Abstract:A novel tandem reductive amination/intermolecular nucleophilic aromatic substitution (SNAr) sequence has been established for the synthesis of amine containing pyrimidine in formation of one carbon-oxygen and one carbon-nitrogen bonds in a one-pot fashion. Treatment of aldehyde with arylamine, 2-methanesulfonyl-4,6-dimethoxypyrimidine and sodium borohydride provides good overall yield. The p-toluenesulfonic acid (PTSA) can be used as activator and is generally needed in the reaction. Dioxane is the preferred reaction solvent, but reactions can also be carried out in tetrahydrofuran (THF), MeCN, toluene and dichloromethane. The procedure is carried out effectively in the presence of K2CO3. The reaction proceeds smoothly with aromatic aldehydes and arylamines possessing electron-donating or -withdrawing groups. This method can be applied to the synthesis of the oilseed rape herbicide and is superior to the classical one in several aspects: cutting out several purification steps, minimizing solvent use and chemical waste, and saving time. Its advantages such as operational convenience, high-efficient synthesis, and starting material availability make it a desirable method for preparing amines with molecular diversity and biological activity.

Abstract:A synthesis route to access triazole-pyrazole hybrids via pyrazolotriazenes was developed. Contrary to existing methods, this route allows the facile N-functionalization of the pyrazole before the attachment of the triazole unit via a copper-catalyzed azide-alkyne cycloaddition. The developed methodology was used to synthesize a library of over fifty novel multi-substituted pyrazole-triazole hybrids. We could also demonstrate a one-pot strategy that renders the isolation of potentially hazardous azides obsolete. In addition, the compatibility of the method with solid-phase synthesis was shown exemplarily.

Abstract:We have developed a novel one pot three steps process, including phosphine catalyzed Rauhut−Currier reaction of γ-alkyl allenoate, Diels-Alder reaction with di-tert-butyl azodicarboxylate and followed by deprotection of Boc group, to prepared tetra-substituted 1,6-dihydropyridazine in good yield. The 1,6-dihydropyridazine was easily converted to pyridazine derivatives via oxidative aromatization by DDQ.

Abstract:Immobilized l-glutamic acid β-methyl ester was sulfonylated with 4-nitrobenzenesulfonyl chloride and alkylated with various α-haloketones. The resulting sulfonamides were reacted with potassium trimethylsilanolate. Then, upon cleavage from the polymer support, tetrasubstituted pyridines were produced as the result of one-step C-arylation, aldol condensation, and oxidation. When cleavage from the resin occurred before the trimethylsilanolate treatment, C-arylation was followed by enamination, which yielded trisubstituted pyrazines. Through the developed protocols, targeted synthesis of novel heterocyclic derivatives was performed using mild reaction conditions and a number of readily available starting materials.

Abstract:The azomethine ylides are generally used in 1,3-dipolar cycloadditions with various dipolarophiles. In this work, a new and diverse route has been developed for the azomethine ylides, for synthesis of novel pyrrole derivatives. The azomethine ylide, produced via C–H activation of unreactive C(sp3)–H bond of 2-methylquinoline, by molecular iodine, in the presence of pyridine. Herein, we represent novel pyrrole derivatives, synthesized from the reaction of pyridinium ylide with olefins, which formed via a reaction of isatin, dialkyl acetylenedicarboxylate derivatives and pyridine as a base in moderate to excellent yields. Abstract:Practical and efficient methods have been developed for the synthesis of 4,6,8,9-tetrasubstituted 8,9-dihydro-5H-pyrimido[4,5-e][1,4]diazepin-7(6H)-ones from 4,6-dicholoropyrimidine aldehyde, N-substituted amino acid esters, and amines in five steps. This synthetic strategy is based on suitably substituted pyrimidines as bis-electrophilic species reacting with various amines to construct the pyrimido[4,5-e][1,4]diazepine core with a strategically anchored functional group for further derivatization. The utility of this methodology was demonstrated through the preparation of a 33-membered library of representative 8,9-dihydro-5H-pyrimido[4,5-e][1,4]diazepin-7(6H)-ones in good to excellent yields.

Abstract:A biologically inspired organocatalytic one-pot synthesis of highly functionalized pyridazines, which are ubiquitous structural units in a number of biologically active compounds, has been developed by starting from readily available diazo compounds and Morita-Baylis-Hillman (MBH) carbonates. Under mild reaction conditions, this synthetic route tolerated significant substrate variation to deliver a broad range of substituted products, including CF3 -substituted pyridazines derivatives. Moreover, the introduction of trifluoromethyl groups into the ring of pyridazine could be completed conveniently from 2,2,2-trifluorodiazoethane.

Abstract:Differently substituted pyrrolidine derivatives have been prepared by a three-component reaction of heterocyclic aldehydes, amino acid methyl or ethyl esters, and <em>trans</em>-1,2-dibenzoylethylene in toluene under catalyst-free conditions. This simple one-step procedure allows to obtain the desired 3,4-dibenzoyl-5-hetarylpyrrolidines in high yields ensuring high diastereoselectivity. Given the fact that the fundamental stage of the proposed reaction mechanism is based on 1,3-dipolar cycloaddition, DFT and TDDFT quantum-chemical calculations have been used for better understanding and characterization of geometry and electronic structure of the reaction intermediates and products.

Abstract:A palladium-catalyzed cross-coupling reaction of aryl iodides with cyclic vinyldiazo ester was developed. The reaction provides various 3-aryl-2-pyrones in good yields with high functional group tolerance. The synthetic application of the resulting 3-aryl-2-pyrones as the diene component in a Diels-Alder reaction was also described.

Abstract:A novel series of pyrazolyl[1,2,4]triazoles has been synthesized by the reaction of polyfunctionalized-triazole 1 with symmetric and asymmetric β -dicarbonyl compounds in ortho -phosphoric acid. Also, we found that the reaction between triazole 1 and arylidene malononitriles in DMF afforded Schiff base compounds 9a,b instead of the target pyrazolyl[1,2,4]triazole 8a,b . The same products 9a,b were synthesized via the reaction of triazole 1 with the corresponding aromatic aldehydes. Moreover, a new series of 6-methyl-6-aryl-5,6-dihydro[1,2,4]triazolo[4,3- b ][1,2,4,5]tetrazine-3-thiol 11a,b and 9-amino-5-methyl-3,5-diphenyl-8-sulfanyl-2,4,5,9-tetrahydro-3 H -[1,2,4]triazolo[5,1- c ][1,2,4]triazepin-3-ol 12a has been successfully synthesized by the reaction of triazoles 1 with acetophenone derivatives.

Abstract:An effective route to functionalized N 1 -[(2,6-dimethyl-4-oxo-4 H-pyran-3-yl)carbonyl]-1-arylsulfonamide and 3-(arylsulfonyl)-6-methyl-2 H-1,3-oxazine-2,4(3 H)-dione derivatives is described. This involves the reaction of arylsulfonyl isocyanates and diketene in the presence of N-heteroaromatic compounds such as 1,3,5-triazine, 1-methyl-1 H-imidazole, or pyridine as catalyst. The reactive 1:1 zwitterions obtained from the addition of N-heteroaromatic compound to arylsulfonyl isocyanates were trapped by diketene to produce functionalized sulfonamide and 1,3-oxazine-2,4(3 H)-dione derivatives.

Abstract:Herein, synthesis of a new series of pyrano[2,3- d ]pyrimidine scaffolds is presented. Ethyl 6-amino-4-(4-chlorophenyl)-5-cyano-2-methyl-4 H -pyran-3-carboxylate has been reacted with formamide, formic acid, urea, thiourea, semicarbazide, and thiosemicarbazide to give six new pyrano[2,3- d ]pyrimidine derivatives in moderate to high yields (48–78%). Acylation and intramolecular cyclization of pyran with carbon disulfide, ethyl cyanoacetate, acetic anhydride, or chloroacetyl chloride has resulted in formation of the corresponding desired bicyclic pyrimidine derivatives. The chloromethyl derivative was subjected to nucleophilic substitution by its reaction with morpholine, piperidine or aniline resulting in the corresponding 2-substituted pyrimidine derivatives in good yields.

Abstract:A simple, modular method to prepare highly substituted pyridines is disclosed. The method employs a cascade reaction comprising (1) a novel N-iminative, Cu-catalyzed cross-coupling of alkenylboronic acids at the N-O bond of alpha,beta-unsaturated ketoxime O-pentafluorobenzoates, (2) electrocyclization of the resulting 3-azatriene, and (3) air oxidation affording highly substituted pyridines in moderate to excellent isolated yields (43-91%). Starting materials are readily available, and functional group tolerance is very good.

Abstract:Herein, a novel protocol was constructed for the synthesis of highly functionalized 2,4-diarylpyridines. In this reaction, α,β-unsaturated ketoxime acetates and enaminones were used as substrates in a one-pot cascade reaction, which included a Michael reaction, intramolecular cyclization, dehydrogenation, and the removal of HOAc. This reaction resulted in the formation and cleavage of two bonds by refluxing a mixture of the starting materials in toluene and in the presence of the catalyst Fe(OTf)3. As a result, a series of functionalized pyridines were produced through a complex cascade reaction. This protocol can be used to synthesize functionalized asymmetrical 2,4-diarylpyridines. This approach is suitable for the combinatorial and parallel syntheses of pyridine derivatives in a one-pot reaction rather than traditional tedious multi-step reactions.

Abstract:In this study, a novel functionalized metal–organic frameworks MIL-125(Ti)-N(CH 2 PO 3 H 2 ) 2 was designed and synthesized via post-modification methodology. Then, MIL-125(Ti)-N(CH 2 PO 3 H 2 ) 2 as a mesoporous catalyst was applied for the synthesis of a wide range of novel tetrahydropyrido[2,3- d ]pyrimidines as bioactive candidate compounds by one-pot condensation reaction of 3-(1-methyl-1 H -pyrrol-2-yl)-3-oxopropanenitrile, 6-amino-1,3-dimethylpyrimidine-2,4(1 H ,3 H )-dione and aromatic aldehydes at 100 °C under solvent-free condition. Interestingly, the preparation of tetrahydropyrido[2,3- d ]pyrimidine was achieved via vinylogous anomeric based oxidation mechanism with a high yield and short reaction time.
