Intrleukin-1β Mrna Expression after Permanent Cerebral Ischemia in Rats

Abstract:Objective To study the regularity and mechanism of geneexpression of IL-1β after permanent focal stroke in rats.Methods Quantitative reverse transcription and polymerase chain reaction(RT-PCR) technique were used to examine the mRNA expression profile of IL-1β,ELISA was used to assay IL-1β protein level at various time after permanent occlusion of the middle cerebral artery(MCAO) in rats.Resullts IL-1βmRNA expressed very early in ischemic cortex at all time points;IL-1βmRNA expressed obviously after 1 h of ischemia(0.34±0.05),then the expression increased gradully,peaked at 24 th(1.23±0.15).IL-1β protein increased very early at 3 rd(384.1±57.7pg/g),peaked at 48 th(802.3±61.7pg/g),IL-1β protein still remained at high level at 5 th(753.9±46.5 pg/g).Conclusions The gene expression of IL-1β after focal stroke elevates,expression of IL-1β protein increase latedly and parallelly,it may suggest that the role of IL-1β in ischemic injury not only confines to acute phase.
