have neuroprotective effect.
